Graphene oxide enhances aphid resistance in sorghum via the miR319-SbTCP7-SbLOX3 Pathway.

The aphid (Melanaphis sacchari) has emerged as a formidable pest, devastating sorghum plants and highlighting the need for sustainable management strategies. Graphene oxide (GO), as a novel material, has garnered attention for its use in crop cultivation and management, but its effects on biotic stresses remain elusive. Here, we used 10 mg/L GO to spray aphid-stressed sorghum seedlings four times in total. GO exposure reduced 50% H2O2 from the reactive oxygen species (ROS) burst induced by the aphid. Further analysis revealed that GO within the cells acts as a nanozyme, mimicking and enhancing…

Valorisation of insect infested sweet sorghum reeds towards production of a fermented beverage.

Sweet sorghum variety (Sorghum bicolour (L)) commonly known as sweet reeds, Ntšhe, in Setswana, is a valuable cash crop mostly for small scale farmers in Botswana and other southern African countries. These reeds are widely consumed as a delicacy and contribute significantly to food security, employment, and rural incomes. However, infestations by the larval stages of Chilo partellus (stem borer moths) lead to substantial economic losses, as consumers reject worm-infested reeds. To mitigate these losses, valorisation of condemned sweet reeds is attractive. Here, we took advantage of our unders…

Genome-wide identification and characterization of greenbug-inducible NAC transcription factors in sorghum.

BACKGROUND: Sorghum (Sorghum bicolor) is an important cereal crop grown worldwide because of its multipurpose uses such as food, forage, and bioenergy feedstock and its wide range of adaption even in marginal environments. Greenbug can cause severe damage to sorghum plants and yield loss. Plant NAC transcription factors (TFs) have been reported to have diverse functions in plant development and plant defense but has not been studied in sorghum yet. METHODS AND RESULTS: In this study, a comprehensive analysis of the sorghum NAC (SbNAC) gene family was conducted through genome-wide analysis. A …

Comparative Transcriptome Analysis Reveals Genetic Mechanisms of Sugarcane Aphid Resistance in Grain Sorghum.

The sugarcane aphid, Melanaphis sacchari (Zehntner) (Hemiptera: Aphididae) (SCA), has become a major pest of grain sorghum since its appearance in the USA. Several grain sorghum parental lines are moderately resistant to the SCA. However, the molecular and genetic mechanisms underlying this resistance are poorly understood, which has constrained breeding for improved resistance. RNA-Seq was used to conduct transcriptomics analysis on a moderately resistant genotype (TAM428) and a susceptible genotype (Tx2737) to elucidate the molecular mechanisms underlying resistance. Differential expression …

Matrix-Assisted Laser Desorption/Ionization-Mass Spectrometry Imaging of Metabolites during Sorghum Germination.

Dhurrin is the most abundant cyanogenic glucoside found in sorghum (Sorghum bicolor) where it plays a key role in chemical defense by releasing toxic hydrogen cyanide upon tissue disruption. Besides this well-established function, there is strong evidence that dhurrin plays additional roles, e.g. as a transport and storage form of nitrogen, released via endogenous recycling pathways. However, knowledge about how, when and why dhurrin is endogenously metabolized is limited. We combined targeted metabolite profiling with matrix-assisted laser desorption/ionization-mass spectrometry imaging to in…

Effects of sorghum variety on fermentation microecology and flavor characteristics of sesame-flavor Baijiu.

Sorghum, the principal raw material for Baijiu fermentation, plays a key role in shaping fermentation microecology and flavor formation. However, the effects of sorghum varietal differences on sesame-flavor Baijiu fermentation remain insufficiently understood. In this study, three sorghum varieties, Hongyingzi (R), Hei'e (B), and a domestic cultivar (G), were compared over two consecutive fermentation rounds. Fermented grain (Jiupei) samples collected at 0, 15, and 50 days were analyzed for physicochemical properties, enzyme activities, microbial communities, volatile compounds in Jiupei and b…

Effects of Aspergillus niger on cyanogenic glycosides removal and fermentation qualities of ratooning sorghum.

INTRODUCTION: Cyanogenic glycosides (CNglcs) are bioactive plant products involving in plant defense against herbivores by virtue of their abilities to release toxic hydrogen cyanide (HCN). Aspergillus niger has been shown to be effective in producing β-glucosidase, which could degrade CNglcs. However, whether A. niger could remove CNglcs under ensiling conditions is still unknown. METHODS: In this study, we first investigated the HCN contents in ratooning sorghums for two years, then the sorghums were ensiled with or without the addition of A. niger. RESULTS: Two years' investigation indica…

Epicuticular wax of sweet sorghum influenced the microbial community and fermentation quality of silage.

