Interleukin-4 receptor-Stat6 signaling in murine infections with a tissue-dwelling nematode parasite.

Interleukin-4 (IL-4) has been shown to be crucial in parasite expulsion in several gastrointestinal nematode infection models. Data from both epidemiological studies with humans and experimental infections in animals imply a critical role for the type II helper response, dominated by IL-4, in host protection. Here we utilized inbred mice on two distinct backgrounds to document the involvement of IL-4 in the clearance of a primary infection of Brugia from the murine host. Our data from infections of IL-4 receptor(-/-) and Stat6(-/-) mice further indicate that IL-4 exerts its effects by activati

Nematode infections in dog breeding kennels in The Netherlands, with special reference to Toxocara.

Faecal samples from 286 adult dogs and 159 pups and dust and soil samples from 32 dog breeding kennels in the Netherlands were examined for nematode eggs. Dogs that shed nematode eggs were found in 41% of the kennels. The kennel prevalence of nematode infection of adult dogs was 33%. The kennel prevalence for infection of adult dogs and pups with nematode species was 21% and 48% for Toxocara canis, respectively, 29% and 0% for Trichuris vulpis, and 20% and 0% for Toxascaris leonina. Kennels with more than two litters per year and with regular import of new animals had a significantly higher pr

Epidemiology of human intestinal nematode infections in Wujiang and Pizhou counties, Jiangsu Province, China.

Intestinal nematode infections are considered highly endemic in the Chinese province of Jiangsu. In May 1997, the prevalence of intestinal nematodes infections was determined among all of the inhabitants aged 5 to 65 of the southern Jiangsu village of Yaojiakon (Wujiang County) and the northern Jiangsu village of Jianmiao (Pizhou County). It was determined that the prevalence of ascariasis, trichuriasis and hook worm infection was greatly reduced compared to when it was last measured in 1990. The reduction was noted to be particularly impressive in Yaojiakon village where the prevalence of asc…

Gastrointestinal nematode infections of goats in a sub-tropical and humid zone of India.

Twelve hundred and twenty-eight goats (Capra hircus L.) from a sub-tropical and humid zone of India were examined for gastrointestinal nematodes. The species encountered in the region were: Haemonchus contortus, Bunostomum trigonocephalum, Oesophagostomum columbianum, Trichuris globulosa, O. aspersum, and T. ovis. The overall infection rate was 86.8%. Among various species found, H. contortus emerged as the most prevalent, although B. trigonocephalum and O. columbianum were also significantly in evidence. The seasonal fluctuation in infection was assessed by monitoring the faecal egg count of

Experimental Infection of Mice with the Parasitic Nematode Strongyloides ratti.

Strongyloides ratti is a parasitic nematode that naturally infects wild rats. However, most laboratory rat and mouse strains are fully susceptible to infection. Immunocompetent BALB/c and C57BL/6 mice terminate S. ratti infections within a month in the context of a canonical type 2 immune response and remain semi-resistant to a re-infection. The course of infection can be divided into three phases: (a) the tissue migration phase of the infective third-stage larvae during the first two days; (b) the early intestinal phase, including the molting to the adult parasites and embedding in the mucosa

Transcriptomic Insights into the Insect Immune Response to Nematode Infection.

Insects in nature interact with a wide variety of microbial enemies including nematodes. These include entomopathogenic nematodes that contain mutualistic bacteria and together are able to infect a broad range of insects in order to complete their life cycle and multiply, filarial nematodes which are vectored by mosquitoes, and other parasitic nematodes. Entomopathogenic nematodes are commonly used in biological control practices and they form excellent research tools for understanding the genetic and functional bases of nematode pathogenicity and insect anti-nematode immunity. In addition, cl

Anisakis Nematodes in Fish and Shellfish- from infection to allergies.

Anisakidosis is a zoonotic parasitosis induced by members of the family Anisakidae. The anisakid genera includes Anisakis, Pseudoterranova, Hysterothylacium and Contracaecum. The final definitive hosts of these nematodes are marine mammals with a complex life cycle. These nematode parasites use different crustaceans and fish species as intermediate or paratenic hosts and humans are accidental hosts. Human anisakiasis, the infections caused by members of the genus Anisakis, occurs, when seafoods, particularly fish, contaminated with the infective stage (third stage larvae [L3]) of this parasite

Immune-mediated regulation of chronic intestinal nematode infection.

Gastrointestinal nematode infection is extremely prevalent worldwide in humans and animals. Infection levels vary between individuals in infected populations and exhibit a negative binomial distribution, and some individuals appear to be predisposed to certain infection levels. Moreover, infection tends to be chronic, despite evidence for the acquisition of some degree of acquired immunity. The host is subject to constant and repeated antigenic challenge, and individuals vary in the response they make. While a considerable amount of information is emerging on the immunoregulatory mechanisms op

Responses of inbred mouse strains to infection with intestinal nematodes.

