Four of six cockatiels died within a week after being purchased from a commercial breeder. A fifth bird was euthanatized and necropsied during this time, and tissues were collected for microscopic examination. The small intestine had moderate numbers of Cryptosporidium sp. parasites present. A few large, basophilic intranuclear inclusions were present in renal ductular epithelium. Cryptosporidium sp. oocysts were found in the feces of the surviving bird. Thirty oocysts were 7.2 by 5.5 microns. The shape index was 1.31. Morphological examinations of the fecal oocysts indicated that the Cryptosp…

The antigenic constituents of sporulated Cryptosporidium parvum oocyst antigens were characterized with antisera from mice immunized against C. parvum. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis followed by silver staining defined the major proteins. Six of seven lectins used recognized as many as 15 bands. The lectins concanavalin A, Dolichos biflorus, and wheat germ agglutinin showed strong activity against the same eight bands with molecular weights ranging from 72,000 to greater than 100,000. An enzyme-linked immunosorbent assay was used to detect antibody to C. parvum. Anti

The absence of a standardized method for detecting oocysts in water samples makes it difficult to characterize them, including in water for reuse. This study aimed to detect Toxoplasma gondii oocysts using two extraction methods. Using method 1693/2014 USEPA, 30 L of water for reuse from two wastewater treatment plants (WWTPs) in the city of São Paulo, Brazil, was concentrated, totaling 20 samples. The supernatant generated from the immunomagnetic separation (IMS) step was collected for detection of T. gondii oocysts. For DNA extraction, two techniques were used: the commercial kit DNeasy Powe…

Toxoplasma gondii infection can result in toxoplasmosis and potential psychological effects. Research commonly focuses on infection through contact with cat fecal matter or consumption of contaminated meat. However, T. gondii oocysts can persist in the environment for years and may be present in soils and on soil-grown produce. Rates of oocyst DNA recovery from produce were high, with 18% of vegetable samples testing positive for T. gondii via PCR test and melt curve analysis. Radishes had significantly higher oocyst counts than arugula, collard greens, kale, lettuce, and spinach. There were n…

BACKGROUND: Toxoplasma gondii is the third most important contributor to health burden caused by food-borne illness. Ingestion of tissue cysts from undercooked meat is an important source of horizontal transmission to humans. However, there is an increasing awareness of the consumption of fresh fruit and vegetables, as a possible source for oocyst transmission, since this stage of the parasite can persist and remain infective in soil and water for long time. Herein, we outline findings related with detection of T. gondii oocysts in vegetables and berry fruits, which are usually raw consumed. T…

Toxoplasma gondii is a zoonotic protozoan parasite that can cause morbidity and mortality in humans, domestic animals, and terrestrial and a … felids. This manuscript further provides a comprehensive review of the pathways by which T. gondii oocysts can infect animals and people through the environment, including in contaminated foods, water or soil. We conclude by identifying critical control points for reducing risk of exposure to oocysts as well as opportunities for future synergies and new …

Water scarcity leads to an increased use of reclaimed water, which in turn calls for an improvement in water reclamation procedures to ensure adequate quality of the final effluent. The presence of infectious Cryptosporidium oocysts (IOO) in reclaimed water is a health hazard for users of this resource. Here, we gathered information on Cryptosporidium (concentrations, infectivity and genotype) in order to perform quantitative microbial risk assessment (QMRA). Moreover, data concerning the spores of sulphite-reducing clostridia (SRC) were used to undertake QMRA at a screening level. Our results…

Irrigation water and washing water have been inferred to be associated with contamination of fresh fruits and vegetables with pathogenic microorganisms infectious for humans. The objective of the present study was to determine whether apples experimentally contaminated with Cryptosporidium oocysts represent a food safety concern. Laser scanning confocal microscopy revealed no morphological changes in Cryptosporidium parvum oocysts attached to apples after 6 weeks of cold storage, suggesting that oocysts might remain viable and possibly infectious during prolonged storage. Mice were fed apple p…

