Hemoglobin stability: observations on the denaturation of normal and abnormal hemoglobins by oxidant dyes, heat, and alkali.

Several unstable mutant hemoglobins have alterations which affect areas of the molecule involved in the attachment of heme to globin. Loss o … ed denaturation to 12-20%. Precipitation was decreased to less than 3.5% by the presence of 0.015 M potassium cyanide. Increasing the ionic strength of the medium increased precipitation. Cyanide prevented the formation of inclusion bodies when red cells containing unstable hemoglobin Philly, beta35 tyr --> phe, were incubated with the redox dye new methylene blue. …

Characterisation of a cyanide hydratase gene in the phytopathogenic fungus Leptosphaeria maculans.

A gene encoding a cyanide hydratase was cloned from an aggressive isolate of Leptosphaeria maculans, the fungus which causes blackleg diseas … ses. Transcription of cyanide hydratase in an aggressive L. maculans isolate is induced strongly by potassium cyanide. Transcription of the gene is detectable in cotyledons of Brassica juncea and B. napus during infection. L. maculans can utilise the reaction product, formamide, as a sole source of nitrogen.

Deflocculation effects due to chemical perturbations in sequencing batch reactors.

Toxic shock-induced deflocculation was examined for activated sludge exposed to six different classes of industrially relevant chemical toxins: an electrophilic solvent (1-chloro-2,4-dinitrobenzene, CDNB), a heavy metal (cadmium), a hydrophobic chemical (1-octanol), an uncoupling agent (2,4-dinitrophenol, DNP), alkaline pH, and weakly complexed cyanide. The concentrations required to inhibit respiration by 50% were used to shock sequencing batch reactors (SBRs) containing a nitrifying (10-day solids retention time (SRT)) and a non-nitrifying (2-day SRT) biomass. Effluent total suspended solids

Acute cyanide Intoxication: A rare case of survival.

A 30-year-old male jewellery factory worker accidentally ingested silver potassium cyanide and was brought to the emergency department in a state of shock and profound metabolic acidosis. This patient was managed hypothetically with use of injection thiopentone sodium intravenously until the antidote was received. Cyanide is a highly cytotoxic poison and it rapidly reacts with the trivalent iron of cytochrome oxidase thus paralysing the aerobic respiration. The result is severe lactic acidosis, profound shock, and its fatal outcome. The patient dies of cardio-respiratory arrest secondary to dy…

Selective gold extraction from electronic waste using high-temperature-synthesized reagents.

For over two hundred years, cyanide has served as the primary reagent for gold extraction. However, due to its high toxicity, the use of cyanide poses significant risks. Traditional low-toxicity leaching reagents have limitations that restrict their widespread industrial application, leading to the necessity for the development of new, efficient, and low-toxic gold leaching reagents to support sustainable gold production. In this study, a novel, efficient, and low-toxicity gold extraction reagent was synthesized at high temperatures by combining urea, sodium carbonate, and a specific iron salt

Ferrate(VI) oxidation of cyanide in water.

Experiments were conducted to test removal of cyanide (free cyanide and several cyanide complexes) in water, under alkaline medium (pH > or = 11), by a new potassium ferrate salt. The removal rate of free cyanide by oxidation with Fe(VI) was greater at pH 11.0 than at pH 12.0. A complete oxidation was obtained with a 2.67 Fe(VI)/CN ratio at pH 11.0. In these conditions, the rate of cyanide oxidation by Fe(VI) was slow, with a reaction rate constant estimated at 0.95 +/- 0.10 s(-1) l mol(-1) at pH 11.0 and 19.6 degrees C in this study. This study revealed that Fe(VI) did not decompose all cyani

Transmetallation by Halogen-Cyanide Metathesis in Water: Application to Cyanide Ion-Free Palladium Catalyzed Cyanation.

Aryl nitriles are important synthons in organic chemistry, yet conventional cyanation methods mostly generate free cyanide ions, creating acute safety hazards and wastewater contamination issues. We report a Pd-catalyzed cyanide ion-free cyanation of aryl halides featuring a transmetallation potentially via halogen-cyanide (CN-) metathesis using potassium ferrocyanide as a bound CN- reservoir. Mechanistic studies, CN--sensing strip tests, and IR spectroscopy confirm that no free CN⁻ is released at any stage, eliminating toxicity and downstream CN- liabilities. Ferrate(VI) oxidation of zinc-cyanide complex.

Zinc-cyanide complexes are found in gold mining effluents and in metal finishing rinse water. The effect of Zn(II) on the oxidation of cyanide by ferrate(VI) (Fe(VI)O(4)(2-), Fe(VI)) was thus investigated by studying the kinetics of the reaction of Fe(VI) with cyanide present in a potassium salt of a zinc cyanide complex (K(2)Zn(CN)(4)) and in a mixture of Zn(II) and cyanide solutions as a function of pH (9.0-11.0). The rate-law for the oxidation of Zn(CN)(4)(2-) by Fe(VI) was found to be -d[Fe(VI)]/dt=k[Fe(VI)][Zn(CN)(4)(2-)](0.5). The rate constant, k, decreased with an increase in pH. The e

Metabolic inhibitors reversibly alter the basal membrane potential of the gecko retinal pigment epithelium.

The effects of metabolic inhibitors on the apical and basal membrane potentials were studied in the isolated retinal pigment epithelium of the lizard Gekko gekko. Adding dinitrophenol or cyanide or cooling the tissue to 15 degrees C first depolarized the apical membrane and then hyperpolarized the basal membrane. The basal hyperpolarization was accompanied by an apparent increase in basal resistance. These effects were fully reversible. Adding ouabain to inhibit specifically the apical Na(+)-K+ pump irreversibly depolarized the apical membrane but did not produce a basal membrane hyperpolariza

Utilization of 35S from radioactive methionine or sulfate in the detoxification of cyanide by rats.