Comparisons were made of the immune and inflammatory responses of four strains of inbred mice to infection with the intestinal nematodes Trichinella spiralis and Nippostrongylus brasiliensis to determine whether genetically determined 'high responsiveness' to infection, seen most clearly in intestinal responses, is independent of the parasite concerned and necessarily correlated with protection. The time course of infection was followed by counting adult worms at intervals after infection. Mucosal mast cells and Paneth cell numbers were determined as indices of the intestinal inflammatory resp

Investigating development of infective stage larvae of filarial nematodes.

Lymphatic filariasis is a parasitic disease affecting millions of individuals and one of the leading causes of disability in the world. However, considering the magnitude of filariasis as a public health problem, relatively little is known about the basic biology of the disease and its causative agents, filarial nematodes. In this paper, the biology of the infective stage larvae of the nematodes responsible for lymphatic filariasis and the approaches used to study their development are reviewed.

Reducing intestinal nematode infection: efficacy of albendazole and mebendazole.

The widespread use of mebendazole and albendazole for treating intestinal nematode infections in human populations is raising concerns that careful monitoring pro- cedures should be in place to identify any emergence of drug resistance. In this article, Andy Bennett and Helen Guyatt discuss whether benchmark parasitological drug efficacy rates can be defined for these anthelmintics, by analysing published data on cure rates and egg reduction rates in the treatment of Ascaris lumbricoides, Trichuris trichiura and hookworm.

[Nematode larva migrans. On two cases of filarial infection].

With rapid air travel, so-called parasitic infections are becoming more important in northern hemisphere and temperate climates. Parasitic disease is usually taken to imply infections caused by protozoa and helminths. The most important helminthic infections in man and with world-wide incidence are schistosomiasis, hookworm, and filariasis. We report the clinico-pathological findings of two patients with filarial infection of soft tissue and lymphatic nodes.

DNA analysis in the diagnosis of infection and in the speciation of nematode parasites.

DNA analysis is playing an increasingly important role in characterising and classifying nematode parasites. Though less emphasis has been placed on utilising DNA elements to study nematodes of veterinary importance, correct diagnosis is, nevertheless, critical to proper treatment and control. The genus Trichinella presents a particularly interesting problem since the level of classification within this genus remains unclear. Herein we discuss the application of DNA analysis to the diagnosis and speciation of the parasitic nematode Trichinella spiralis, where the study of cloned repetitive DNA

Systemic autoimmune disease and concurrent nematode infection in a dog.

Systemic lupus erythematosus was diagnosed in a dog with concurrent nematode infection. The clinical signs of disease were unusually severe and included multiple neurologic deficits, polyarthritis, and weight loss. The dog was thrombocytopenic, and serotest results included positive lupus erythematosus test, positive rheumatoid factor test, positive antinuclear antibody test, hypergammaglobulinemia, and high platelet-associated IgG concentration. After treatment of hookworm, whipworm, and heartworm infections concurrently with corticosteroid and empiric treatment, the dog's condition improved.

Importance of TGFβ in Cancer and Nematode Infection and Their Interaction-Opinion.

Historically, there has been little interaction between parasitologists and oncologists, although some helminth infections predispose to the development of tumours. In addition, both parasites and tumours need to survive immune attack. Recent research suggests that both tumours and parasites suppress the immune response to increase their chances of survival. They both co-opt the transforming growth factor beta (TGFβ) signalling pathway to modulate the immune response to their benefit. In particular, there is concern that suppression of the immune response by nematodes and their products could

Intestinal nematode infection exacerbates experimental visceral leishmaniasis.

Leishmania donovani exposure often results in subclinical infection in immunocompetent individuals, and the factors dictating development of visceral leishmaniasis (VL) are not known. Infection with intestinal worms skew immunity towards type 2 and regulatory responses, thereby theoretically increases susceptibility to intracellular infections controlled by type 1 responses. Here we have tested how chronic infection with the intestinal nematode Heligmosomoides polygyrus affected immunity to a secondary infection with L donovani. We found that mice infected with H polygyrus displayed higher Lei

The host immune response to gastrointestinal nematode infection in sheep.

Gastrointestinal nematode infection represents a major threat to the health, welfare and productivity of sheep populations worldwide. Infected lambs have a reduced ability to absorb nutrients from the gastrointestinal tract, resulting in morbidity and occasional mortality. The current chemo-dominant approach to nematode control is considered unsustainable due to the increasing incidence of anthelmintic resistance. In addition, there is growing consumer demand for food products from animals not subjected to chemical treatment. Future mechanisms of nematode control must rely on alternative, sust
