Are there structural and functional modules in the vertebrate olfactory bulb?

A number of different recording methods have shown that odorants elicit patterns of neuronal activity widely distributed across cells of the olfactory receptor epithelium, olfactory bulb, and piriform cortex in the vertebrate olfactory system. These findings suggest that the physicochemical properties of odorant molecules are processed by distributed coding mechanisms activated in parallel in olfactory circuits in order to characterize a single, "monomolecular" odorant. These findings also suggest that the response patterns seen at higher levels are set up by differential responses in peripher…

The olfactory bulb: A neuroendocrine spotlight on feeding and metabolism.

Olfaction is the most ancient sense and is needed for food-seeking, danger protection, mating and survival. It is often the first sensory modality to perceive changes in the external environment, before sight, taste or sound. Odour molecules activate olfactory sensory neurons that reside on the olfactory epithelium in the nasal cavity, which transmits this odour-specific information to the olfactory bulb (OB), where it is relayed to higher brain regions involved in olfactory perception and behaviour. Besides odour processing, recent studies suggest that the OB extends its function into the reg…

Organizational Principles of the Centrifugal Projections to the Olfactory Bulb.

Centrifugal projections in the olfactory system are critical to both olfactory processing and behavior. The olfactory bulb (OB), the first relay station in odor processing, receives a substantial number of centrifugal inputs from the central brain regions. However, the anatomical organization of these centrifugal connections has not been fully elucidated, especially for the excitatory projection neurons of the OB, the mitral/tufted cells (M/TCs). Using rabies virus-mediated retrograde monosynaptic tracing in Thy1-Cre mice, we identified that the three most prominent inputs of the M/TCs came fr…

Congenital Agenesis of the Olfactory Bulbs: What to Suspect?

Complete agenesis of the olfactory bulbs (OB) constitutes a rare cause of congenital anosmia, which is more often associated with cerebral malformations or genetic disorders. The authors present a very rare case of a 28-year-old caucasian male with complaints of complete lack of the sense of smell since childhood. Radiologic study confirmed complete bilateral agenesis of the OB, with no other radiologic or clinical findings, such as delayed sexual development. Laboratorial investigation confirmed no signs of adrenocortical insufficiency. Despite being immature in the term neonate, the OB are a…

Calcification of the olfactory bulbs in three patients with hyposmia.

An estimated two million Americans suffer from chemosensory disorders. We present the clinical and imaging findings in three hyposmic patients with bilateral olfactory bulb calcification detected by CT. To our knowledge, these are the first cases of olfactory bulb calcification reported in the literature. A review of the literature concerning calcification of cranial nerves, olfactory neuritis, and the potential etiology and clinical significance of olfactory bulb calcification in our patients is presented.

Electrophysiological differentiation of new neurons in the olfactory bulb.

The subventricular zone produces neuroblasts that migrate to the olfactory bulb (OB) and differentiate into interneurons throughout postnatal life (Altman and Das, 1966; Hinds, 1968; Altman, 1969; Kishi et al., 1990; Luskin, 1993; Lois and Alvarez-Buylla, 1994). Although such postnatally generated interneurons have been characterized morphologically, their physiological differentiation has not been thoroughly described. Combining retroviral-mediated labeling of newly generated neurons with patch-clamp electrophysiology, we demonstrated that soon after new cells enter the layers of the olfactor…

Novel microglomerular structures in the olfactory bulb of mice.

The murine olfactory system consists of two primary divisions: (1) a main olfactory system, in which olfactory sensory neurons (OSNs) located in the main olfactory epithelium (MOE) send their axons to glomeruli in the main olfactory bulb (MOB); and (2) an accessory olfactory system, in which OSNs located in the vomeronasal organ send their axons to glomeruli in the accessory olfactory bulb (AOB). In labeling studies using the lectin Ulex europaeus agglutinin (UEA), we discovered a novel subset of small neuropilar structures in the MOB that are distinct from other glomeruli both in the MOB and …

Structure of the olfactory bulb in tadpoles of Xenopus laevis.

The structure of the olfactory bulb in tadpoles of Xenopus laevis (stages 54-56) was studied using axon tracing (with biocytin or low-weight dextran) and immunocytochemical techniques. Filling the olfactory nerve with biocytin made the nerve layer and the glomeruli visible. Dye injections into the glomerular layer labeled the lateral olfactory tract. Vice versa, dye injections into the lateral olfactory tract made mitral cells and their glomerular branching patterns visible. Anti-GABA antiserum stained periglomerular and granule cells, while the olfactory nerve and mitral cells were labeled by…

[Calretinin immunoreactivity in olfactory bulb and mucosa in mice].

