Mutation of E1 glycoprotein of classical swine fever virus affects viral virulence in swine.

Transposon linker insertion mutagenesis of a full-length infectious clone (IC) (pBIC) of the pathogenic classical swine fever virus (CSFV) strain Brescia was used to identify genetic determinants of CSFV virulence and host range. Here, we characterize a virus mutant, RB-C22v, possessing a 19-residue insertion at the carboxyl terminus of E1 glycoprotein. Although RB-C22v exhibited normal growth characteristics in primary porcine macrophage cell cultures, the major target cell of CSFV in vivo, it was markedly attenuated in swine. All RB-C22v-infected pigs survived infection remaining clinically …

H1N1 G4 swine influenza T cell epitope analysis in swine and human vaccines and circulating strains uncovers potential risk to swine and humans.

BACKGROUND: Pandemic influenza viruses may emerge from animal reservoirs and spread among humans in the absence of cross-reactive antibodies in the human population. Immune response to highly conserved T cell epitopes in vaccines may still reduce morbidity and limit the spread of the new virus even when cross-protective antibody responses are lacking. METHODS: We used an established epitope content prediction and comparison tool, Epitope Content Comparison (EpiCC), to assess the potential for emergent H1N1 G4 swine influenza A virus (G4) to impact swine and human populations. We identified an…

First study to describe the prevalence and epidemiology of African swine fever, classical swine fever, porcine reproductive and respiratory syndrome and swine flu in Kazakhstan.

BACKGROUND: Kazakhstan, the ninth-largest country in the world, located in Central Asia and bordering China, Kyrgyzstan, Russia, Turkmenistan, and Uzbekistan, hosts a diverse population of domestic pigs across various environments, providing potential hosts for highly pathogenic viral diseases of swine. Here we monitored African Swine Fever Virus (ASFV), Classical Swine Fever Virus (CSFV), Porcine Reproductive and Respiratory Syndrome Virus (PRRSV), and Swine Influenza Virus (SIV). RESULTS: During the spring and fall of 2019, we sampled 1,459 domestic pigs in northern, central and eastern Kaz…

Adenovirus-vectored novel African Swine Fever Virus antigens elicit robust immune responses in swine.

African Swine Fever Virus (ASFV) is a high-consequence transboundary animal pathogen that often causes hemorrhagic disease in swine with a case fatality rate close to 100%. Lack of treatment or vaccine for the disease makes it imperative that safe and efficacious vaccines are developed to safeguard the swine industry. In this study, we evaluated the immunogenicity of seven adenovirus-vectored novel ASFV antigens, namely A151R, B119L, B602L, EP402RΔPRR, B438L, K205R and A104R. Immunization of commercial swine with a cocktail of the recombinant adenoviruses formulated in adjuvant primed strong A…

Bovine viral diarrhea virus in swine: neutralizing antibody in naturally and experimentally infected swine.

Neutralizing antibodies against bovine viral diarrhea virus (BVDV) were detected in sera of Iowa farm sows, their piglets derived by hysterectomy and in specific-pathogen-free (SPF) pigs in a closed herd. No antibodies against BVDV were detected in sera of 134 fetal pigs from 67 sows in an Iowa slaughter-house. Neutralizing antibodies against BVDV or hog cholera virus, produced experimentally in SPF pigs, were demonstrated to be due to different antigens rather than to a common antigen, although slight cross-neutralization reactions sometimes occur.

[Activation of proteinases and acid phosphatase in swine leukocytes infected with the swine classical plague virus].

The activities of proteinases active at neutral pH and of acid phosphatase increases after freezing and defrosting and hypotonic shock in porcine leukocytes in vitro infected with the classical hog cholera virus in comparison with intact cells. The enzymes in the cells from animals both immune and nonimmune to hog cholera were activated upon infection with both virulent and vaccine strains of the virus. No proteinase activity was detected in the culture medium in which infected porcine leukocytes were cultured. Possible pathological reactions of a cell to viral infection are discussed.

Spontaneous and diet-induced coronary atherosclerosis in normal swine and swine with von Willebrand disease.

We have observed that pigs with impaired platelet function in the form of severe von Willebrand's disease (vWd) are resistant to spontaneous and to diet-induced aortic atherosclerosis. However, it has been reported that vWd pigs are susceptible to coronary atherosclerosis produced by balloon-induced injury of coronary arteries combined with an atherogenic diet. We have evaluated the development of coronary atherosclerosis in normal control (NC) and homozygous vWd pigs in two prospective studies: 1) as a spontaneous process in five NC and vWd pigs receiving a regular diet from the age of 3 mont

Bronchodilator responsiveness in swine veterinarians.

OBJECTIVE: Swine veterinarians are known to be at risk for respiratory symptoms and airflow obstruction. The present study reassessed the prevalence of respiratory complaints and pulmonary function abnormalities in swine veterinarians and sought to characterize their response to bronchodilators. METHODS: A cross-sectional study was conducted during the American Association of Swine Veterinarians annual meeting. Subjects completed a respiratory symptom and workplace exposure history questionnaire and spirometry. Subjects with airflow obstruction were assessed for a post-bronchodilator response…

General anesthetic techniques in swine.

General anesthesia techniques for swine can be challenging due to the animal's temperament, anatomic traits, physical condition, and the environment in which the clinician may be working. Taking these factors into consideration, this article provides specific information on preanesthetic considerations, venous catheterization, drug selection, monitoring, perioperative complications and therapy, recovery, and analgesia.

Halothane gene and swine performance.

One hundred thirty-one pigs representing seven different breed groups (Minnesota No. 1 [M], Pietrain [P], Yorkshire [Y], and crosses PY, P[PY], P[NP], and P[YP]P) and three halothane gene genotypes (NN, Nn, and nn) were tested for breed, sex, and halothane gene (HAL) effects on growth and carcass performance. Breed effects were significant for all traits measured. Sex effect was significant for most traits except for meat scores. The HAL-locus linkage group explained 20 to 30% of the total variation for meat quality scores and 1 to 10% for meat quantity and growth traits. Pietrain x Yorkshire

Riboflavin requirement of gestating swine.

Forty crossbred gilts (9 to 10 mo of age) were used to determine the riboflavin requirement for gestating swine. Upon mating, gilts were assigned to one of five levels of dietary riboflavin (.77, 1.77, 2.77, 3.77 or 4.77 ppm) and individually fed 2.0 kg daily of the assigned diet throughout gestation. Gilts were bled on d 7, 24, 30, 35, 40, 56, 70, 84, 90 and 97 postcoitum. On d 1 postpartum, all sows and their live piglets were bled. Hemolysates of isolated, washed erythrocytes were prepared from blood samples and assayed to establish the erythrocyte glutathione reductase activity coefficient…
