The relaxing effects of barbiturates in vascular smooth muscle of rat aorta.

The effects of thiamylal and pentobarbital on contractions mediated through the influx of extracellular Ca(++) and the release of intracellularly stored Ca(++) were compared in rat aortic strips. Thiamylal (3 x 10(-5) M to 10(-3) M) and pentobarbital (10(-4) to 10(-3) M) significantly attenuated the contraction induced by KCl (20 mM), and shifted the dose-response curve for Ca(++) of KCl (20 mM)-treated strips downwards and to the right. Caffeine (10(-2) M)-induced contraction was significantly attenuated by thiamylal at concentrations greater than 10(-4) M and by pentobarbital at 3 x 10(-4) M

FOXC1 Controls Smooth Muscle Cell Differentiation and Plasticity in Vascular Disease.

Smooth muscle cell-related transcription factors have been associated with coronary artery disease in large genome-wide association studies, increasing their translational value for therapeutic investigations. Our study shows for the first time that FOXC1 is a major transcriptional regulator in vascular injury and atherosclerotic lesions, causally implicated in the disease. Functionally, our data suggest that FOXC1 modulates smooth muscle cell activation vs quiescence by regulating key pathways involved in cell adhesion, cell cycle progression, and actin cytoskeleton organization. Further inve

PCSK9 expression in human atherosclerotic lesions and vascular smooth muscle cells.

AIMS: Proprotein convertase subtilisin/kexin type 9 (PCSK9) is the third gene involved in autosomal dominant hypercholesterolemia, an atherosclerosis major cause. Its role in regulating plasma cholesterol through hepatocyte functions has been widely reported, and anti-PCSK9 drugs have been developed. However, direct interactions between PCSK9 and arterial pathology are not fully elucidated. As vascular smooth muscle cells (vSMC) play a major role in arterial wall pathophysiology, we describe PCSK9's presence in human atherosclerotic lesions and its interaction with vSMC. METHODS: Healthy and

Defective autophagy in vascular smooth muscle cells promote uremic accelerated atherosclerosis.

Individuals with chronic kidney disease (CKD) exhibit a significantly elevated risk of premature mortality, predominantly attributable to cardiovascular etiologies. Atherosclerosis is a multifactorial vascular pathology driven by dysregulated lipid metabolism. Notably, patients with CKD exhibit an accelerated progression of atherosclerosis, a condition referred to as uremia-accelerated atherosclerosis (UAAS). Autophagy represents an evolutionarily conserved cellular process that plays a pivotal role in maintaining intracellular homeostasis across cardiovascular cell types. However, the involve

Deficiency of Smooth Muscle Adar1 Exacerbates Vascular Remodeling and Pulmonary Hypertension.

BACKGROUND: ADAR1 (adenosine deaminase acting on RNA 1) catalyzes the conversion of adenosine to inosine in double stranded RNAs (dsRNAs), which is critical to prevent autoinflammatory responses mediated by activation of the type I IFN (interferon) signaling. We define the role of ADAR1-dependent RNA editing in IFNβ activation in pulmonary arterial smooth muscle cells (PASMCs) from idiopathic pulmonary arterial hypertension (IPAH), a devastating disease leading to right heart failure and death. METHODS: RNA editing levels were analyzed in PASMCs from IPAH patients versus healthy controls. A c

Smooth Muscle LRRC8A Knockout Preserves Vascular Function in Ang II Hypertension.

BACKGROUND: Ang II (angiotensin II) causes hypertension and vascular inflammation both directly and indirectly via cytokines, including TNFα (tumor necrosis factor-α). In vascular smooth muscle cells (VSMCs), Ang II and TNFα activate Nox1 (NADPH oxidase 1) to produce superoxide. TNFα receptors associate with Nox1 and LRRC8A (leucine-rich repeat containing 8A) anion channels to modulate inflammation and contractility in a RhoA-dependent manner. VSMC-specific LRRC8A knockout mesenteric arteries are protected from TNFα-induced injury, and vasodilation is preserved. We hypothesized that LRRC8A kno

pH-sensing GPR68 inhibits vascular smooth muscle cell proliferation through Rap1A.

