Cryptic Interactions in a Marine Microcosm: Sponges Shape the Microbiome of a Resident Goby.

Microbial symbionts influence ecological and evolutionary processes in their multicellular hosts; yet they remain poorly characterised in marine systems. Sponges host exceptionally diverse microbiomes, potentially providing rich microbial reservoirs for their residents. The tusked goby (Risor ruber), an obligate sponge-dweller, and its sponge hosts provide an ideal model to investigate the causes and consequences of shared microbial taxa in host-resident systems. Using 16S rRNA sequencing, we characterised the microbiomes of R. ruber skin and gut, their sponge hosts, a non-obligate, sponge-ass

Complete ectosome formation from the choanosome during whole-body regeneration of the sponge Hymeniacidon heliophila (Wilson 1911) using an in vitro microexplant model.

The regeneration of the sponge Hymeniacidon heliophila was investigated using the in vitro microexplant model. This model differs from cell dissociation, surface ablation, and fragment culture models because it preserves the choanosome attached to the substrate, maintaining the apical-basal axis while entirely lacking the ectosome and the water flow polarity (ostia-osculum). The complete ectosome regenerative development was assessed using stereoscopy and scanning electron microscopy until complete adult formation. The observed stages of ectosome regeneration from the preserved choanosome alig

Fimsbactin Siderophores From a South African Marine Sponge Symbiont, Marinomonas sp. PE14-40.

Low iron levels in marine habitats necessitate the production of structurally diverse siderophores by many marine bacterial species for iron acquisition. Siderophores exhibit bioactivities ranging from chelation for iron reduction in hemochromatosis sufferers to antimicrobial activity either in their own right or when coupled to known antibiotics for targeted delivery or for molecular imaging. Thus, marine environments are a sought-after resource for novel siderophores that could have pharmaceutical or industrial application. The fimsbactins A-F (1-6) are mixed catechol-hydroxamate siderophore

Role of layered architecture in marine sponge root fibres: new lessons from nature for the design of tension cables.

Patterns found in structural materials of biological origin are an excellent source of inspiration for engineers. The root fibres (basalia spicules) of the marine sponge Euplectella aspergillum anchor it to the ocean floor and exhibit a lamellar architecture. It is generally thought that the spicule's architecture contributes to the spicule's fracture toughness. However, in recent experiments, the spicules' architecture did not contribute to their fracture toughness in a statistically significant way, with their fracture initiation toughness being similar to that of synthetic glass. In this ar

Fighting nature with nature: antiviral compounds that target retroviruses.

The human immunodeficiency virus (HIV) is a type of lentivirus that targets the human immune system and leads to acquired immunodeficiency syndrome (AIDS) at a later stage. Up to 2021, there are millions still living with HIV and many have lost their lives. To date, many anti-HIV compounds have been discovered in living organisms, especially plants and marine sponges. However, no treatment can offer a complete cure, but only suppressing it with a life-long medication, known as combined antiretroviral therapy (cART) or highly active antiretroviral therapy (HAART) which are often associated with

Enigmazole Phosphomacrolides from the Marine Sponge Cinachyrella enigmatica.

Enigmazole B (1) and four new analogues, cis-enigmazole B (2), dehydroenigmazole B (3), enigmimide B (4), and enigmimide A (5), were isolated from the marine sponge Cinachyrella enigmatica. Their planar structures were elucidated by detailed NMR and MS data analyses, which established 1-3 to be oxazole-substituted 18-membered phosphomacrolides, while 4 and 5 were oxazole ring-opened congeners. The relative and absolute configurations in 1 were determined by a combination of chemical transformations and spectroscopic analyses. Photooxidation of the oxazole moiety in 1 gave enigmimide B (4), thu

In situ determination of Si, N, and P utilization by the demosponge Tethya citrina: A benthic-chamber approach.

Sponges consume dissolved silicon (DSi) to build their skeletons. Few studies have attempted to quantify DSi utilization by these organisms and all available determinations come from laboratory measurements. Here we measured DSi consumption rates of the sponge Tethya citrina in its natural habitat, conducting 24h incubations in benthic chambers. Sponges consumed DSi at an average rate of 0.046 ± 0.018 μmol h-1 mL-1 when DSi availability in its habitat was 8.3 ± 1.8 μM. Such DSi consumption rates significantly matched the values predicted by a kinetic model elsewhere developed previously for th

Miuramides A and B, Trisoxazole Macrolides from a Mycale sp. Marine Sponge That Induce a Protrusion Phenotype in Cultured Mammalian Cells.

