Covalent adduction of serotonin-derived quinones to the SARS-CoV-2 main protease expressed in a cultured cell.

The SARS-CoV-2 main protease is an essential molecule for viral replication and is often targeted by medications to treat the infection. In this study, we investigated the possible inhibitory action of endogenous quinones on the enzyme. Recombinant SARS-CoV-2 main protease was exposed to tryptamine-4,5-dione (TD) or quinone from 5-hydroxyindoleacetic acid (Q5HIAA). As a result, the protease activity was considerably decreased in a dose-dependent manner. The IC50 values of the quinones toward the enzyme were approximately 0.28 μM (TD) and 0.49 μM (Q5HIAA). Blot analyses using specific antibodie…

Voltammetric method for determining the trace moisture content of organic solvents based on hydrogen-bonding interactions with quinones.

Voltammetry experiments were performed on the natural quinone, vitamin K(1) (VK(1)), in a range of organic solvents of varying dielectric constant that are commonly used for electrochemical measurements [dimethyl sulfoxide (DMSO), N,N-dimethylformamide (DMF), acetonitrile (MeCN), propionitrile (EtCN), butyronitrile (PrCN), 1,2-dichloroethane (DCE), dichloromethane (DCM), and 1,1,2,2-tetrachloroethane (TCE)]. The water content of the solvents was accurately measured using Karl Fischer (KF) coulometric titrations, and the voltammetric data were used to estimate the degree of hydrogen-bonding int

Quinone-Based Mediator Immobilized Mesoporous Electrodes for Bioelectrocatalysis of Glucose Dehydrogenase.

Bioelectrocatalysis, which couples enzymatic catalytic and electrode reactions, is the basic principle behind enzyme electrochemical biosensors, biofuel cells, and bioelectrosynthesis systems. Electron mediators facilitate the transfer of electrons between electrodes and oxidoreductases during catalytic reactions, so ideally the mediator remains near the electrode for higher catalytic efficiency. Additionally, electrode materials with three-dimensional space at the enzyme volume mesoscale are expected to improve electrode performance (e.g., increased current density and stability). In previous

Chemical Proteomics Reveal the Inventory of Pyrroloquinoline Quinone Binding Proteins in Bacteria.

Pyrroloquinoline quinone (PQQ) is a bacterial redox cofactor enabling enzyme catalysis in various sugar and alcohol dehydrogenases. However, its proposed additional role as a "longevity vitamin" lacks a clear molecular basis and is thus highly debated. Here, we applied chemical proteomics to identify previously unknown classes of PQQ-binding proteins. We designed and synthesized a structurally diverse suite of five PQQ probes equipped with a diazirine photo-cross-linker and an alkyne handle for target identification. The fidelity of the probes was first evaluated for two well-characterized bac

Pyrroloquinoline Quinone and NAD+ Metabolism in Glaucoma: A Molecular Rationale for Combined Neuroprotection.

Glaucoma is the leading cause of irreversible blindness worldwide and a paradigmatic age-related neurodegenerative disease in which retinal ganglion cells (RGCs) are selectively lost through mechanisms that extend beyond intraocular pressure. Age-dependent NAD+ depletion in RGCs, compounded by the progressive impairment of NAD+ biosynthesis and by the hyperactivation of NAD+-consuming enzymes under oxidative stress, defines a metabolic vulnerability that current pressure-lowering therapy does not address. Pyrroloquinoline quinone (PQQ) is a tricyclic ortho-quinone present in plant-derived food

Anti-inflammatory effects of para-quinone methide derivatives on ulcerative colitis.

A series of para-quinone methide derivatives were evaluated their anti-inflammatory activity. Through the screening of the lipopolysaccharide (LPS)-induced inflammatory cell model in Raw264.7 cells, it was found that the inhibitory activity of meta-substituted derivatives on NO production was superior to that of ortho- and para-substituted derivatives. Among them, in the inflammatory cell model, the meta-trifluoromethyl substituted para-quinone methide derivative 1i had the best activity in inhibiting LPS-induced excess generation of NO. And 1i could effectively inhibit the increase of ROS in

Sulfide-quinone oxidoreductase is required for cysteine synthesis and indispensable to mitochondrial health.

Mitochondrial dysfunction is related to common age-related disorders, including neurodegenerative diseases, metabolic syndrome, and carcinogenesis. Therefore, maintaining the functionality and integrity of mitochondria is important for human health. Herein, we found that sulfide:quinone oxidoreductase (Sqr), which oxidizes hydrogen sulfide to reactive sulfur species (RSS), was indispensable to mitochondria health in the eukaryotic model microorganism Schizosaccharomyces pombe. Sqr knock-out led to morphological changes and functional deficiencies of mitochondria and apoptosis in S. pombe. The

Labeling of Proteins by BODIPY-Quinone Methides Utilizing Anti-Kasha Photochemistry.

A novel approach for the photolabeling of proteins by a BODIPY fluorophore is reported that is based on an anti-Kasha photochemical reaction from an upper singlet excited state (Sn) leading to the deamination of the BODIPY quinone methide precursor. On the other hand, the high photochemical stability of the dye upon excitation by visible light to S1 allows for the selective fluorescence detection from the dye or dye-protein adduct, without concomitant bleaching or hydrolysis of the protein-dye adduct. Therefore, photolabeling and fluorescence monitoring can be uncoupled by using different exci

The effect of Pyrroloquinoline quinone and Resveratrol on the Survival and Regeneration of Cerebellar Granular Neurons.

