Fate of platinum metals in the environment.

For many years now automotive exhaust catalysts have been used to reduce the significant amounts of harmful chemical substances generated by car engines, such as carbon monoxide, nitrogen oxides, and aromatic hydrocarbons. Although they considerably decrease environmental contamination with the above-mentioned compounds, it is known that catalysts contribute to the environmental load of platinum metals (essential components of catalysts), which are released with exhaust fumes. Contamination with platinum metals stems mainly from automotive exhaust converters, but other major sources also exist

Platinum-induced autoantibodies target nucleoplasmic antigens related to active transcription.

Research on autoimmune diseases has revealed that autoimmunity can be induced by heavy metals such as mercury and gold. Following the introduction of platinum-containing catalytic converters in automobiles, the emission of platinum compounds constitutes an abundant environmental pollutant, however, potential immunological hazards resulting from platinum-containing emissions were not yet examined. In our previous studies on molecular mechanisms of heavy metal-induced autoimmunity, we showed a platinum-dependent subcellular redistribution of the autoantigen fibrillarin from the nucleolus to the …

Silica-Supported Mononuclear Methyl-Platinum Complexes Stabilized by Trimethylphosphine-Synthesis, Structure, and Reactions Under Reducing Conditions.

The chemistry of single-site platinum surface complexes with phosphine ligands is rarely understood, in contrast to its well-known chemistry in solution. This report describes the synthesis of a supported bis(phosphine)methylplatinum(II) complex on silicon dioxide (PtMe(PMe3)2/SiO2) and its reactivity with carbon monoxide, hydrogen, and under inert gas. The parent compound and reaction intermediates are characterized by IR-, solid-state NMR, and X-ray absorption spectroscopy. The sterically non-demanding monodentate phosphine ligands stabilize the platinum centers, allowing the formation of se

Selective targeting of pentoxifylline to hepatic stellate cells using a novel platinum-based linker technology.

Targeting of antifibrotic drugs to hepatic stellate cells (HSC) is a promising strategy to block fibrotic processes leading to liver cirrhosis. For this purpose, we utilized the neo-glycoprotein mannose-6-phosphate-albumin (M6PHSA) that accumulates efficiently in HSC during liver fibrosis. Pentoxifylline (PTX), an antifibrotic compound that inhibits HSC proliferation and activation in vitro, was conjugated to M6PHSA. We employed a new type of platinum-based linker, which conjugates PTX via coordination chemistry rather than via covalent linkage. When incubated in plasma or in the presence of t

Beyond platinums: gold complexes as anticancer agents.

The accidental discovery of the anticancer properties of cisplatin in the mid-1960s triggered the development of alternative platinum-based drugs. However, the platinum-based treatment of tumor diseases is massively hampered by severe side-effects and development of resistance. Sulfur-containing biomolecules play a significant role in platinum anticancer chemotherapy because of their high affinity to the platinum(II) ion. Sulfur is involved in the entire metabolic processing of platinum drugs. Strong and irreversible binding of cisplatin to intracellular thiolato ligands is considered a major

Synthesis, characterization, cytotoxic activity, and cellular accumulation of dinuclear platinum complexes derived from N,N'-di-(2-aminoethyl)-1,3-diamino-2-propanol, aryl substituted N-benzyl-1,4-butanediamines, and N-benzyl-1,6-hexanediamines.

This work describes the synthesis and characterization of six new dinuclear platinum complexes having N,N'-di-(2-aminoethyl)-1,3-diamino-2-propanol, aryl substituted N-benzyl-1,4-butanediamines and N-benzyl-1,6-hexanediamines as ligands. They were prepared by the reaction of cis-[PtCl(2)(DMSO)(2)] (DMSO=dimethyl sulfoxide) with the appropriate ligand in water, except for one of them, which was prepared from K(2)PtCl(4). We also report the cytotoxic activity and cellular accumulation of three of these complexes in a human small-cell lung carcinoma cell line and its resistant subline. Resistant

Mechanisms of Copper-Induced Autophagy and Links with Human Diseases.

As a structural and catalytic cofactor, copper is involved in many biological pathways and is required for the biochemistry of all living organisms. However, excess intracellular copper can induce cell death due to its potential to catalyze the generation of reactive oxygen species, thus copper homeostasis is strictly regulated. And the deficiency or accumulation of intracellular copper is connected with various pathological conditions. Since the success of platinum-based compounds in the clinical treatment of various types of neoplasias, metal-based drugs have shown encouraging perspectives f

Metals as mutagens.

A number of metals are mutagenic in bacteria or phage. These include compounds of arsenic, chromium, copper, iron, manganese, molybdenum, platinum, and selenium. Compounds containing alumium, antimony, arsenic, cadmium, copper, lead, mercury, nickel, and tellium have been shown to induce chromosomal aberrations or abnormal cell divisions in animal or plant cells. Genetic evidence suggests that arsenic, chromium, and molybdenum compounds may influence the accuracy of DNA repair processes in microorganisms.

Exploring the mechanisms of metal-based pharmacological agents via an integrated approach.

The peculiar chemical properties of metal-based drugs impart innovative pharmacological profiles to this class of therapeutic and diagnostic agents, most likely in relation to novel molecular mechanisms still poorly understood. However, inorganic drugs have been scarcely considered for medicinal applications with respect to classical organic compounds due to the prejudice of the relevant toxic effects evidenced in certain cases. Thus, the development of improved metallodrugs requires clearer understanding of their physiological processing and molecular basis of actions. Among the various issue

Comparative Binding Affinities of Platinum-Based Drugs toward Purine Alkaloids and Nucleobases.

Platinum-based anticancer drugs are among the most commonly used agents in cancer therapy. In particular, carboplatin, cisplatin, and oxaliplatin have been used in both monotherapies and combination therapies for many years. The main targets of these compounds are nucleobases in DNA. By creating cross-links, they inhibit cell development and lead to apoptosis. Recent studies indicate that platinum-(II) drugs can interact with other compounds with structures similar to nucleobases. For this reason, the current study analyzed the interactions of carboplatin, cisplatin, and oxaliplatin with nucle

Cisplatin reacts with the RING finger domain of RNF11 and interferes with the protein functions.

Protein reactions play important roles in the mechanism of action of cisplatin. In this work, we found that cisplatin is highly reactive to the RING finger domain of RNF11, a key protein involved in tumorigenesis and metastasis. The results show that cisplatin binds to RNF11 at the zinc coordination site and leads to zinc ejection from the protein. The formation of S-Pt(II) coordination and Zn(II) ions release have been confirmed by UV-vis spectrometry using zinc dye and thiol agent, showing reducing the contents of thiol groups while forming S-Pt bonds and releasing zinc ions. Oxidation of Dichloromethane over Au, Pt, and Pt-Au Containing Catalysts Supported on γ-Al2O3 and CeO2-Al2O3.

Au, Pt, and Pt-Au catalysts supported on Al2O3 and CeO2-Al2O3 were studied in the oxidation of dichloromethane (DCM, CH2Cl2). High DCM oxidation activities and HCl selectivities were seen with all the catalysts. With the addition of Au, remarkably lower light-off temperatures were observed as they were reduced by 70 and 85 degrees with the Al2O3-supported and by 35 and 40 degrees with the CeO2-Al2O3-supported catalysts. Excellent HCl selectivities close to 100% were achieved with the Au/Al2O3 and Pt-Au/Al2O3 catalysts. The addition of ceria on alumina decreased the total acidity of these catal
