Identification of a Divergent Lineage Porcine Pestivirus in Nursing Piglets with Congenital Tremors and Reproduction of Disease following Experimental Inoculation.

Congenital tremors is a sporadic disease of neonatal pigs characterized by action-related repetitive myoclonus. A majority of outbreaks of congenital tremors have been attributed to an unidentified virus. The objectives of this project were to 1) detect potential pathogen(s) in samples from piglets with congenital tremors and 2) develop an infection model to reproduce disease. Using next-generation sequencing, a divergent lineage pestivirus was detected in piglets with congenital tremors. The virus was originally most closely related to a bat pestivirus but is now more closely related to a rec…

Detection of a Hobi-like virus in archival samples suggests circulation of this emerging pestivirus species in Europe prior to 2007.

The first reported incidence of Hobi-like viruses in Europe dates to a 2010 outbreak of respiratory disease in cattle in Italy. In this study, a Hobi-like virus was detected in archival samples, collected in 2007 in Italy from a cattle herd displaying respiratory disease, during the validation of a nested PCR protocol for rapid characterization of bovine pestiviruses. Phylogeny conducted with full-length pestivirus genomes and three informative genomic sequences, placed the strain detected in the samples, Italy-129/07, into the Hobi-like virus branch. Italy-129/07, similar to other Hobi-like v…

A multiplex real-time PCR assay for identification porcine circovirus 2, atypical porcine pestivirus, and classical swine fever virus.

BACKGROUND: Porcine circovirus 2 (PCV2), atypical porcine pestivirus (APPV), and classical swine fever virus (CSFV) are significant pathogens causing congenital tremor in swine, resulting in substantial economic losses worldwide. METHODS: A TaqMan multiplex real-time PCR method was developed to detect PCV2, APPV, and CSFV, simultaneously. This method was then applied to investigate 476 clinical samples from newborn piglets with congenital tremor symptoms. RESULTS: This method exhibited high specificity with no cross-reactivity with other viruses. Furthermore, the assay demonstrated remarkable …

Evolution and genetic diversity of atypical porcine pestivirus (APPV) from piglets with congenital tremor in Guangxi Province, Southern China.

Atypical porcine pestivirus (APPV) was identified and associated with congenital tremor (CT) type A-II in new born piglets and has been reported in many countries. In China, the first APPV identification in swine herds was reported in Guangdong province in 2016. To investigate the genetic characteristics of APPV in Guangxi province, 53 tissue samples from neonatal piglets with CT were collected and detected from October 2017 to May 2019. Five APPV strains which were named as GX04/2017, GX01-2018, GX02-2018, GX01-2019 and GX02-2019 were obtained. Sequence analysis revealed that all six APPV str…

Maximum likelihood and Bayesian analyses of a combined nucleotide sequence dataset for genetic characterization of a novel pestivirus, SVA/cont-08.

Bovine viral diarrhoea virus 1 (BVDV-1) and Bovine viral diarrhoea virus 2 (BVDV-2) are two recognised bovine pestivirus species of the genus Pestivirus. Recently, a pestivirus, termed SVA/cont-08, was detected in a batch of contaminated foetal calf serum originating from South America. Comparative sequence analysis showed that the SVA/cont-08 virus shares 15-28% higher sequence identity to pestivirus D32/00_'HoBi' than to members of BVDV-1 and BVDV-2. In order to reveal the phylogenetic relationship of SVA/cont-08 with other pestiviruses, a molecular dataset of 30 pestiviruses and 1,896 chara…

Proteomics computational analyses suggest that hepatitis C virus E1 and pestivirus E2 envelope glycoproteins are truncated class II fusion proteins.

Class II fusion proteins encoded by tick-borne encephalitis virus (TBEV), dengue virus, and Semliki Forest virus have a fusion peptide located at the end of a rod-like molecule comprised of three antiparallel beta sheet domains. Proteomics computational analyses suggest that hepatitis C virus (HCV) envelope glycoprotein E1 and pestivirus envelope glycoprotein E2 are truncated class II fusion proteins. Similarities were also detected between the receptor-binding portion of TBEV E and HCV E2, and between TBEV small membrane protein precursor prM and pestivirus E1. The proposed models of Flavivir…

Presence and shedding dynamics of Atypical Porcine Pestivirus (APPV) in semen of breeding boars: implications for virus transmission in pig production.

BACKGROUND: Atypical porcine pestivirus (APPV), genus Pestivirus, is the causative agent of congenital tremor type A-II, a neurological disease in newborn piglets that is characterized by tremors. Although the virus can spread horizontally, clinical disease is observed only after in utero infection. While venereal transmission has been proposed as a potential route, the pathogenesis and transmission dynamics of APPV remain incompletely understood. In this study, we aimed to investigate the presence of APPV in semen from Norwegian breeding boars, its temporal shedding patterns, and the potentia…

Cerebellar hypomyelination, white matter vacuolization, and prolonged presence of atypical porcine pestivirus in pigs with congenital tremor type A-II.

Atypical porcine pestivirus (APPV) is responsible for congenital tremor (CT) type A-II in pigs, a globally distributed neurological disease, with many unresolved questions regarding its pathogenesis and pathology. This descriptive case-control study assessed the viral load of APPV and its association with lesions in the central nervous system (CNS), as piglets born with severe clinical signs of CT recovered from clinical disease. The virus was found in all pigs with CT across 3 age groups (newborn, 3-week-old, 4- to 5-month-old CT pigs) using reverse transcription-quantitative polymerase chain…

Enhancing the understanding of coinfection outcomes: Impact of natural atypical porcine pestivirus infection on porcine reproductive and respiratory syndrome in pigs.

