[Immunizing and immunomodulating activity of a wild epidemic parotitis virus at a parotitis infection focus].

The immune status of 18 children previously vaccinated or not with live parotitis vaccine and living in a focus of parotitis was tested over time. Specific antibodies were found in the blood sera or nasal secretion of 88.9% children by follow-up days 10-20, and memory antigen-reactive cells were forming in 55.6%. Functional temporary T-immunodeficiency was observed at the same period in 66.7% children. Although there were no cases of clinically apparent epidemic parotitis, immunologic studies confirmed the circulation of wild type virus. Immunologic studies detected 2 cases of inapparent cours

[Indices of the humoral response to respiratory syncytial virus infection in children with acute bronchitis].

Examinations of 30 children with acute bronchitis have revealed a respiratory syncytial (RS) viral infection in 18. A reduced response to RS virus was seen in patients with bronchitis complicated by obstruction, in contrast to that in cases without obstruction. Humoral response to this virus was reduced and delayed in younger children as against that in elder ones. Even high levels of specific IgG did not result in vitro neutralization of RS virus.

Experimental respiratory syncytial virus pneumonia in young calves: microbiologic and immunofluorescent findings.

Young calves were inoculated with respiratory syncytial virus (RSV) intranasally or by a combined intranasal and intratracheal route and were killed between postinoculation (initial) days (PID) 1 and 14. Viral antigens were detected by immunofluorescence in nasopharyngeal cells from calves killed between PID 2 and 10. Evidence of infection of the trachea and lungs with RSV was obtained by immunofluorescence and virus isolation in calves inoculated by the combined route, but not in calves inoculated intranasally. Within the lungs, RSV antigens were observed in epithelial cells of bronchioli and

Atopy predisposing to acute bronchiolitis during an epidemic of respiratory syncytial virus.

Thirty-one infants admitted to hospital with acute bronchiolitis during an epidemic of respiratory syncytial virus were compared with a control group of 32 infants to establish whether the two groups differed in atopic background. Past history of respiratory illness, eczema, and present reactions to skin testing differed significantly between the two groups. Thus, infants with acute bronchiolitis had a significantly higher atopic predisposition than the controls.

Non-productive paramyxovirus infection: Nariva virus infection in hamsters.

The pathogenesis of infection with Nariva virus (NV)--recently classified as a paramyxovirus--was studied in the hamster, an animal closely related to the natural host. Intracranial inoculation of suckling hamsters produces an acute necrotizing encephalitis with large amounts of infectious virus and virus antigen in the brain. In contrast, weanling hamsters have only small amounts of infectious virus and only early in the disease, when they are well; later, when clinically ill, they have a non-productive infection with continuing evidence of viral antigen, but no detectable infectious virus. Rapid diagnosis of viral infections with fluorescent antisera.

Viral immunofluorescence tests were performed with commercial antisera on 1046 specimens of respiratory, conjunctival corneal and dermal origin to compare the diagnostic effectiveness of direct examination by immunofluorescence with culture. The fluorescence assays were found to be highly sensitive for respiratory syncytial and measles virus in nasopharyngeal aspirates from children, but less satisfactory for other respiratory viruses. The test detected 38% of the adenovirus positives from conjunctival specimens and 67% of the combined herpes simplex virus positives from the latter 3 groups. D

Viral pneumonia in older adults.

Viruses account for a substantial portion of respiratory illnesses, including pneumonia, in the elderly population. Presently, influenza virus A H3N2 and respiratory syncytial virus are the most commonly identified viral pathogens in older adults with viral pneumonia. As diagnostic tests such as reverse-transcription polymerase chain reaction become more widely used, the relative importance of additional viruses (such as parainfluenza, rhinoviruses, coronaviruses, and human metapneumovirus) will likely increase. Influenza virus should be considered as a cause of pneumonia during the winter mon

[Once more a discussion of the RSV affair].

During autumn- and winter epidemics respiratory syncytial (RS) virus accounts for the majority of respiratory infections in infants and young children. In case of an acute lower respiratory tract infection, RS virus can induce serious symptoms. These are age-dependent. The most important symptoms in babies and toddlers are dyspnea, wheezing, cyanosis and apneas. In the case of respiratory insufficiency or fatigue, as well as recurrent apneas, mechanical ventilation is required. Diagnosis can be made using a direct immunofluorescence technique with monoclonal antibodies. To control the risk of

[Severe respiratory syncytial virus infections. Study of 87 infants hospitalized in an epidemic].

