Transketolase regulates endoplasmic reticulum stress independent of enzymatic activity in human retinal Müller cells.

Transketolase (TKT) expression in the nucleus of the retina has been reported, but its function beyond metabolism remains unclear. In this study, we investigated the role of TKT in the regulation of endoplasmic reticulum (ER) stress in the retina. Using a VEGF-overexpressing mouse model of age-related macular degeneration (AMD), we analyzed the expression and enzymatic activity of Tkt, and found that while expression levels remained unchanged, enzymatic activity was significantly reduced. Tkt localized primarily to the nucleus of the inner nuclear layer in healthy retinae but shifted outside t…

Transketolase abnormality in cultured fibroblasts from familial chronic alcoholic men and their male offspring.

We have investigated a thiamine-dependent enzyme, transketolase, in cultured fibroblasts from 41 human subjects, including patients with alcoholism-associated Wernicke-Korsakoff syndrome (n = 3), familial chronic alcoholic males (n = 7), their sons (n = 7), nonalcoholic men (n = 7), their male offspring (n = 7), and three generations of an Amish family (n = 10) without any history of alcoholism. This study was undertaken to delineate whether transketolase abnormality (i.e., high Michaelis Menton constant (Km) for thiamine pyrophosphate), previously reported in patients with Wernicke-Korsakoff …

Investigating Coenzyme Function of Thiamine Triphosphate Using Its Novel Hydrolysis-Resistant Analog and Transketolase.

Thiamine (vitamin B1) and its phosphates are essential for almost all organisms. Thiamine diphosphate (ThDP) is the major intracellular deri … ed. Given that ThTP accumulation is caused by cytosolic THTPA suppression, cytosolic ThDP-dependent transketolase (TKT) is the primary target for probing (bm)ThTP's coenzyme function. Indeed, bmThTP acts as a TKT coenzyme, with the apparent Km(bmThTP) of 16.3 µM. However, bmThTP binding slightly differs from that of ThTP. Molecular docking was used to estimate …

Design, Synthesis, and Herbicidal Activity Study of Novel Pyrazole-Carboxamides as Potential Transketolase Inhibitors.

Transketolase (TKL; EC 2.2.1.1) has been identified as a potential new herbicide target. In order to discover highly herbicidal active compounds targeting TKL and improve their structural diversity for lead compounds, a series of pyrazole-carboxamides 7a-7v were designed and synthesized through structural optimization for pyrazole-containing phenoxy amide compound 4u. Among the synthesized compounds, compound 7r possessed excellent herbicidal efficacy against Digitaria sanguinalis (Ds) and Amaranthus retroflexus (Ar) by the small cup method (the inhibition about 95%, 100 mg/L) and the foliar s…

Salmonella enterica serovar Typhimurium has three transketolase enzymes contributing to the pentose phosphate pathway.

The genus Salmonella is responsible for many illnesses in humans and other vertebrate animals. We report here that Salmonella enterica serovar Typhimurium harbors three transketolases that support the non-oxidative branch of the pentose phosphate pathway. BLAST analysis identified two genes, STM14_2885 and STM14_2886, that together encode a putative transketolase (TktC) with 46-47% similarity to the known TktA and TktB isoforms. Assessing the mRNA and protein expression for each of the three transketolases, we determined that all are expressed in WT cells and regulated to varying extents by th…

In thiamine deficiency, activation of erythrocyte transketolase by thiamine in vivo exceeds activation by cofactor in vitro.

In 60 thiamine deficient patients, the mean erythrocyte transketolase activity after activation by thiamine diphosphate cofactor in vitro, representing the apparent sum of holoenzyme and apoenzyme activities, was 0.609 (SD 0.166) U/g Hb before thiamine therapy and rose to 0.772 (SD 0.152) U/g Hb immediately after the administration of thiamine to the patients. The difference between these values, 0.163 (SD 0.130) U/g, is the mean activity of transketolase protein which can be activated by thiamine in vivo but not by thiamine diphosphate in vitro. This difference correlated with low initial ery…

The effect of reduced dietary protein level on the activity of transketolase and glutathione reductase in pig erythrocytes.

The effect was investigated of decreased protein level in a food ration on covering the requirements for vitamins B1 and B2. Two feeding experiments were conducted, each on 32 fattening pigs of Polish Landrace breed. The activity of transketolase and glutathione reductase in erythrocytes and vitamin B1 and B2 level in blood and liver were determined. It was shown that a reduction of the diatery protein level by 25% caused a decrease of thiamine utilization by pigs. It indicates the necessity of a dietary supplementation with a synthetic vitamin B1 preparation in such a condition. A reduced act…

The determination of thiamin pyrophosphate in blood and other tissues, and its correlation with erythrocyte transketolase activity.

A sensitive method for the specific measurement of thiamin pyrophosphate (TPP) has been developed using the apoenzyme recombination concept. … are fed a thiamin deficient diet, the erythrocyte TPP level falls more rapidly than the erythrocyte transketolase activity. After 8 days, the level of TPP in the erythrocytes of deficient animals was 10% of the level in pair-fed controls. At this time, however, there was no appreciable decrease in their respective transketolase activities. The level of TPP in t …

Overexpression of transketolase-like gene 1 is associated with cell proliferation in uterine cervix cancer.

BACKGROUND: Tumor cells need large energy and nucleic acids to proliferate and grow. For most of their energy needs, cancer cells depend more on glycolysis. For most of their nucleic acids needs, cancer cells depend more on the nonoxidative pathway of the pentose phosphate pathway. Transketolase(TKT) is a crucial enzyme in the nonoxidative pathway of the PPP. METHODS: The real-time quantity PCR was used to determine the expression of transketolase gene family in uterine cervix cancer. Transketolase activity of cell was determined by using enzyme-linked method. Cell proliferation was detected …

Expression of transketolase-like 1 (TKTL1) and p-Akt correlates with the progression of cervical neoplasia.

AIM: It is supposed that increased glycolysis is crucial for the energy supply during tumor progression. Unfortunately, the relevance of glycolysis in cervical neoplasia is unknown, but what is certain is the fact that cervical cancer shows a high expression of glucose membrane transporters, which are necessary for glucose uptake as an energy source. Transketolase-like enzyme 1 (TKTL1) and the oncogene p-Akt have been described to play an important role in glycolysis during tumorigenesis. Thus, we were interested in their expression in cervical tissue. METHODS: We examined the expression of T…

Production and purification of Penicillium citreoviride toxin and its effect on TPP-dependent liver transketolase.

The production isolation and purification of a yellow mycotoxin from Penicillium citreoviride NRRL 2579 in different culture media was described. When injected subcutaneously to albino rats it alters the kinetic pattern of transketolase (EC 2.2.1.1) in liver in vivo in a competitive manner. In vitro, the inhibition is noncompetitive in nature. However, addition of thiamine diphosphate (TPP) to the in vitro system relieved the inhibitory effect. These findings suggested a relationship between citreoviridin-induced beriberi and the probable antithiamine effect of the toxin.

Activities of thiamine-dependent enzymes in two experimental models of thiamine deficiency encephalopathy: 3. Transketolase.

Chronic thiamine deprivation in the rat leads to ataxia, loss of righting reflex and neuropathological damage to lateral vestibular nucleus. Before onset of neurological symptoms, transketolase (TK) activities were found to be selectively reduced by 25% in lateral vestibular nucleus and surrounding pons. Further progression of thiamine deprivation resulted in a generalized reduction in TK activity. Measurement of enzyme activity in the presence of added TPP cofactor in vitro did not lead to normalisation of enzyme activities suggesting loss of apoenzyme. Administration of thiamine to symptomat…