Molecular characterization of Cryptosporidium spp.oocysts in clinical samples is useful for public health since it allows the study of sources of contamination as well as the transmission in different geographical regions. Although widely used in developed countries, in Brazil it is restricted to academic studies, mostly using commercial kits for the extraction of genomic DNA, or in collaboration with external reference centers, rendering the method expensive and limited. The study proposes the application of the modifications recently introduced in the method improving feasibility with lower …

Control of cryptosporidiosis is important in public health. Rivers that are polluted with Cryptosporidium and drinking water that is treated for drinking water production from polluted rivers could result in the waterborne disease of cryptosporidiosis. We carried out an epidemiological study of natural water supplies in Hokkaido, one of the largest dairy prefectures in Japan. To detect Cryptosporidium oocysts in environmental water, the filtration method was used for 28 samples, which were collected from 10 rivers. A method adapted from the United States Environmental Protection Agency (U.S. E…

Fecal samples were collected from 257 dogs in four areas in Korea during the period of January 1996 to November 1997 and examined by immunofluorescence assay for Cryptosporidium oocysts using a commercial diagnostic kit (Meridian Diagnostics, Cincinnati, Ohio). Of the 257 samples, 25 (9.7%) were positive for Cryptosporidium. Differences were noted in the prevalence of canine cryptosporidiosis in both areas and dog types. The results provide a further evidence of environmental contamination and widespread distribution of the parasite in Korea.

In animal and clinical studies of Cryptosporidium infection, counting Cryptosporidium oocysts in collected fecal samples is often critical to interpretation of results and validity of statistical analyses. In this experiment, 3 methods of counting oocysts derived from feces of infected mice were compared. These included sequential counting of oocysts in 25 adjacent microscopic fields, random counting of oocysts in 25 scattered microscopic fields, and hemacytometer counting of oocysts using standard procedures. Numerical results are expressed as the numbers of oocysts per microscopic field for …

With a view to optimize the etiological diagnosis of diarrhoea the authors have attempted to improve the diagnostic methodology for the parasite Cryptosporidium spp. as a current general technique, as well as a screening technique. A comparative evaluation of the various methods used for the concentration and the staining of cysts of Cryptosporidium spp. was done after quantification according to a five parameter score: electiveness, morphology, chromatic contrast, rapidity and accessibility. The importance of the screening tests in the diagnosis of cryptosporidiosis was evaluated according to

Cryptosporidium parvum is a zoonotic parasite of the intestine and poses a threat to human and animal health. However, it is difficult to obtain a large number of oocysts for genome sequencing using in vitro culture. To address this challenge, we describe a procedure for purification of C. parvum from a clinically positive sample and a method for isolation of a single oocyst of C. parvum using a three-axis hydraulic micromanipulator. We employ the strategy of whole-genome amplification (WGA) of 10 oocysts, and purify the whole-genome DNA after amplification, and finally meet the basic conditio…

Toxoplasma gondii is a coccidian parasite with the cat as its definitive host but any warm-blooded animal, including humans, may act as intermediate hosts. It has a worldwide distribution where it may cause acute and chronic toxoplasmosis. Infection can result from ingestion either of tissue cysts in infected meat of intermediate hosts or oocysts found in cat faeces via contaminated water or food. In this review, we highlight how the oocyst and sporocyst walls sustain the persistence and transmission of infective T. gondii parasites from terrestrial and aquatic environments to the host. We fur…

Toxoplasma (T.) gondii is a protozoan parasite with a broad range of intermediate hosts. Humans are often infected by ingestion of tissue cysts in raw or undercooked meat or meat products. Turkeys as food-producing animals can also serve as intermediate hosts. The aim of the present study was to investigate occurrence and predilection sites of T. gondii infection in turkeys after oral infection with oocysts. Experimental infections with different doses of T. gondii oocysts were performed in 36 turkeys to mimic natural infection. Systemic distribution of parasitic stages was investigated by scr…
