The selection of allergen-specific B cells into the plasma cell (PC) pool is a critical step in the immune dysregulation that leads to the production of IgE in allergic diseases. We sought to characterize the murine IgE repertoire. In particular, we questioned whether the IgE repertoire of plasmablasts (PBs)/PCs differs from the IgE repertoire of non-PCs. Therefore, we sorted splenocytes from OVA-sensitized BALB/c mice into CD138(pos) (PBs/PCs) and CD19(pos)/CD138(neg) (non-PCs) B cell fractions. Using reverse transcription PCR, we amplified, cloned, and sequenced IgE mRNA transcripts and anal

The proliferative rate of malignant plasma cells, as measured by the plasma cell labelling index (PCLI), is an important prognostic factor in multiple myeloma (MM); however, the PCLI alone is probably Inadequate to describe tumour growth because it ignores the idea that myeloma cells may have a reduced rate of apoptosis. The aims of this study were to develop a flow cytometric method to measure the apoptosis index of fresh marrow plasma cells and develop a plasma cell growth index (PCGI) that related both proliferation and apoptosis to disease activity. Marrow aspirates were obtained from 91 p…

Numerous IgG4 plasma cells and a high IgG4/IgG plasma cell ratio are important criteria in the diagnosis of IgG4-related sclerosing disease (IgG4-RSD), a steroid-responsive fibroinflammatory disorder. There is also a growing list of other inflammatory disorders that may mimic IgG4-RSD with many IgG4 plasma cells; however, limited data exist concerning whether plasma cell (Zoon) balanitis (PCB) is among these disorders. We, therefore, reviewed the clinical, histologic, and immunohistochemical features of PCB in 17 patients, including evaluation of CD3, CD20, CD138, κ, λ, IgG, IgG4, IgM, IgA, an…

Autoimmune hepatitis (AIH), post-transplant recurrent AIH (rAIH), and plasma cell-rich rejection (PCR) are clinical diagnoses with the shared histopathologic hallmark of plasma cell hepatitis (PCH). As these histologically and serologically indistinguishable diagnoses are differentiated by clinical context, it remains uncertain whether they represent distinct immunologic phenomena. Improved understanding of immunoglobulin subclass 4-producing plasma cells (IgG4-PC) has brought attention to IgG4 as an immunophenotypic biomarker. To date, degree and clinical significance of IgG4-PC infiltration …

BACKGROUND: Multiparameter flow cytometry (MFC) identification and characterization of plasma cells (PCs) is a useful tool to support diagnosis, prognostication, and monitoring of PC diseases (PCD). Currently, the number of MFC markers suited for the identification of PC remains limited. Moreover, antibody therapies against PC-associated markers further compromise the utility of the most widely used reagents (e.g., CD38). Despite markers other than CD38 and CD138 are recognized as potentially useful PC-identification markers, no study has comparatively evaluated their performance in combinatio…

Long-lived plasma cells (LLPCs) are primarily found in the CD19-CD38hiCD138+ plasma cell subset in the human bone marrow. They produce antibodies continuously for several years or even decades after initial infection or vaccination, and are essential for durable anti-infective immunity. Compared to short-lived plasma cells, LLPCs express higher levels of surface molecules such as CD27, CD38, CD138, C-X-C chemokine receptor 4 (CXCR4), interleukin 6 receptor (IL-6R), and B-cell maturation antigen (BCMA). However, specific markers for LLPCs have not been identified yet. Currently, transgenic mous…

Cell membranes undergo biophysical remodeling as an adaptation to the surroundings and to perform specific biological functions. However, the extent and relevance of such changes in human immune cells remain unknown, largely because of the lack of single-cell and multidimensional methodologies. Here we apply a cytometry-based method to fill this gap by combining biophysical profiling with simultaneous analysis of immune cell markers. This platform reveals notable cell-type-dependent plasma membrane order heterogeneity in immune cells. By sorting immune cells according to their membrane order a

Secreting a continuous, and sometimes life-long antibody supply, plasma cells are the effector arm of humoral immune system. With an incredibly diverse array of binding specificities, antibodies play critical roles in homeostasis and disease. Traditional views of plasma cells have them function at a distance, relying on circulation to ferry their antibodies to peripheral tissues. However, this review focuses on plasma cells that operate locally within tissues that lack ready access to circulating antibody, specifically, we explore plasma cells that accumulate within the central nervous system …

