Meta-analysis of the efficacy of amifostine in the prevention of cisplatin ototoxicity.

OBJECTIVE: The effectiveness of amifostine in the prevention of cisplatin ototoxicity remains controversial. The objective of this meta-analysis was to determine whether amifostine is successful in preventing ototoxicity secondary to cisplatin chemotherapy. DESIGN: Meta-analysis. METHODS: We conducted a systematic review of all randomized, controlled trials using amifostine in patients of all ages receiving cisplatin chemotherapy. Data extraction was performed by two independent reviewers using predefined data fields, including study quality indicators. Heterogeneity was evaluated using the

Ferulic acid alleviates ionizing radiation-induced developmental and neurobehavioral toxicity in zebrafish via modulation of apoptosis-related genes.

High-dose ionizing radiation (IR), as encountered in radiotherapy-related and radiological emergency contexts, induces severe acute developmental and neurobehavioral toxicity. However, safe and effective natural radioprotective agents remain limited. Ferulic acid (FA), a natural phenolic compound widely found in medicinal and edible plants, possesses well-established antioxidant and anti-inflammatory properties. Nevertheless, its protective effects and underlying mechanisms against high-dose IR-induced acute in vivo injury remain largely unclear. This study aimed to evaluate the in vivo protec

Melatonin as a radioprotective agent against flattening filter and flattening filter-free beam in radiotherapy-induced lung tissue damage.

BACKGROUND: Radiotherapy is a widely used treatment method in oncology, applied by delivering high-energy particles or waves to the tumor tissue. Although tumor cells are targeted with radiotherapy, it can cause acute or long-term damage to healthy tissues. Therefore, the preservation of healthy tissues has been an important subject of various scientific researches. Melatonin has been shown to have a radioprotective effect on many tissues and organs such as liver, parotid gland, brain, and testicles. This study aimed to evaluate the protective effect of melatonin against the radiation at vario

A historical perspective of radioprotection studies in India with a focus on plant-derived radioprotectors.

This article reviews the evolution of radioprotection research in major Indian laboratories over the past seventy years, tracing a systematic progression in the exploration of various protective agents. The significant contribution of premier Indian institutions dedicated to 'Radioprotection Development Programme' is highlighted. Early investigations focused on synthetic radiopro-tectors, particularly sulphydryl compounds, which demonstrated strong radioprotective effects in laboratory settings. However, their clinical application was limited due to severe tissue toxicity. Subsequent research

Metformin Protects against Radiation-Induced Acute Effects by Limiting Senescence of Bronchial-Epithelial Cells.

Radiation-induced damage to normal lung parenchyma remains a dose-limiting factor in thorax-associated radiotherapy (RT). Severe early and late complications with lungs can increase the risk of morbidity in cancer patients after RT. Herein, senescence of lung epithelial cells following RT-induced cellular stress, or more precisely the respective altered secretory profile, the senescence-associated secretory phenotype (SASP), was suggested as a central process for the initiation and progression of pneumonitis and pulmonary fibrosis. We previously reported that abrogation of certain aspects of t

Current concepts. Photoprotection.

Photoprotection encompasses all methods to prevent UV radiation (UVR) damage to the skin, including sunscreens, clothing, seeking shade, and duration and time of the day spent outdoors under UVR. As scientific research validates short- and long-term detrimental effects of UVR, physicians and the public must become increasingly aware of these problems to avoid them. Photoaging is defined. Choice of sunscreens, their Food and Drug Administration labeling, and future sunscreen products are reviewed. Hazards of UVR on the skin include acute sunburn, photocarcinogenesis, immunologic suppression, an

The effect of sunscreen on melanoma risk.

Total cumulative sun exposure is associated with the development of squamous cell and basal cell cancers, whereas intense intermittent sun exposure is associated with the development of melanoma. Exposure to UV radiation is the only known modifiable cause of melanoma, but the role of sunscreen in melanoma prevention remains somewhat controversial. This article discusses how UV radiation contributes to the pathogenesis of melanoma, how sunscreen modulates the action of UV radiation on the skin, and the effect of sunscreen on the risk of developing melanoma. A review of available sunscreen agent

[Prevention of occupational solar UV radiation-induced epithelial skin cancer].

