A Novel Sparse Cellular Labeling System with Tunable Gradients and Long-Term Stability.

The dense packing of cells within tissues poses challenges for studying cellular properties. Sparse labeling, genetically targeting a small subset of densely distributed cells, provides a powerful approach to investigate cellular morphology, connectivity, dynamics, and functions, especially in neuroscience. However, current sparse labeling methods are generally restricted to fixed labeling densities and often exhibit a decline in sparsity over time. In this study, by maximizing the utility of commonly used recombinases and the relational recognition sites, we constructed a versatile sparse lab

Visual Detection of Inorganic Orthophosphate in Phloem Cells.

The phloem plays a vital role in actively transporting soluble organic compounds-such as sugars, minerals, and proteins-from source to sink tissues, a process driven by turgor pressure and adenosine triphosphate (ATP)-dependent energy. Since ATP levels in phloem cells are closely linked to inorganic orthophosphate (Pi) availability, precise and efficient detection of Pi in these cells is essential for understanding phloem function. To address this, we developed the inorganic orthophosphate staining assay (IOSA), a method for visually detecting Pi in phloem cells. As a demonstration, we have su

Mapping subcellular microenvironments using oligonucleotide-directed proximity labeling.

Understanding how cells compartmentalize their biomolecules into discrete structures remains one of the fundamental goals of cell biology. The proliferation of proximity labeling (PL) technologies has been invaluable toward this goal, enabling biochemical "dissection" of compartments that would be intractable to classical biochemical methods. While robust PL approaches have long been established for targeting proteins of interest, targeting nucleic acids-RNAs and genomic loci-has remained significantly more challenging. Here, we review recent advancements in the field that overcome this longst

Elucidation of Ganglioside Microdomains Using Proximity Labeling.

It has long been known that lipid microdomain structures (also called "lipid rafts") are formed in the cell membrane, and gangliosides are concentrated in these domains. Many functional molecules (membrane proteins, receptors, and their associated signaling molecules, etc.) are concentrated in these domains along with gangliosides, and it has been pointed out that these domains play an important role in signal transduction, for instance, from outside the cell. Interestingly, it has been pointed out that each microdomain contains different types of gangliosides and functional molecules, and it

Current Landscape of Advanced Imaging Tools for Pathology Diagnostics.

Histopathology relies on century-old workflows of formalin fixation, paraffin embedding, sectioning, and staining tissue specimens on glass slides. Despite being robust, this conventional process is slow, labor-intensive, and limited to providing two-dimensional views. Emerging technologies promise to enhance and accelerate histopathology. Slide-free microscopy allows rapid imaging of fresh, unsectioned specimens, overcoming slide preparation delays. Methods such as fluorescence confocal microscopy, multiphoton microscopy, along with more recent innovations including microscopy with UV surface

Inherent lack of CMFDA/FDA staining in certain algae and its implication for ballast water testing.

The approved method for testing the efficacy of ballast water management systems with respect to killing 10-50 μm organisms uses movements of the organisms or the vital stains CMFDA/FDA. The present study demonstrates that certain freshwater coccoid chlorophytes, known or suspected to contain a highly resistant cell wall component (algaenan), stain poorly with CMFDA/FDA, resulting in false negatives. The staining rates for the most dominant species were determined and were approx. 3-70 %. The use of Crystal Violet as an indicator for the presence of algaenan gave inconclusive results. The numb

Application of the Method of Melanin Bleaching After Immunohistochemical Staining of Melanin-containing Tissues.

OBJECTIVE: The purpose of this research was to investigate the method of melanin bleaching in immunohistochemical staining of melanin-containing tissues. MATERIALS AND METHODS: Forty melanin-containing tissue samples were selected, including 30 with malignant melanoma and 10 with blue naevi. The samples were divided into the following 3 groups: the prestaining bleaching group, nonbleaching group, and poststaining bleaching group. Each group was subjected to HMB45 and MelanA immunohistochemical staining. The detection system used was Roche ventana ultraview universal alkaline phosphatase red k

Histomorphometric and histochemical characteristics of the oesophagus of the greater mouse-eared Bat, Myotis myotis (Borkhausen, 1797).

The aim of this study was to examine the histomorphometric and histochemical structure of oesophagus of the wild-caught greater mouse-eared bat M. myotis. For this purpose, 10 specimens of oesophagus were collected and processed. The oesophagus wall of M. myotis is composed of four layers: tunica mucosa, tunica submucosa, tunica muscularis and tunica adventitia. The mucosa is covered by a non-keratinized stratified squamous epithelium. There were no glands in the wall of the oesophagus. Tunica muscularis is composed of two muscle layers: internal circular muscular layer and an external longitu

A Histopathology-based Assessment of Biological Behavior in Oral Hyalinizing Extraosseous Lesions by Differential Stains.

AIM: The assessment of hyalinization to determine aggressive behavior in oral pathological lesions is a scarcely researched field that requires further exploration. The current study aims to predict the biological behavior of oral hyalinizing extraosseous lesions (OHEOL) by employing four differential stains with clinicopathologic correlation. MATERIALS AND METHODS: The study was performed on retrospectively diagnosed formalin-fixed paraffin-embedded cases of salivary gland tumors (SGTs) (n = 13), benign soft tissue (BST) lesions (n = 24), and oral submucous fibrosis (OSMF) (n = 53). The hema