The vertebrate olfactory system is capable of recognizing and discriminating thousands of different odorant molecules. In vitro biochemical studies have shown that some odorants elicit an increase in the intercellular second messenger cyclic AMP, whereas other odorants cause an increase in the second messenger inositol triphosphate (IP3). If IP3 plays an important role in the signal transduction, calcium-binding protein will be needed. We investigated the expression of calretinin, which belongs to the E-F hand family, in the neonatal and adult olfactory mucosa and olfactory bulb. Mice were sac…

The connections between the olfactory bulb and the brain in the goldfish.

We have investigated olfactory bulb connections in the goldfish by using both horseradish peroxidase applied to olfactory tract lesions and wheat germ agglutinin-conjugated peroxidase administered to the olfactory bulb. The projections from the bulb pass to the brain in two major bundles: the lateral and the medial olfactory tracts. The lateral tract innervates the posterior terminal field in the area dorsalis. The medial tract divides into two rami. The dorsolateral ramus is the most substantial olfactory bundle in the brain and innervates several targets in both the area dorsalis and the are…

Glutamate decarboxylase localization in neurons of the olfactory bulb.

Glutamate decarboxylase (GAD), the enzyme that synthesizes the neurotransmitter gamma-aminobutyric acid (GABA), has been localized in the rat olfactory bulb by immunocytochemical methods with both light and electron microscopy. The light microscopic results demonstrated GAD-positive puncta concentrated in the external plexiform layer and in the glomeruli of the glomerular layer. In addition, GAD-positive reaction product stained the dentrites and somata of granule and periglomerular cells. The electron microscopic observations confirmed the presence of GAD-positive reaction product within gran…

Transcutaneous Vagus Nerve Stimulation at Olfactory Bulb-Relevant Frequencies Modulates Human Olfactory Function.

Transcutaneous vagus nerve stimulation (tVNS) is a noninvasive technique that engages central neuromodulatory pathways and has been shown to … nner. The selective effect of gamma frequency, that is, the known processing frequency of the human olfactory bulb, implies inhibitory interneuron engagement in the olfactory bulb. These findings demonstrate that noninvasive tVNS can modulate human olfactory processing in a frequency-specific manner and highlight stimulation frequency as a critical parameter for …

Reduced olfactory bulb volume accompanies olfactory dysfunction after mild SARS-CoV-2 infection.

Despite its high prevalence, the determinants of smelling impairment in COVID-19 remain not fully understood. In this work, we aimed to examine the association between olfactory bulb volume and the clinical trajectory of COVID-19-related smelling impairment in a large-scale magnetic resonance imaging (MRI) analysis. Data of non-vaccinated COVID-19 convalescents recruited within the framework of the prospective Hamburg City Health Study COVID Program between March and December 2020 were analyzed. At baseline, 233 participants underwent MRI and neuropsychological testing as well as a structured …

Integrated age-related immunohistological changes occur in human olfactory epithelium and olfactory bulb.

Olfactory epithelium (OE) is capable of lifelong regeneration due to presence of basal progenitor cells that respond to injury or neuronal loss with increased activity. However, this capability diminishes with advancing age and a decrease in odor perception in older individuals is well established. To characterize changes associated with age in the peripheral olfactory system, an in-depth analysis of the OE and its neuronal projections onto the olfactory bulb (OB) as a function of age was performed. Human olfactory tissue autopsy samples from 36 subjects with an average age of 74.1 years were …

MR Imaging-Based Evaluations of Olfactory Bulb Atrophy in Patients with Olfactory Dysfunction.

BACKGROUND AND PURPOSE: Although the olfactory bulb volume as assessed with MR imaging is known to reflect olfactory function, it is not always measured during olfactory pathway assessments in clinical settings. We aimed to evaluate the utility of visual olfactory bulb atrophy and neuropathy analyses using MR imaging in patients with olfactory dysfunction. MATERIALS AND METHODS: Thirty-four patients who presented with subjective olfactory loss between March 2016 and February 2017 were included. Patients underwent a nasal endoscopic examination, olfactory testing with the Korean Version of the…
