BACKGROUND: Similar to other recent mumps genotype G outbreaks worldwide, most mumps patients during the recent mumps genotype G outbreaks in the Netherlands had received 2 doses of measles, mumps and rubella (MMR) vaccine during childhood. Sera taken from two cases of mumps reinfection were tested against different strains of mumps virus in a plaque reduction neutralisation assay. Antibodies to mumps in the sera reacted with all strains, but the pattern of emergence of these antibodies differed with the strains tested. These findings raise the possibility of emergence of a mutant strain under the selective pressure of immunisation, with limited or no cross protection induced by the vaccine strain.

By immunoprecipitation analysis using antisera against oligo peptides synthesized based on the deduced N-terminal and C-terminal amino acid sequences of the SH proteins of the mumps virus, the SH protein was detected in mumps virus-infected cells. The SH protein expressed from cDNA by the vaccinia-T7 expression system was recovered in the membrane fraction. A pleocytosis of the CSF occurred in all of 10 children with mumps meningitis and persisted for weeks and months in some patients. Oligoclonal IgG proteins were detected in the cerebrospinal fluid (CSF) during the 2nd week after onset of meningitis or later in 4 out of 10 patients, and could be detected as late as 11 and 12 months after meningitis in 2 patients.
