In vitro antibiotic susceptibility of the newly recognized agent of ehrlichiosis in humans, Ehrlichia chaffeensis.

Ehrlichiosis in humans, a rickettsial disease recently discovered in the United States, is generally treated successfully with tetracyclines; however treatment with these agents is usually avoided with children and pregnant women. The in vitro susceptibility of Ehrlichia chaffeensis, the agent of human ehrlichiosis in the United States, was assessed by a quantitative evaluation of infected DH82 cells cultivated in 96-well microtiter plates in the presence of different concentrations of selected antibiotics. Extracellular MICs and MBCs were evaluated after 72 h of exposure to the antibiotics. D

Mechanisms of resistance in anaerobes and new developments in testing.

Anaerobic bacteria currently demonstrate increased resistance to antimicrobial agents, primarily by the production of beta-lactamase. A number of species of Bacteroides, most notably those in the Bacteroides fragilis group, produce these enzymes. A few species of Fusobacterium and Clostridium produce beta-lactamase as well. Fortunately, this mechanism of resistance is readily overcome by administering beta-lactamase inhibitors coupled with a beta-lactam antibiotic that would otherwise be inactivated. Other types of resistance encountered in anaerobic bacteria include inactivating enzymes such

Antibiotic sensitivities of Aeromonas hydrophila cultured from medicinal leeches.

Increased use of medicinal leeches (Hirudo medicinalis) for the treatment of venous congestion in flaps and replanted parts has coincided with reports of soft tissue infections following leech application. We cultured the gullets of 20 medicinal leeches to re-examine the antibiotic sensitivities of Aeromonas hydrophila, the leech enteric organism associated with reported infections. These isolates reflected reported resistance to penicillin and first generation cephalosporins as well as sensitivity to gentamicin, tetracycline and chloramphenicol. Additionally, the cultures were sensitive to ce

Susceptibility of Legionella spp. to imipenem and 27 other beta-lactam antibiotics.

With 28 beta-lactam antibiotics the susceptibilities of 60 strains of Legionella spp. (49 Legionella pneumophila and 11 ATCC type strains of other Legionella species) were determined. Agar dilution testing was used on buffered charcoal-yeast extract agar to which 0.1% alpha-ketoglutarate was added. The most active of the penicillins tested were ampicillin (MIC = 0.06-8 mg/l) and temocilin (MIC = 0.125 - 16 mg/l); the most active cephalosporins were ceftazidime (MIC = 0.03 - 0.5 mg/l), HR 810 (MIC = 0.06 - 1 mg/l) and cefoxitin (MIC = 0.125 - 2 mg/l). Of all the drugs tested imipenem had the mo

Rapid drug susceptibility testing of mycobacteria in tissue culture medium.

Mycobacteria grew rapidly in FST medium (tissue culture medium F12 supplemented with 5% serum and 0.05% Tween 80). Growth of Mycobacterium tuberculosis and other niacin-negative mycobacteria in flat-bottomed, 96-well tissue culture plates was estimable by the naked eye within 3 to 5 days when mycobacteria were inoculated at 0.1 to 0.01 Klett units (5 X 10(4) to 0.5 X 10(4) CFU) per well. Spontaneous resistant variants of M. tuberculosis to isoniazid arose and grew in the medium within 2 weeks of culture. A total of 56 clinically isolated mycobacteria whose drug susceptibilities had been tested

[Characteristics of the resistance of Pseudomonas aeruginosa plasmids].

A total of 503 clinical strains of Ps. aeruginosa isolated in hospitals of 4 towns in 1976 - 1978 were studied. It was shown that a significant number of the isolates were resistant to high concentrations of streptomycin (83.3 per cent), kanamycin (71 per cent), sulfanilamide (70.2 per cent) and mercuric chloride (61.6 per cent). Strains resistant to gentamicin (29.8 per cent) and carbenicillin (20.2 per cent) occurred comparatively rare. Resistance to polymyxin and rifampicin was recorded in 2 and 4 per cent f the isolates respectively. It was found that 77 per cent of the isolates could tran

In vitro susceptibility of strains of Pseudomonas pseudomallei to rifampin.

Reported high activity of rifampin for Pseudomonas pseudomallei could not be verified by extensive in vitro tests conducted with 31 recently isolated strains. Minimal inhibitory concentrations of rifampin were 25 mug/ml for three strains and greater than 25 mug/ml for 28 strains. Rifampin had relatively poor in vitro activity when compared with tetracycline drugs and chloramphenicol antibiotics now commonly used for treating melioidosis.

Potent antimicrobial peptides against Legionella pneumophila and its environmental host, Acanthamoeba castellanii.