Morphology-guided cell-based screening of the extract of a Mycale sp. marine sponge led to the isolation of two trisoxazole macrolides, miuramides A (1) and B (2), which induced characteristic morphological changes in 3Y1 cells. The structure of 1 including absolute configuration was elucidated by a combination of the analysis of spectroscopic data, derivatization, and degradation. Both compounds exhibit potent cytotoxicity against 3Y1 cells.

Zampanolides B-E from the Marine Sponge Cacospongia mycofijiensis: Potent Cytotoxic Macrolides with Microtubule-Stabilizing Activity.

Four new compounds (2-5) structurally related to the microtubule-stabilizing agent (-)-zampanolide (1) have been isolated from the Tongan marine sponge Cacospongia mycofijiensis. Three of these new structures, zampanolides B-D (2-4), exhibit nanomolar cytotoxicity toward the HL-60 cell line, are antimitotic, and induce in vitro tubulin polymerization at levels comparable to 1. Zampanolide E (5), saturated at C-8/C-9, was significantly less potent and does not stabilize purified tubulin, even at 10-fold higher concentrations. The structural differences across these compounds reveal a plasticity

Allocation of trisoxazole macrolides in the sponge Penares cf. nux and the impact on epiphytic bacterial cohabitants.

The correlation between the allocation of trisoxazole macrolides in the capitums, appendages, and bases of the sponge Penares cf. nux and the surface-attached bacteria on the corresponding parts was examined. The kabiramide contents were highest in the capitums, followed by the appendages and bases. Conversely, direct counts of cultivable surface-attached bacteria showed that the bacteria aggregate more densely on the surfaces of the bases. This suggested the repelling effects of the kabiramides against the fouling bacteria, particularly on the capitums and appendages. Twenty-two bacterial str

Model system for cell adhesion mediated by weak carbohydrate-carbohydrate interactions.

The multivalent carbohydrate-carbohydrate interaction between membrane-anchored epitopes derived from the marine sponge Microciona prolifera has been explored by colloidal probe microscopy. An in situ coupling of sulfated and non-sulfated disaccharides to membrane-coated surfaces was employed to mimic native cell-cell contacts. The dynamic strength of the homomeric self-association was measured as a function of calcium ions and loading rate. A deterministic model was used to estimate the basic energy landscape and number of participating bonds in the contact zone.

Marine natural products as prototype agrochemical agents.

In the interest of identifying new leads that could serve as prototype agrochemical agents, 18 structurally diverse marine-derived compounds were examined for insecticidal, herbicidal, and fungicidal activities. Several new classes of compounds have been shown to be insecticidal, herbicidal, and fungicidal, which suggests that marine natural products represent an intriguing source for the discovery of new agrochemical agents.

Anti-Proliferative Potential of Secondary Metabolites from the Marine Sponge Theonella sp.: Moving from Correlation toward Causation.

Marine sponges have been recognized as a rich source of potential anti-proliferative metabolites. Currently, there are two sponge-derived anti-cancer agents (a macrolide and a nucleoside) isolated from the Porifera phylum, suggesting the great potential of this sponge as a rich source for anti-neoplastic agents. To search for more bioactive metabolites from this phylum, we examined the EtOAc extract of Theonella sp. sponge. We isolated seven compounds (1-7), including four 4-methylene sterols (1-4), two nucleosides (5 and 6), and one macrolide (7). Among them, theonellasterol L (1) was identif…

In vitro and in vivo antifilarial potential of marine sponge, Haliclona exigua (Kirkpatrick), against human lymphatic filarial parasite Brugia malayi: antifilarial activity of H. exigua.

The present study reports on the antifilarial activity of a marine sponge Haliclona exigua (phylum Porifera). The crude methanol extract and n-butanol-soluble fraction killed adult Brugia malayi at 31.25-microg/ml concentration (both in motility and 3-(4,5 dimethylthiazol-2-yl)-2,5 diphenyl tetrazolium bromide assay) while the chloroform fraction was lethal at a lower concentration of 15.6 microg/ml. The activity could be located in a single molecule araguspongin C which brought about mortality of worm at 15.6 microg/ml. In vivo evaluation of the crude extract (5 x 500 mg/kg, orally) and the c…

Immunosuppressive sesquiterpene phenols from the marine sponge Dactylospongia elegans.

Two new sesquiterpene phenols, dactyltone A (1) and dactyltone B (2), were isolated from the marine sponge Dactylospongia elegans collected from the South China Sea. Their structures were elucidated by comprehensive spectroscopic analysis (including NMR and HRESIMS) combined with experimental and calculated electronic circular dichroism (ECD) data. Notably, the carbonylation at C-2 observed in ring A of marine sponge-derived sesquiterpene phenols represents an uncommon structural feature. Biological evaluation revealed that both compounds showed selective immunosuppressive activity, specifical