Phenotypic transformation of vascular smooth muscle (VSM) from a contractile state to a synthetic, proliferative state is a hallmark of cardiovascular disease (CVD). In CVD, diseased tissue often becomes acidic from altered cellular metabolism secondary to compromised blood flow, yet the contribution of local acid/base imbalance to the disease process has been historically overlooked. In this study, we examined the regulatory impact of the pH-sensing G protein-coupled receptor GPR68 on vascular smooth muscle (VSM) proliferation in vivo and in vitro in wild-type (WT) and GPR68 knockout (KO) mal

PRDM16 deficiency in vascular smooth muscle cells aggravates abdominal aortic aneurysm.

Abdominal aortic aneurysm (AAA) is usually asymptomatic until life-threatening complications occur, predominantly involving aortic rupture. Currently, no drug-based treatments are available, primarily due to limited understanding of AAA pathogenesis. The transcriptional regulator PR domain-containing protein 16 (PRDM16) is highly expressed in the aorta, but its functions in the aorta are largely unknown. By RNA-seq analysis, we found that vascular smooth muscle cell-specific (VSMC-specific) Prdm16-knockout (Prdm16SMKO) mice already showed extensive changes in the expression of genes associated

Calcium promotes vascular smooth muscle cell phenotypic switching in Marfan syndrome.

Fibrillin 1 (Fbn1) mutations cause Marfan syndrome (MFS), with aortic root dilatation, dissection, and rupture. Few studies reported the blood calcium and lipid profile of MFS, and the effect of vascular smooth muscle cell (VSMC) phenotypic switching on MFS aortic aneurysm is unclear. Here, we aimed to investigate the role of calcium-related VSMC phenotypic switching in MFS. We retrospectively collected MFS patients' clinical data, performed bioinformatics analysis to screen the enriched biological process in MFS patients and mice, and detected markers of VSMC phenotypic switching on Fbn1C1039

FHL5 Controls Vascular Disease-Associated Gene Programs in Smooth Muscle Cells.

BACKGROUND: Genome-wide association studies have identified hundreds of loci associated with common vascular diseases, such as coronary artery disease, myocardial infarction, and hypertension. However, the lack of mechanistic insights for many GWAS loci limits their translation into the clinic. Among these loci with unknown functions is UFL1-four-and-a-half LIM (LIN-11, Isl-1, MEC-3) domain 5 (FHL5; chr6q16.1), which reached genome-wide significance in a recent coronary artery disease/ myocardial infarction GWAS meta-analysis. UFL1-FHL5 is also associated with several vascular diseases, consis

ATP-sensitive potassium channels in zebrafish cardiac and vascular smooth muscle.

ATP-sensitive potassium channels (KATP channels) are hetero-octameric nucleotide-gated ion channels that couple cellular metabolism to excitability in various tissues. In the heart, KATP channels are activated during ischaemia and potentially during adrenergic stimulation. In the vasculature, they are normally active at a low level, reducing vascular tone, but the ubiquitous nature of these channels leads to complex and poorly understood channelopathies as a result of gain- or loss-of-function mutations. Zebrafish (ZF) models of these channelopathies may provide insights to the link between mo

Smooth Muscle Cell-Specific Disruption of the BBSome Causes Vascular Dysfunction.

The BBSome-a complex consisting of 8 Bardet-Biedl syndrome proteins-is involved in the regulation of various cellular processes. Recently, the BBSome complex has emerged as an important regulator of cardiovascular function with implications for disease. In this study, we examined the role of the BBSome in vascular smooth muscle and its effects on the regulation of cardiovascular function. Smooth muscle-specific disruption of the BBSome through tamoxifen-inducible deletion of Bbs1 gene-a critical component of the BBSome complex-reduces relaxation and enhances contractility of vascular rings and

Cadherins in vascular smooth muscle cell (patho)biology: Quid nos scimus?

Vascular smooth muscle cells (SMCs) phenotypes span a reversible continuum from quiescent/contractile (differentiated) to proliferative/synthetic (dedifferentiated) enabling them to perform a diversity of functions that are context-dependent and important for vascular tone-diameter homeostasis, vasculogenesis, angiogenesis or vessel reparation after injury. Dysregulated phenotype modulation and failure to maintain/regain the mature differentiated and contractile phenotypic state is pivotal in the development of vascular diseases such as atherosclerosis and restenosis after angioplasty and coro

Apelin/APJ system: A critical regulator of vascular smooth muscle cell.