Epicuticular wax, as secondary metabolites (hydrophobic compounds) covering plant surface, plays important roles in protecting plants from abiotic and biotic stresses. However, whether these compounds will influence fermentation process of silage is still not clear. In this study, two sweet sorghum cultivars with varying epicuticular wax on sheath (bloom), Yajin 2 (YJ, less bloom), and Jintian (JT, dense bloom), were harvested at flowering and maturing stages, and ensiled with or without bloom, aiming to evaluate the effects of bloom on fermentation quality, feed nutrition and microbial commun…

An investigation into the influence of age on the standardized amino acid digestibility of wheat and sorghum in broilers.

Standardized ileal digestibility coefficients (SIDC) of nitrogen (N) and amino acids (AA) in wheat and sorghum at 6 different ages (d 7, 14, 21, 28, 35, and 42) of broilers were determined. Two assay diets were formulated to contain 93.8% of each grain as the sole source of AA in the diet. Titanium dioxide (0.5%) was added as an indigestible marker. Each assay diet was fed to 6 replicate cages housing 14 (d 7), 12 (d 14), 10 (d 21), 8 (d 28), 8 (d 35), and 6 (d 42) birds per cage for 4 d prior to ileal digesta collection. The apparent ileal digestibility coefficients (AIDC) were standardized b…

Pangenomic characterization and grain-development expression analysis of the OVATE gene family in sorghum.

The OVATE family proteins (OFPs) are plant-specific regulators that modulate organ morphogenesis and grain development. To elucidate their roles in sorghum (Sorghum bicolor L.) grain development, we performed a pan-genome-informed comparative analysis of the OFP family using 17 published sorghum genomes and identified 498 SbOFP genes. These 498 genes were classified into 35 SbOFP orthologous gene families, which were grouped into 5 Core, 9 Softcore, and 21 Shell families based on presence/absence variation (PAV). Collinearity and Ka/Ks analyses showed that 2 Softcore (OFP7, OFP13) and 3 Shell …

Replacing maize partially or totally with sorghum alters mycotoxin profile in their silages in microsilos.

Heat- and drought-tolerant sorghum is an attractive alternative to maize, yet its silage mycotoxin trade-offs remain unclear. We compared maize (Zea mays L.), sorghum (Sorghum bicolor (L.) Moench; cultivars Goliath and Sucrosorgo 506) and 1:1 maize-sorghum silages after 60 d ensiling (microsilo scale), with or without a commercial multi-strain lactic acid bacteria (LAB) inoculant. All silages fermented well (pH 4.06 - 4.21; lactic acid 18.1 - 27.9 g/kg dry matter). Maize silage showed the highest Fusarium toxins-deoxynivalenol (DON) 499 - 516 and nivalenol (NIV) 173 - 216 μg/kg dry matter. In …

Comparative genomics and stress-responsive expression of the IMP gene family in Sorghum bicolor.

Inositol monophosphatase (IMP) is a key enzyme in the inositol signaling pathway and ascorbic acid (AsA) biosynthesis, playing crucial roles in plant growth, development, and abiotic stress responses. Sorghum (S. bicolor) is an important cereal crop with remarkable drought and salinity tolerance, yet a systematic analysis of the IMP gene family in sorghum has been lacking. In this study, we identified eight IMP genes (SbIMP-1-SbIMP-8) in the sorghum genome. Phylogenetic analysis classified them into six subfamilies, consistent with rice and maize, suggesting a common ancestral origin. Gene str…

Genome editing in maize and sorghum: A comprehensive review of CRISPR/Cas9 and emerging technologies.

The increasing changes in the climate patterns across the globe have deeply affected food systems where unparalleled and unmatched challenges are created. This jeopardizes food security due to an ever-increasing population. The extreme efficiency of C4 crops as compared to C3 crops makes them incredibly significant in securing food safety. C4 crops, maize (Zea mays L.) and sorghum (Sorghum bicolor L. Moench) in particular, have the ability to withstand osmotic stress induced by oxidative stress. Osmotic stress causes a series of physical changes in a plant thus facilitating reduced water uptak…

Effects of PEG-induced osmotic stress on growth and dhurrin levels of forage sorghum.

Sorghum (Sorghum bicolor L. Moench) is a valuable forage crop in regions with low soil moisture. Sorghum may accumulate high concentrations of the cyanogenic glucoside dhurrin when drought stressed resulting in possible cyanide (HCN) intoxication of grazing animals. In addition, high concentrations of nitrate, also potentially toxic to ruminants, may accumulate during or shortly after periods of drought. Little is known about the degree and duration of drought-stress required to induce dhurrin accumulation, or how changes in dhurrin concentration are influenced by plant size or nitrate metabol…