Legionella pneumophila, the major causative agent of Legionnaires' disease, is most often found in the environment in close association with free-living amoebae, leading to persistence, spread, biocide resistance, and elevated virulence of the bacterium. In the present study, we evaluated the anti-Legionella and anti-Acanthamoeba activities of three alpha-helical antimicrobial peptides (AMPs), namely, NK-2, Ci-MAM-A24, and Ci-PAP-A22, already known for the extraordinary efficacy against other microbes. Our data represent the first demonstration of the activity of a particular AMP against both

Isolation, identification and antimicrobial susceptibility of brucella blood culture isolates.

UNLABELLED: Isolation of slowly growing and fastidious Brucella spp strains from clinical specimens is difficult, because of varying factors, including species specificities, stadium of disease, and previous antibiotic treatment of the patients. The use of automated blood culture systems has overcome some cultivation problems. The automated identification system such as VITEK 2 compact allows more precise identification, as well. AIM: To present our own experience in the isolation of Brucella species from blood cultures, by the Bact/Alert automated system, identification by the VITEK 2 compac

Inhibition and disruption of bacterial biofilms by a bipyridine-ruthenium complex: a broad-spectrum in vitro study.

Biofilms, communities of microorganisms, are important in the medical and pharmaceutical sectors due to their potential to contaminate medical implants and other equipment. Because their thick matrix makes them resistant to antiseptics, disinfectants, and drugs, biofilms are difficult to eliminate. Therefore, this study investigated the antibiofilm potential of a ruthenium-based complex incorporating the bipyridine ligand CS27. Using the XTT reduction assay, the inhibitory and disruptive effects of CS27 on bacterial biofilms were assessed in vitro for a panel of clinically and industrially rel

Evolution under low antibiotic concentrations: a risk for the selection of Pseudomonas aeruginosa multidrug-resistant mutants in nature.

Antibiotic pollution of non-clinical environments might have a relevant impact on human health if resistant pathogens are selected. However, this potential risk is often overlooked, since drug concentrations in nature are usually below their minimal inhibitory concentrations (MICs). Albeit, antibiotic resistant bacteria can be selected even at sub-MIC concentrations, in a range known as the sub-MIC selective window. Using short-term evolution experiments, we have determined the sub-MIC selective windows of the opportunistic pathogen Pseudomonas aeruginosa for seven antibiotics of clinical rele

Ciprofloxacin induced antibiotic resistance in Salmonella Typhimurium mutants and genome analysis.

Antibiotic resistance of Salmonella species is well reported. Ciprofloxacin is the frontline antibiotic for salmonellosis. The repeated exposure to ciprofloxacin leads to resistant strains. After 20 cycles of antibiotic exposure, resistant bacterial clones were evaluated. The colony size of the mutants was small and had an extended lag phase compared to parent strain. The whole genome sequencing showed 40,513 mutations across the genome. Small percentage (5.2%) of mutations was non-synonymous. Four-fold more transitions were observed than transversions. Ratio of < 1 transition vs transversion

Non-albicans oral candidosis in HIV-positive patients.

Specimens from HIV-positive patients with oral candidosis were taken for culture, species identification and azole susceptibility testing, which was correlated with treatment outcome. Of 921 specimens, 95 yielded non-albicans species, mainly from patients with low CD4 lymphocyte counts and extensive previous azole exposure. Most non-albicans isolates were from specimens co-infected with Candida albicans, complicating the interpretation of in-vitro susceptibility results, which accurately predicted antifungal failure when the non-albicans species was isolated alone. Eighty-five non-albicans iso

In vitro activity of cefpirome against selected clinical enterobacterial isolates with beta-lactamase-mediated resistance.

Previous studies have shown that there is a high incidence of resistance to cephalosporins among enterobacteria isolated in Greek hospitals. This resistance is mainly due to either the derepression of chromosomal cephalosporinases or the acquisition of plasmids coding for SHV-5 type beta-lactamase. In the present study the activity of cefpirome against a number of enterobacteria (Escherichia coli, Klebsiella pneumoniae, Enterobacter cloacae, Enterobacter aerogenes and Serratia marcescens) possessing the mechanisms mentioned above was examined. Cefpirome was found active against all the strains

[Antibiotic sensitivity of Mycoplasma pathogenic for man].

Human pathogen mycoplasmas (Mycoplasma pneumoniae, Mycoplasma hominis, Ureaplasma urealyticum) are intrinsically resistant to antibiotics which inhibit the cell wall biosynthesis (beta-lactams, vancomycin, bacitracin), to polymyxins, rifamycins, sulfonamides, trimethoprim, 5-nitroimidazoles, nitrofurans and to presently available quinolones. These three species are moderately susceptible to aminoglycosides, susceptible to chloramphenicol and highly susceptible to tetracyclines. M. pneumoniae is susceptible to macrolides, lincosamins and streptogramins. M. hominis is resistant to early macrolid