Weanling rats were fed a semipurified diet based on 10% casein and 1,500 ppm potassium cyanide. The diet was supplemented with varying levels of DL-methionine and/or potassium sulfate such that all diets were iso-sulfurous except for the basal which had no additional sulfur. The animals were fed for 2 weeks after which half of the rats in all groups except the basal were used in a radioactive study. Urine was collected and analyzed for thiocyanate. Animals which received labeled methionine were more efficient in producing labeled thiocyanate than those which received labeled sodium sulfate, al…

Inhibition of mitochondrial respiration by sodium nitroprusside and the mechanism of cyanide liberation.

The influence of sodium nitroprusside (SNP) on mitochondrial respiration was examined in rat liver mitochondria. The addition of SNP 1 mmol litre-1 during state 3 respiration inhibited the oxygen uptake by 63.4%. A mixture of SNP 1 mmol litre-1 and glutathione (GSH) 1 mmol litre-1 inhibited the oxygen uptake more markedly (by 75.9%). The cyanide concentrations were 0.01 mmol litre-1 with SNP alone and 0.15 mmol litre-1 with the mixture of SNP and GSH. Cyanide production from SNP in the presence of various reducing agents was studied in potassium phosphate 0.1 mol litre-1 buffer solution (pH 7.

Sequelae of attempted suicide by cyanide ingestion: a case report.

A twenty-eight-year-old man presented emergently after ingesting 800 mg of potassium cyanide in a suicide attempt. He survived only with intensive medical and psychiatric intervention, and went on to develop severe parkinsonian symptoms, including profound micrographia and hypersalivation. Bilateral, symmetrical basal ganglial abnormalities were demonstrated with magnetic resonance imaging. Survival following cyanide poisoning is rare; the clinical, radiologic, and neuropathologic sequelae in other documented cases are reviewed.

Effect of temperature on the uptake and metabolism of cyanide by weeping willows.

Plants' uptake and metabolism of cyanide in response to changes in temperature was investigated. Pre-rooted weeping willows (Salix babylonica L.) were exposed to hydroponic solution spiked with potassium cyanide for 2-3 d. Ten different temperatures were used, ranging from 11 degrees C to 32 degrees C. Cyanide in water, plant tissue, and air was analyzed spectrophotometrically. The results revealed that significant amounts of the applied cyanide were removed from the aqueous solutions in the presence of plants. Small amounts of free cyanide were detected in plant materials in all treatments, b…

Blood cyanide levels in mice after administration of amygdalin.

Oral doses of amygdalin and intraperitoneal (i.p.) doses of potassium cyanide (KCN) in the near-lethal range were administered to CD2F1 female mice. Blood cyanide levels were then measured as a function of time. The maximum cyanide level after amygdalin administration was reached at about 11/2 to 2 h and was within the range of values seen after KCN administration. Behaviour of mice correlated with the time of maximum blood cyanide level. Acute distress was observed at times when the cyanide level was highest. There was great variability in the nature and magnitude of the response in individua…

Acute cyanide poisoning complicated by lactic acidosis and pulmonary edema.

Massive cyanide poisoning occurred in a 21-year-old man who had ingested 600 mg of potassium cyanide. The clinical course was marked by acute pulmonary edema and lactic acidosis. Because the poison was unidentified until nine hours after ingestion, the patient received only supported treatment which included diuresis, oxygen, bicarbonate, and assisted ventilation. A review of the literature shows that many case reports are poorly documented and do not provide a firm basis for evaluating therapy. To our knowledge, only four patients, including ours, have had blood levels of cyanide measured. In…

Oxidation of cyanide to the cyanyl radical by peroxidase/H2O2 systems as determined by spin trapping.

The cyanyl radical was formed during the oxidation of potassium or sodium cyanide by horseradish peroxidase, lactoperoxidase, chloroperoxidase, NADH peroxidase, or methemoglobin in the presence of hydrogen peroxide. The spin adducts of the cyanyl radical with 5,5-dimethyl-1-pyrroline-N-oxide and N-tert-butyl-alpha-phenylnitrone were quite stable at neutral pH. The identity of these spin adducts could be demonstrated using 13C-labeled cyanide and by comparison with the spin adducts of the formamide radical, a hydrolysis product of the cyanyl radical adduct. The enzymatic conversion of cyanide t

Availability of ferrocyanide and ferricyanide complexes as a nitrogen source to cyanogenic plants.

The effects of additional nitrogen on the toxicity and removal of ferrocyanide and ferricyanide by cyanogenic plants were investigated. Maize (Zea mays L. var. ZN 304) seedlings were grown in the hydroponic solutions with or without additional nitrogen, and amended with either potassium ferrocyanide or potassium ferricyanide at 25.0 +/- 0.5 degrees C for 144 h. Various physiological parameters were monitored to determine the responses of the plant seedlings to the exposure of these two chemicals. Potassium Amide-Promoted Direct Synthesis of α-Aryl Nitriles from α-Aryl Alkenes.

The synthesis of organic nitriles remains heavily dependent on methodologies that often involve transition-metal catalysts or toxic cyanide reagents, limiting their sustainability. Here, we report a cyanide- and transition-metal-free chemical looping strategy that converts α-aryl alkenes and ammonia (NH3) into α-aryl nitriles using potassium amide (KNH2) as a reaction mediator. The reaction initially proceeds via the formation of α-aryl nitrile potassium salts. Subsequent treatment of the potassium salts with NH3 liberates nitriles and regenerates KNH2, thereby closing the chemical loop. Combi