The inability of central nervous system (CNS) to regenerate following traumatic brain injury (TBI) can be attributed to apoptotic cell death, inhibitory extrinsic environment, and the limited ability of neurons to regenerate. Thus, fostering the intrinsic regenerative potential and minimizing neuronal cell death could be a promising therapeutic approach. Pyrroloquinoline quinone (PQQ) was previously reported for its neuroprotective and regenerative potential on peripheral nerves. Here, we investigated the ability of PQQ to induce neurite re-growth in a wound healing model on cultured cerebella

Anti-inflammatory and Quinone Reductase-Inducing Compounds from Beilschmiedia mannii.

Previous studies on the therapeutic potential of plant species found in the diet of chimpanzees living in Taï National Park have shown that they could be potential candidates for the search of new molecules useful for humans. Based on the screening of some of these plants, the fruits of Beilschmiedia mannii, whose dichloromethane extract showed cancer chemopreventive properties, were selected. Bioactivity-guided fractionation of the extract resulted in the isolation and identification of two γ-pyrones, including desmethoxydihydromethysticin (1: ), found in a natural source for the first time,

Efficient Coupling Reaction of Quinone Monoacetal with Phenols Leading to Phenol Biaryls.

A simple and efficient synthesis of phenol biaryls by the cross-couplings of quinone monoacetals (QMAs) and phenols is reported. The Brønsted acid catalytic system in 1,1,1,3,3,3-hexafluoro-2-propanol was found to be particularly efficient for this transformation. This reaction can be extended to the synthesis of various phenol biaryls, including sterically hindered biaryls, with yields ranging from 58 to 90 % under mild reaction conditions and in a highly regiospecific manner.

Induction of quinone reductase and glutathione transferase in rat tissues by juglone and plumbagin.

The ability of the naturally-occurring naphthoquinone derivatives, juglone and plumbagin, to increase tissue activities of the Phase II detoxification enzymes quinone reductase (QR) and glutathione transferase (GT) has been investigated in rats. Groups of female Sprague-Dawley rats were dosed by oral intubation on 5 consecutive days with either juglone or plumbagin at 12.5, 25, 50, 75, 100 or 125 mumoles/kg/day. The animals were then killed and the activities of QR and GT determined in tissue homogenates. The naphthoquinone derivatives had no significant effect on enzyme activities in the live

Chemistry of puupehenone: 1,6-conjugate addition to its quinone-methide system.

The marine natural product puupehenone (1), isolated in good yields from sponges of the genus Hyrtios, has been shown to undergo stereospecific 1,6-conjugate addition to its quinone-methide system. Several nucleophilic agents such as hydrogen cyanide, Grignard reagents, and nitroalkanes were studied, producing structurally diverse compounds. This lead optimization study was initiated due to the bioactivity of puupehenone and its natural analogues, which includes numerous previous reports of potential anticancer and antiinfective activity.

Quinone mediated electron transport system in the filarial parasite Setaria digitata.

Setaria digitata, a cattle filarial parasite, is known to have peculiarities such as hydrogen peroxide (H2O2) production, cyanide insensitivity, absence of cytochromes and presence of quinones. Estimation of mitochondrial H2O2 with different substrates and inhibitors showed that salicylhydroxamic acid (SHAM), the alternative oxidase inhibitor, inhibited the H2O2 production maximally. Based on the inhibitory studies with rotenone, antimycin A, o-hydroxydiphenyl, SHAM and 2 thenoyltrifluoroacetone, a mechanism for the electron transport is proposed. Quinone Q8 seems to have a central role, hence…

Enantioselectivity in blood-cerebrospinal fluid barrier permeability of substituted p-phenylenediamine quinones in patients with leptomeningeal metastasis.

Substituted p-phenylenediamine quinones (PPD-Qs), widespread environmental contaminants arising from rubber antioxidants, exhibit documented toxicity and have been detected in human biological matrices. Penetration behaviors of various PPD-Qs across the blood-cerebrospinal fluid barrier remain unclear, particularly at the enantiomer-specific level. This study investigated this penetration behaviors using enantiomer-specific analysis of six PPD-Qs in 138 matched serum-cerebrospinal fluid sample pairs from Chinese adult patients with leptomeningeal. All targeted PPD-Q homologues were frequently …

p-Phenylenediamines and their derived quinones: A review of their environmental fate, human exposure, and biological toxicity.

p-Phenylenediamines (PPDs) are widely used as antioxidants in numerous rubber products to prevent or delay oxidation and corrosion. However, their derived quinones (PPD-Qs), generated through reactions with ozone, are ubiquitous in the environment and raise significant health and toxicity concerns. This review summarizes the current state of knowledge on environmental distribution and fate, human exposure, and biological toxicity of PPDs and PPD-Qs, and makes recommendations for future research directions. Although PPDs and PPD-Qs have been monitored in a variety of environmental matrices, stu…

Synthesis of polycyclic aromatic quinones by continuous flow electrochemical oxidation: anodic methoxylation of polycyclic aromatic phenols (PAPs).

The electrochemical oxidation of polycyclic aromatic phenols (PAPs) has been developed in a microfluidic cell to synthesize polycyclic aromatic quinones (PAQs). Methanol was used as nucleophile to trap the phenoxonium cation formed in the oxidation as an acetal, that later were hydrolysed to the quinone. Formation of hydrogen gas as the cathode reaction caused challenges in the flow cell and were overcome by recycling the reaction mixture through the cell at increased flow rate several times. The specific quinones formed were guided by the position of an initial hydroxy group on the polycyclic…