APJ, an orphan G protein-coupled receptor, is first identified through homology cloning in 1993. Apelin is endogenous ligand of APJ extracted from bovine stomach tissue in 1998. Apelin/APJ system is widely expressed in many kinds of cells such as endothelial cells, cardiomyocytes, especially vascular smooth muscle cell. Vascular smooth muscle cell (VSMC), an integral part of the vascular wall, takes part in many normal physiological processes. Our experiment firstly finds that apelin/APJ system enhances VSMC proliferation by ERK1/2-cyclin D1 signal pathway. Accumulating studies also show that

S100B promotes injury-induced vascular remodeling through modulating smooth muscle phenotype.

S100B is a biomarker of nervous system injury, but it is unknown if it is also involved in vascular injury. In the present study, we investigated S100B function in vascular remodeling following injury. Balloon injury in rat carotid artery progressively induced neointima formation while increasing S100B expression in both neointimal vascular smooth muscle (VSMC) and serum along with an induction of proliferating cell nuclear antigen (PCNA). Knockdown of S100B by its shRNA delivered by adenoviral transduction attenuated the PCNA expression and neointimal hyperplasia in vivo and suppressed PDGF-B

Homocysteine regulates endothelin type B receptors in vascular smooth muscle cells.

Vascular smooth muscle endothelin type B (ETB) receptor is involved in the pathogenesis of cardiovascular diseases (CVDs). Hyperhomocysteinemia is an independent risk factor for CVDs. The present study was designed to examine the hypothesis that homocysteine (Hcy) up-regulates vascular smooth muscle ETB receptors. In vitro experiments were performed in rat superior mesenteric artery (SMA) and vascular smooth muscle cells (VSMCs). The rat SMA or VSMCs were cultured in serum-free medium for 24h in the presence and absence of Hcy with or without specific inhibitors for the ERK1/2 signaling pathwa

Vascular smooth muscle cell phenotypic changes in patients with Marfan syndrome.

OBJECTIVE: Marfan's syndrome is characterized by the formation of ascending aortic aneurysms resulting from altered assembly of extracellular matrix microfibrils and chronic tissue growth factor (TGF)-β signaling. TGF-β is a potent regulator of the vascular smooth muscle cell (VSMC) phenotype. We hypothesized that as a result of the chronic TGF-β signaling, VSMC would alter their basal differentiation phenotype, which could facilitate the formation of aneurysms. This study explores whether Marfan's syndrome entails phenotypic alterations of VSMC and possible mechanisms at the subcellular level

Congenital smooth muscle hamartoma on the face treated using vascular laser.

A congenital smooth muscle hamartoma is a rare, benign proliferation of smooth muscle bundles in the dermis that is usually diagnosed in the neonatal period or infancy. Surgical excision is the first-line therapeutic option, but in certain areas such as the face, surgery may be too aggressive, and different treatments should be considered. We present the case of a congenital smooth muscle hamartoma on the face treated using pulsed dye laser with good response.

Molecular control of vascular smooth muscle cell differentiation and phenotypic plasticity.

Although the primary role of vascular smooth muscle cells (SMCs) is contraction, they exhibit extensive phenotypic diversity and plasticity during normal development, during repair of vascular injury, and in disease states including arteriosclerosis and tumour angiogenesis. Results of recent studies indicate that there are unique as well as common transcriptional regulatory mechanisms that control expression of various SMC marker genes within vascular SMC subtypes, and that these mechanisms are complex and dynamic even at the single cell level. This chapter will review recent progress in our u

A membrane potential-sensitive dye for vascular smooth muscle cells assays.

Changes in membrane potential of rat aorta smooth muscle cells were investigated using the bis-oxonol sensitive probe DIBAC2(3). We compared the changes in membrane potential induced by a high external KCl concentration in aorta smooth muscle cells from normotensive 2 kidney (2K) and from renal hypertensive 2 kidney-1 clip (2K-1C) rats. The spectral properties of the membrane potential were first characterized in aqueous buffers and in cultured smooth muscle cells from 2K and 2K-1C rat aortas. Fluorescence emission and the images were recorded using a laser scanning confocal microscope. The re
