Outcome of photodynamic therapy with Rose Bengal in conjunction with topical PHMB and chlorhexidine combination in Acanthamoeba keratitis.

PURPOSE: To report the outcome of Acanthamoeba keratitis, with early addition of Photo-dynamic antimicrobial therapy with Rose Bengal (PDAT-RB) to the medical treatment (combination of 0.02% Polyhexamethylene Biguanide (PH)and 0.02% chlorhexidine(CH)). METHODS: Patients were recruited based on the infiltrate size being < 8 mm and limited to the mid stroma, < 300µ, and confirmed microbiological diagnosis. Along with the continuation of PHMB + CH, patients were also treated with PDAT-RB twice with a gap of one week using 0.1% w/v RB and green LED (525 nm) array immediately after the confirmatio…

Development of Glycerol-Rose Bengal-Polidocanol (GRP) foam for enhanced sclerosis of a cyst for cystic diseases.

Polycystic kidney disease (PKD) is a common genetic disorder that results in a proliferating and enlarging cyst and ultimately leads to loss of kidney function. Because an enlarged cyst is a primary factor for limited kidney function, the large cyst is surgically removed by laparoscopic deroofing or sclerosant. This a relatively nascent treatment method entails complications and sometimes fail due to the cyst fluid refilling and infection. This study proposes using a more stable and effective polidocanol foam with glycerol and Rose Bengal (GRP form) to prevent cyst regeneration and irritation,…

Rose bengal staining in detection of oral precancerous and malignant lesions with colorimetric evaluation: a pilot study.

Early detection of oral precancerous and malignant lesions is still a diagnostic challenge for most of clinicians, and ideal adjuncts for detection of these lesions are currently unavailable. Our preliminary study has indicated that rose bengal (RB) staining might have the potency as a diagnostic aid; however, its clinical significance and reliability in hospital-based population are still not clear. In the present study, we investigated the efficacy of RB staining in detection of oral precancerous and malignant lesions. RB staining was conducted in 132 patients, and staining results were dete…

Rose Bengal acetate-mediated photodynamic therapy alleviates Cutibacterium acnes-induced acne-like lesions in a mouse model.

Acne vulgaris is a wordwide distressing skin condition which is often treated by topical antibiotics or retinoids, and in severe cases with oral medications. It is generated by a series of factors including excess sebum production clogging sebaceous glands and infection with Cutibacterium acnes producing inflammation. Photodynamic therapy (PDT) with topical aminolevulanic acid or its methyl ester plus red light has shown good results, especially for inflammatory lesions. The present study investigated the use of a new photosensitzer Rose Bengal acetate (RBAc) for acne PDT treatment. RBAc plus …

Accessing oxy-functionalized N-heterocycles through rose bengal and TBHP integrated photoredox C(sp3)-O cross-coupling.

Herein, we report a practical and simple mono- and di-C(sp3)-O cross-coupling of tautomerizable N-heterocycles (dihydrophthalazine-1,4-diones, pyridone, quinoxalinone and pyrimidinone) with ketones, β-dicarbonyl compounds and nitroalkane, leading to substituted imidate derivatives under visible-light conditions. The combination of rose bengal as the photocatalyst and TBHP enables sustainable reaction conditions, operational simplicity, and high chemo- and regioselectivity with exceptional yields (up to 94%), good functional group tolerance and substrate generality. In the case of unsymmetrical…

Photodynamic anti-biofilm PHEMA/Rose Bengal hydrogels enhanced by iodide: evidence for hydrogen peroxide and triiodide formation.

Biofilm-associated infections remain a major challenge for biomedical devices, motivating the development of light-activated antimicrobial materials. Here we report the antibiofilm activity (against Staphylococcus aureus and Pseudomonas aeruginosa) of a photodynamic hydrogel consisting of Rose Bengal (RB) physically entrapped within a crosslinked poly(2-hydroxyethyl methacrylate) (PHEMA) matrix. Under visible-light irradiation, RB-PHEMA alone produced only modest reductions (ca. 1 log), whereas addition of iodide markedly enhanced antibiofilm performance, yielding a 4.2-log reduction for S. au…

Rose Bengal-functionalized chitosan spheres as singlet oxygen generators for photodegradation of water contaminants and photodynamic inactivation.

Millimeter-sized chitosan spheres covalently functionalized with Rose Bengal (ME-CS-RB) were developed as reusable heterogeneous photosensitizers capable of generating singlet oxygen (1O2) under visible-light irradiation. Structural and spectroscopic characterization confirmed the effective immobilization of the photosensitizer within the chitosan matrix without compromising its integrity. The photoactivity of ME-CS-RB was evaluated through the photosensitized degradation of furosemide as a model water contaminant under green LED irradiation, with scavenger experiments indicating that 1O2 is t…

Rose Bengal Electromagnetic Activation With Green Light for Infection Reduction: Follow-Up of a Randomized Clinical Trial.

IMPORTANCE: Infectious keratitis results in poor outcomes despite maximal medical therapy. While rose bengal photodynamic therapy (RB-PDT) has not been shown to be beneficial at improving best spectacle-corrected visual acuity (BSCVA) or cure rate, decreasing scar size, or complication rate at a shorter period of 6 months, long-term results are essential to evaluate clinically relevant scarring and visual recovery. OBJECTIVE: To evaluate 12-month outcomes from the clinical trial (REAGIR) comparing adjunctive rose bengal photodynamic therapy (RB-PDT) vs sham in treatment of fungal, acanthamoeb…

Role of reactive oxygen species in cardiac preconditioning: study with photoactivated Rose Bengal in isolated rat hearts.

UNLABELLED: Oxygen radical scavengers have been shown to prevent the development of ischemic preconditioning, suggesting that reactive oxygen species (ROS) might be involved in this phenomenon. In the present study, we have investigated whether direct exposure to ROS produced by photoactivated Rose Bengal (RB) could mimic the protective effects of ischemic preconditioning. METHODS: In vitro generation of ROS from photoactivated RB in a physiological buffer was first characterised by ESR spectroscopy in the presence of 2,2,6,6-tetramethyl-1-piperidone (oxoTEMP) or 5,5-dimethyl-1-pyrroline-N-ox…

Rose bengal score--a possible key parameter when evaluating disease level and progression in primary Sjögren's syndrome.

The rose bengal score is one of the most commonly used tests for evaluation of ocular surface epithelial damage. The test is used in most Sjögren's syndrome criteria. We examined 24 female and four male patients with primary Sjögren's syndrome (primary SS) in order to evaluate possible correlation between the various tests for keratoconjuncivitis sicca (KCS), and for possible correlations to xerostomia and p-IgG levels. Among the KCS tests a high rose bengal score appeared to be the key parameter, being correlated to low break-up time (P less than 0.01), low tear lysozyme (P less than 0.01), a…