Atypical porcine pestivirus (APPV) is a novel member of the Pestivirus genus detected in association with congenital tremor (CT) type A-II outbreaks and from apparently healthy pigs, both as singular infection and as part of multi-pathogen infections. 'Classical' pestiviruses are known to cause immunosuppression of their host, which can increase susceptibility to secondary infections, severely impacting health, welfare, and production. To investigate APPV's effect on the host's immune system and characterise disease outcomes, 12 piglets from a natural APPV CT type A-II outbreak were experiment…

Genetic characterization of atypical porcine pestivirus (APPV) in China and the successful isolation of a novel APPV strain within genotype 2.

Atypical porcine pestivirus (APPV) is one major causative agent of congenital tremor (CT) type A-II. In this study, the geographical distribution and genetic characteristics of APPV strains in China were established. To date, APPV has been detected in at least ten provinces/regions of China. Genetic variability analysis showed that NS3 genes were highly conserved among Chinese APPV strains, while NS5A-, Npro-, and Erns-encoding genes were highly variable. Phylogenetic analysis revealed that all of the reference strains could be classified into 3 genotypes (1-3) and 7 subgenotypes (1.1-1.7). Th…

Genetically distinct pestiviruses pave the way to improved classical swine fever marker vaccine candidates based on the chimeric pestivirus concept.

Classical swine fever (CSF) is one of the most important viral diseases of pigs. In many countries, the use of vaccines is restricted due to limitations of subunit vaccines with regard to efficacy and onset of protection as well as failure of live vaccines to differentiate infected from vaccinated animals (DIVA principle). Chimeric pestiviruses based on CSF virus (CSFV) and the related bovine viral diarrhea virus (BVDV) have been licensed as live marker vaccines in Europe and Asia, but cross-reactive antibodies can cause problems in DIVA application due to close antigenic relationship. To deve…

Full-length sequence determination and isolation of infectious particles of an atypical porcine pestivirus from wild boars in Ishikawa Prefecture, Japan.

Atypical porcine pestivirus (APPV), the causative agent of congenital tremor (CT) type A-II, is a single-stranded (+) RNA virus classified in the genus Pestivirus of the family Flaviviridae that infects domestic pigs and wild boars. While surveys and studies on APPV infection in pigs are ongoing, information on APPV in wild boars is lacking. In Japan, there have been no reports of APPV infection in wild boars. We therefore screened serum samples from 333 wild boars to determine the prevalence of APPV in Ishikawa Prefecture. APPV-RNA was detected in two samples, and the complete viral genome wa…

Development of a TaqMan RT-qPCR for the detection and genotyping of bovine viral diarrhea virus types 1, 2, and HoBi-like pestivirus.

BACKGROUND: HoBi-like pestivirus (HoBiPeV), as a newly discovered virus species in recent years, poses a serious threat to the global cattle industry due to its wide transmission and high pathogenicity. The genetic sequence of HoBiPeV exhibits significant differences when compared to the known strains of BVDV-1 (Pestivirus bovis) and BVDV-2 (Pestivirus tauri).Current detection methods are typically limited to identifying BVDV-1 and BVDV-2, and are unable to effectively differentiate HoBiPeV from the other two genotypes. To facilitate the rapid detection and differentiation of various genotypes…

Validation of a Broad-Range Conventional RT-PCR Assay for Pestivirus Surveillance and Molecular Detection of BVDV-2a in Pigs in Costa Rica.

The genus Orthopestivirus, within the family Flaviviridae, includes important pathogens such as classical swine fever virus (CSFV), bovine viral diarrhea virus types 1 and 2 (BVDV-1 and BVDV-2), and border disease virus (BDV). Although Costa Rica is endemic for BVDV, it remains officially free of CSFV and BDV, highlighting the need for reliable molecular tools for pestivirus surveillance and differential diagnosis in swine populations. This study validated a conventional broad-range in-house RT-PCR assay targeting the 5' untranslated region (5'UTR) of pestiviruses, a genomic region that is hig…

Evaluation of an E2-based indirect ELISA for the serological differentiation of ovine Italy pestivirus from classical swine fever virus in pigs.

The genus Pestivirus includes several species responsible for economically important diseases in domestic and wild animals. In 2017, a novel ovine pestivirus (ovIT PeV) was isolated from aborted lamb fetuses and molecular analysis showed a genetic distinction from known Pestivirus species with the closest phylogenetic relationship to classical swine fever virus (CSFV, Pestivirus suis). This close similarity is concerning because CSF is a notifiable disease to the World Organization for Animal Health (WOAH) and is subject to eradication campaigns. Moreover, ovIT PeV can infect pigs and induce a…

12 novel atypical porcine pestivirus genomes from neonatal piglets with congenital tremors: A newly emerging branch and high prevalence in China.

Atypical porcine pestivirus (APPV), a newly discovered member of the genus Pestivirus, is considered to be associated with congenital tremors (CT) in piglets. From June 2016 to January 2018, 440 serum and tissue samples from CT-affected piglets in Anhui, Guangdong and Guangxi provinces were collected to detect APPV. The results showed a high level of 63.4% preference for APPV in 27 swine farms and complicated co-infection cases between APPV and other 12 swine viruses. Meanwhile, 12 novel APPV genomes were screened and identified. Results showed that complete genomes, Npro and NS5A genes of the…