During a winter epidemic, 87 infants were admitted to Necker-Enfants-Malades hospital with a severe respiratory syncitial virus (RSV) infection. These infants fell into two groups: 37 infants without any medical history and 50 showing an underlying pathology (immune deficiencies, heart disease, CNS disorders, digestive malformations, allergic manifestations). Of the 37 infants with no medical history, most were below the age of 6 months and the RSV infection was manifested clinically by bronchiolitis or bronchitis. Most of the infants in the other group were more than 6 months of age and prese

Virus-specific polypeptides of human parainfluenza virus type 4 and their synthesis in infected cells.

We have studied the structural components of human parainfluenza virus type 4A (PIV-4A) and identified some virus-specific polypeptides by immunoprecipitation with polyclonal and monoclonal antibodies followed by one- or two-dimensional SDS-PAGE. HN polypeptides existed as monomer, disulfide-linked dimer, and disulfide-linked larger oligomer in cells infected with PIV-4A. Interestingly, the nonreduced NP, the nonreduced fusion, and the reduced F1 proteins migrated as doublets. Two F1 polypeptides were derived from different F1 + 2 proteins which migrated separately under nonreducing condition.

Pulmonary infections with respiratory syncytial virus and the parainfluenza viruses.

Respiratory syncytial virus (RSV) and the parainfluenza viruses (PIVs) are the most important causes of acute lower respiratory illness (LRI) in infants and children under 6 years of age. These enveloped viruses are members of the paramyxovirus family. They infect cells in the epithelium lining the trachea and intrapulmonary airways, and cause croup, bronchitis, bronchiolitis, and bronchopneumonia. RSV causes annual midwinter to early spring outbreaks of respiratory disease in temperate climates; epidemics are heralded by the appearance of increased numbers of cases of bronchiolitis, primarily

Respiratory syncytial virus pneumonia in Friesian calves: physiological findings.

Clinical, serological and physiological values were measured during and after naturally occurring respiratory syncytial virus pneumonia in four Friesian calves weighing between 120 and 160 kg. Clinical and serological findings were in agreement with the description of spontaneous calf pneumonia, associated with respiratory syncytial virus infection. Pulmonary function values, recorded one, eight, 15 and 57 days after the clinical onset of the disease, provided good evidence that the clinical syndrome associated with respiratory syncytial virus infection in Friesian cattle is at first an acute

Incidence of RS virus infections in premature children's ward.

In the course of two years (1974-76) four outbreaks of acute respiratory disease in the premature children's ward of a Prague hospital were studied virologically and clinically. RS virus (RSV) was found to be the aetiological agent. The highest isolation rate of RSV was achieved when using two heteroploid cell lines (L-132 and HEp-2 cells) simultaneously. Of the 30 children examined, 60% showed a severe course of disease (pneumonia and/or bronchiolitis) while in 40% of the children the disease had the form of rhinitis with striking abundance of whitish foamy secretions. In one of the outbreaks

Inhibition of multiplication of parainfluenza 3 virus in prostaglandin-treated WISH cells.

Influence of prostaglandins E2 (PGE2) and F2alpha (PGF 2alpha) on multiplication of myxovirus parainfluenza 3 was investigated. At concentrations of 0.01--1 mug/ml prostaglandins had no direct cytotoxic effects. PGE2 and PGF2alpha inhibited multiplication of parainfluenza 3 virus at concentrations 0.1--10 mug/ml. The inhibitory effect was most pronounced when the prostaglandins were added to medium for the whole period of virus multiplication (48 hours) while little or no effect was found when prostaglandins were added before virus inoculation or for 2 hours after infection. It is suggested th

Landscape of Prophylactic Strategies Against Human Parainfluenza Virus Type 3.

Human parainfluenza virus-type 3 (HPIV3) is a respiratory virus responsible for mild illnesses in most of the population and severe illnesses, such as bronchiolitis and pneumonia, in young children, immunocompromised individuals and the elderly. To date, no vaccines nor antiviral drugs have been approved against HPIV3, despite its significant burden in vulnerable people. In this review, we present both past and current prophylactic strategies against HPIV3, such as live-attenuated virus, vector-based, subunit and mRNA vaccine candidates or antibody-based passive protection. For each strategy,

Air in All the Wrong Places.

Extraluminal air can occur through a wide variety of mechanisms. Often, the free air resides in isolated regions including the thorax, the peritoneum, or the mediastinum. We present a pediatric case where there was extensive extraluminal air simultaneously within several regions, one of which has never been reported in the literature.
