An ultrastructural study has been made of a symbiotic association between a sulfur bacterium and the roots of the rice plant (Oryza saliva L.). This association is proposed to have useful economic consequences in ameliorating hydrogen sulfide toxicity and associated Akiochi or Straighthead disease in lowland rice cultivation. The presence of catalase (E.C. 1.11.1.6) in rice roots and in some rhizosphere bacteria has been demonstrated in ultrathin sections of field-grown rice using a catalasespecific medium. Catalase was detected in capsules surrounding the catalase-negative S-bacteria. These c…

The soil microbiome drives soil organic carbon (SOC) transformation, shaped and impacted by plant growth stage and soil management such as tillage. The soil microbial carbon pump conceptually links the degradation of plant- and microbe-derived compounds and the neosynthesis of microbial biomass, as a driver of SOC buildup. Using metatranscriptomic sequencing, we characterized actively expressed microbial functions associated with the microbial carbon pump in the Brassica napus rhizosphere across growth stages and under simulated erosion, using a tailored KEGG Orthology (KO) approach complement…

Rhizosphere microbiomes and metabolomes are influenced by host genotype and developmental stage. However, there has been limited research that simultaneously profiles the rhizosphere microbiome and metabolism of rice across different genotypes and developmental stages. Furthermore, the interactions between rhizosphere microbiomes and metabolism in various plant genotypes and developmental stages are not well understood. Here, we investigated the diversity and composition of rhizosphere microbial community and metabolism across three hybrid rice varieties and their respective parent lines durin…

The rhizosphere, a dynamic interface shaped by plant root exudates, fosters microbial communities with significant biochemical potential. This study investigated the interplay between soil properties and fungal bioactivity in the rhizospheres of Withania coagulans and Justicia adhatoda in Pakistan. Physicochemical analysis revealed silty loam textures with divergent phosphorus [25.7 vs. 71.5 mg/kg] and potassium [108 vs. 78 mg/kg] levels, alongside near-neutral pH, influencing microbial dynamics. Two fungal isolates, Aspergillus luchuensis and A. flavus, were identified through morphological t…

BACKGROUND: Plant biostimulants are an emerging class of agricultural inputs known to enhance plant growth and improve their tolerance to abiotic stress. While biostimulants are widely used, their mechanisms of action remain poorly understood. This study investigates the effects of an Ascophyllum nodosum-based biostimulant (ANE) on the rhizosphere bacterial communities of corn (Zea mays).
RESULTS: Root exudates from ANE root-treated plants promoted the swarming motility of Pseudomonas protegens (CHA0), a plant growth-promoting rhizobacterium. Gene expression analysis showed that root exudates…

Long-term fertilization can result in the changes in carbon (C) cycle in the maize rhizosphere soil. However, there have been few reports on the impacts of microbial regulatory mechanisms on the C cycle in soil. In the study, we analyzed the response of functional genes that regulate the C fixation, decomposition and methane (CH4) metabolism in maize rhizosphere soil to different fertilization treatments using metagenomics analysis. As the dominant C fixation pathway in maize rhizosphere soil, the abundance of the functional genes regulating the reductive citrate cycle (rTCA cycle) including k…

Plant growth promoting rhizobacteria produce chemical compounds with different benefits for the plant. Among them, HCN is recognized as a biocontrol agent, based on its ascribed toxicity against plant pathogens. Based on several past studies questioning the validity of this hypothesis, we have re-addressed the issue by designing a new set of in vitro experiments, to test if HCN-producing rhizobacteria could inhibit the growth of phytopathogens. The level of HCN produced by the rhizobacteria in vitro does not correlate with the observed biocontrol effects, thus disproving the biocontrol hypothe

The diversity and antifungal activity of fluorescent pseudomonads isolated from rhizospheres of tea, gladiolus, carnation and black gram grown in acidic soils with similar texture and climatic conditions were studied. Biochemical characterisation including antibiotic resistance assay, RAPD and PCR-RFLP studies revealed a largely homogenous population. At soil pH (5.2), the isolates exhibited growth with varying levels of siderophore production, irrespective of crop rhizospheres. Two isolates with maximum chitinase production showed antagonism. The bacterial populations in general lacked the ab…