Malignancies of the skin, with an incidence of more than 200,000 newly registered cases/year, are the most frequently notified malignances in Germany. In Europe, squamous cell carcinoma (SCC) and basal cell carcinoma (BCC) account for about 30 cases/100,000 persons and 50-100 cases/100,000 persons, respectively. Ultraviolet (UV) exposure is the main risk factor to induce these cancers. Increased incidence rates were shown for persons having red/blonde hair as well as light eye colour, acquire sun burns easily, hardly tan and develop freckles. The majority of the malignancies and precursor lesi

Taurine provides a time-dependent amelioration of the brain damage induced by γ-irradiation in rats.

Exposure to ionizing radiation (IR) is inevitable since over 80% of total average exposure comes from natural sources. Brain is vulnerable to the deleterious effects of IR. Therefore, scientists paid attention in identifying novel compounds to protect against radiation-induced brain injury. Adult male albino rats weighing 120-150 g were divided into five groups, 18 rats each. Group 1 served as control, group 2 received an oral daily dose of taurine (500 mg/kg) for 2 weeks. Group 3 was exposed to a whole body single dose of γ-irradiation (6 Gy). Groups 4 and 5 received taurine before and after

Molecular Influence of the ATM Protein in the Treatment of Human Cells with Different Radioprotective Drugs: Comparisons between Antioxidative and Pro-Episkevic Strategies.

The radiation protection strategy with chemical agents has long been based on an antioxidative approach consisting in reducing the number of radical oxygen and nitrogen species responsible for the formation of the radiation-induced (RI) DNA damage, notably the DNA double-strand breaks (DSB), whose subset participates in the RI lethal effect as unrepairable damage. Conversely, a DSB repair-stimulating strategy that may be called the "pro-episkevic" approach (from the ancient Greek episkeve, meaning repair) can be proposed. The pro-episkevic approach directly derives from a mechanistic model bas

Black Hoof Medicinal Mushroom Phellinus linteus (Agaricomycetes) Extracts Protect Against Radiation-Induced Hematopoietic Abnormality in Mice.

We investigated the effects of Phellinus linteus extracts (PLEs) against radiation damage in mice. First, BALB/c mice were irradiated once with γ-rays at 4, 5, 6, or 8 Gy and allowed to recover for 20 days. Results reveal that 8-Gy radiation caused death in 100% of mice on day 13, and 6-Gy radiation caused death in 86.7% of mice (13/15) at the end of the experiment, whereas 4- and 5-Gy radiation did not result in any death. We then used 5-Gy γ-ray radiation to examine the protective effects of PLEs. Mice were orally administered a PLE (500, 1000, and 1500 mg/kg) daily for 2 weeks before radiat

A Ternary Flavonoid Formulation Mitigates Fractional Radiation-Induced Brain Injury via Transcriptomic Reprogramming and Synaptic Protection.

UNLABELLED: Fractional ionizing radiation (FIR) is a standard cancer therapy but often induces severe central nervous system complications, including cognitive decline and physiological dysfunction. While natural flavonoids hold therapeutic promise for radiation-induced brain injury (RIBI), optimizing their combinations and elucidating the underlying molecular pathways remain challenging. METHODS: To develop a precise therapeutic strategy, we first integrated network pharmacology and UHPLC-Q-Orbitrap MS/MS analysis to identify three highly effective flavonoid monomers from a radioprotective b

Radiation proctitis in the rat. Sequential changes and effects of anti-inflammatory agents.

Female Wistar rats were treated with single exposure irradiation to 2 cm of distal colon to cause radiation proctitis. All animals were evaluated by examination, colonoscopy and histologic evaluation for changes post-irradiation. Exposures of 10, 12.5, 15, 17.5, 20, 22.5, 25, 27.5 and 30 Gy caused dose-related clinical and histologic changes peaking at 7 to 15 days post-exposure. Rats treated with 20 Gy were colonoscoped and biopsied daily and showed sequential post-irradiation endoscopic changes ranging from mucosal edema and mild inflammatory changes to erosion and ulcers. Histologically, cr

Intrarectal application of amifostine for the prevention of radiation-induced rectal injury.

Clinically symptomatic late injury to the rectal wall occurs in about one third of patients with prostate cancer treated with external beam irradiation. Reducing the physical dose to the anterior rectal wall without a similar reduction in the posterior peripheral zone is difficult because of the proximity of these structures. Based on our previous observations that intrarectal application of amifostine resulted in very high concentrations of amifostine and its active metabolite WR-1065 in the rectal wall of Copenhagen rats, the authors initiated a phase I clinical trial in 1998.