Plasma cells (PCs) are terminally differentiated B cells that are long-lived and have high rates of protein secretion. Because of these features, ex vivo generated PCs are being developed as therapeutics for long-term protein delivery. Here, we used proteomics and flow cytometry-based approaches to comprehensively interrogate the surface proteome of activated B cell versus PC populations. Next, we analyzed adhesion proteins that might facilitate engraftment of ex vivo generated PCs at candidate immune sites. We identified key adhesion proteins including integrin alpha 4 (ITGA4) and ITGB1 that …

Among the five antibody isotypes, immunoglobulin E (IgE) is unique in both its biological potency and clinical associations. Secreted by a specialized subset of plasma cells, IgE plays a critical role in host defense against helminths and other parasitic infections. Its effector functions are mediated through two main mechanisms: (1) opsonization of parasite surfaces, which enables eosinophil recognition of the Fc region of IgE, leading to eosinophil degranulation and parasite destruction; and (2) potent activation of mast cells and basophils through high-affinity binding to FcεRI. Among all i…

Circulating plasma cells (CPCs) represent an accessible subset of antibody-secreting cells that provide valuable insights into immune activation and regulation. This review presents the first comprehensive synthesis of CPC biology, with a particular focus on the mechanisms governing their generation under physiological conditions and the distinct pathways that drive their formation within tumor microenvironments. We further summarize the broader clinical relevance of CPCs as potential biomarkers across infections, autoimmune diseases, and plasma cell disorders. With the rapid advancement of li…

Persistent allergies affect the quality of life of patients and increase economic burdens. Many clinical observations indicate the presence of IgE+ long-lived plasma cells (LLPCs), which account for the persistent secretion of specific IgE; however, the characteristics of the IgE+ LLPCs have yet to be identified clearly. In this review, we summarized the generation of IgE+ PCs, discussed the prosurvival factors in the microenvironment, and reviewed the unique IgE-BCR signaling, which may bring insights into understanding the survival mechanisms of IgE+ LLPCs.

Three hundred thousand new cases of Lyme disease are diagnosed annually in the United States. The earliest manifestation of the disease, erythema migrans, occurs earlier than serologic conversion, and skin biopsies can be very helpful in suggesting the diagnosis. Histopathologic findings vary depending on where in the lesion the specimen is taken, but typically consist of a superficial and deep perivascular and interstitial lymphocytic infiltrate with eosinophils centrally and with histiocytes and plasma cells at the periphery. Rare cases with interstitial histiocytes and rare-to-sparse plasma…

Five pathogenic Candida species were compared in terms of their osmotolerance, tolerance to toxic sodium and lithium cations, and resistance to fluconazole. The species not only differed, in general, in their tolerance to high osmotic pressure (C. albicans and C. parapsilosis being the most osmotolerant) but exhibited distinct sensitivities to toxic sodium and lithium cations, with C. parapsilosis and C. tropicalis being very tolerant but C. krusei and C. dubliniensis sensitive to LiCl. The treatment of both fluconazole-susceptible (C. albicans and C. parapsilosis) and fluconazole-resistant (C

Abundant bone marrow angiogenesis is present in almost all myeloma patients requiring therapy and correlated to treatment response and survival. We assessed the expression of 402 angiogenesis-associated genes by Affymetrix DNA microarrays in 466 samples, including CD138-purified myeloma cells (MMCs) from 300 previously untreated patients, in vivo microcirculation by dynamic contrast-enhanced magnetic resonance imaging, and in vitro angiogenesis (AngioKit-assay). Normal bone marrow plasma cells (BMPCs) express a median of 39 proangiogenic (eg, VEGFA, ADM, IGF-1) and 28 antiangiogenic genes (eg,…

Plasma cells (PCs) develop from B lymphocytes following stimulation by antigen and express a genetic program aimed at the synthesis of immunoglobulins. This program includes the induction of genes coding for transcription factors such as PRDM1, X-box-binding protein 1 and BHLHB3, cell-surface molecules such as CD138/syndecan-1, and for the unfolded protein response. We review how the microarray technology has recently contributed to the understanding of the biology of this rare but essential cell population and its transformation into premalignant and malignant PCs.

The binding of fluvastatin, an inhibitor of hydroxymethylglutaryl coenzyme A reductase, to plasma proteins and red blood cells of rat, dog, and human in vitro was determined by ultrafiltration. Additionally, the stereospecificity of fluvastatin binding to proteins and the potential interaction between fluvastatin and the highly protein bound drugs warfarin, salicylic acid, and glyburide were investigated. Only a small fraction of fluvastatin in blood was taken up by the blood cells, amounting to 19-33% in the rat and < or = 15% in dog and humans. The plasma:blood fluvastatin ratio in these spe