Ubiquitous arsenic (As) nanophases in soils are metastable due to the size effect and become readily available to rice under flooded conditions, making substantial contributions to the As pool. Here, the chemical characteristics of As nanophases in the rhizosphere as a hotspot for rice-soil-microbe interactions were fingerprinted at a single-particle resolution, revealing their chemical speciation and interactions with soil microbiota. The initial As particle characteristics (speciation and size) governed bioavailability through distinct transformation pathways mediated by particle-specific el…

Microbiome-guided crop protection increasingly emphasizes the chemical basis of disease suppression. Here, we identify Streptomyces aurantiacus YS-4, a rhizosphere-enriched actinobacterium selectively recruited by resistant cultivars of Salvia miltiorrhiza, as a producer of metacycloprodigiosin, a previously reported secondary metabolite that is reported for the first time in this strain and further investigated for its role in suppressing Fusarium oxysporum-induced root rot in S. miltiorrhiza. Metacycloprodigiosin inhibited Fusarium oxysporum growth by 81.66% at 200 μg mL-1 and induced extens…

The rhizosphere is a key ecological niche where plants interact with microorganisms, and its health directly affects plant growth, development, and disease resistance. Existing theories have laid an important foundation for understanding plant-microbe interactions. Among them, the biological market theory interprets the mutualistic symbiosis between plants and microorganisms from the perspective of nutrient exchange, offering valuable insights into resource flow and interaction mechanisms within the rhizosphere. On this basis, this study further proposes the conceptual model of 'biological cor…

Arsenic (As) contamination in agroecosystems poses significant risks to food security and human health. The mechanisms of As speciation change in soil within wetland rhizospheres are understood, but their location-precise importance within heterogeneous root-associated microbiomes is uncertain. While microbial processes are often considered dominant drivers of As redox transformations, the role of abiotic factors such as reactive oxygen species (ROS) remains underexplored due to limited in situ evidence. Here, we combined multiple high-resolution in situ techniques to map microscale distributi…

To elucidate genotype-associated differences in rhizosphere microbial community assembly, this study compared the microbiomes of three Fusarium wilt-resistant and three susceptible watermelon cultivars using amplicon sequencing. Results revealed distinct bacterial and fungal community structures between the two groups. Notably, resistant cultivars harbored a higher number of unique operational taxonomic units and displayed greater fungal richness compared to their susceptible counterparts. Beyond taxonomic composition, co-occurrence network analysis demonstrated that the fungal community withi…

The rhizosphere, the zone around the roots of plants, including the root, the soil on the root itself, and the surrounding area soil, is the hotspot for the production of reactive oxygen species (ROS). Functioning as redox signaling mediators, rhizosphere ROS regulate a series of key processes in the rhizosphere microenvironment. These biological and geochemical events cover root morphogenesis, the formation of beneficial plant-microbe symbioses, and element biogeochemical transformations. Nevertheless, the pronounced spatial heterogeneity, drastic redox fluctuations, and intricate interfacial…

Foliar application of metal micronutrients is increasingly adopted in intensive cultivation systems, yet its potential ecological risks to rhizosphere functions remain poorly understood. Here, using the medicinal plant Panax notoginseng as a model, we conducted a gradient foliar amendment experiment with iron (Fe), zinc (Zn), and copper (Cu) to evaluate how aboveground metal inputs regulate rhizosphere soil multifunctionality (MF) through microbial life-history strategies. By integrating 16S rRNA amplicon sequencing, metagenomics, root transcriptomics, and a newly developed quantitative Yield-…

Plant-microbe symbiotic relationships drive ecosystem evolution. This study employed metabolomics and metagenomic technologies to investigate the effects of the aboveground-restricted endophytic fungus Epichloë guerinii in the host plant Melica transsilvanica on the rhizosphere microbial community structure and functional traits. Our results revealed that the presence of E. guerinii significantly increased the secretion of organic acids, amino acids, and sugar alcohols from the host root system. These exudates correlated strongly with abundant, plant growth-promoting rhizosphere microorganisms…

With the rapid expansion of industrial activities, the accumulation of heavy metals in the environment has become a serious threat to ecological security and public health. Rhizosphere microorganisms play a crucial role in supporting the growth and quality of medicinal plants by facilitating nutrient uptake and regulating hormonal balance. However, medicinal plants can absorb heavy metals from contaminated soils during growth, resulting in toxic metal accumulation in plant tissues and reduced efficacy of active compounds. At the same time, excessive heavy metal levels suppress rhizosphere micr…
