Diagnostic accuracy of the light microscope method to detect the eggs of Cardicola spp. in the gill filaments of the bluefin tuna.

Trematode blood flukes of the genus Cardicola are potentially lethal in bluefin tuna cultures. The present study proposed a new method to detect aporocotylid eggs in tuna gills. Aporocotylid eggs were detected by analysing a pair of gill filaments of five transversal areas of the eight holobranches of one hundred Atlantic bluefin tuna and observed with glycerol and a stereomicroscope with an oblique brightfield. Data were gathered according to holobranches, transversal areas and their combination. Eggs were uniformly distributed among the holobranches, but they had the highest prevalence in th…

Levels of mercury and organohalogen compounds in Pacific bluefin tuna (Thunnus orientalis) cultured in different regions of Japan.

Contamination levels of total mercury (T-Hg), p,p'-DDE, and polychlorinated biphenyls (PCBs) in akami (leaner meat) and toro (fatty meat) samples from Pacific bluefin tuna cultured in the southern (four locations) and central (three locations) regions of Japan were analyzed. The contamination level of T-Hg in the akami and toro samples from the southern region tended to decrease with an increase in latitude, whereas those of p,p'-DDE and PCBs tended to increase. These spatial trends in contaminants were similar to those reported previously in wild tuna caught off the coast of Japan (Hisamichi …

Thermal pretreatment-assisted dual-enzyme hydrolysis of steamed skipjack tuna dark muscle for safety-controlled bioactive hydrolysate production.

Steamed skipjack tuna dark muscle (STDM), a protein-rich by-product of tuna canning, is underutilized because of microbial contamination and histamine-related safety concerns. This study developed a safety-controlled hydrolysate through heat pretreatment and sequential Alcalase (AL)-Flavourzyme (FL) hydrolysis. An AL:FL ratio of 4:1 (w/w), 20 mg total enzyme, and sequential addition yielded 56.38% hydrolysis after 5 h. Heat treatment reduced total aerobic bacteria, coliforms, and Escherichia coli below detection limits and was associated with lower relative histamine-forming bacteria-associate…

Staphylococcus aureus Burden in Frozen, Precooked Tuna Loins and Growth Behavior During Typical and "Worst-case" Processing Conditions.

FDA's Seafood HACCP Guidance (SHG) recommends a cumulative exposure time limit of 3 h if product temperature exceeds 21.1 °C (70°F) to minimize the growth of Staphylococcus aureus (SA) and prevent staphylococcal enterotoxin (SE) production. This time limit is insufficient to practically accomplish postthawing activities (e.g., packing, brining, sealing, retort staging, and time for the center of the can to reach the critical temperature of 50 °C (122°F)). The documented evidence supporting the establishment of this exposure time recommendation is minimal and mostly irrelevant to tuna. The goal…

The Southern Bluefin Tuna Mucosal Microbiome Is Influenced by Husbandry Method, Net Pen Location, and Anti-parasite Treatment.

Aquaculture is the fastest growing primary industry worldwide. Marine finfish culture in open ocean net pens, or pontoons, is one of the largest growth areas and is currently the only way to rear high value fish such as bluefin tuna. Ranching involves catching wild juveniles, stocking in floating net pens and fattening for 4 to 8 months. Tuna experience several parasite-induced disease challenges in culture that can be mitigated by application of praziquantel (PZQ) as a therapeutic. In this study, we characterized the microbiome of ranched southern Bluefin Tuna, Thunnus maccoyii, across four a…

Tuna Oil Attenuates Crab Tropomyosin-Induced Food Allergy in Mice via Modulation of Intestinal Immunity, Microbiota, and Proteome.

Food allergies, particularly to crustaceans, are a growing health concern with limited management options. We heterologously expressed and purified crab tropomyosin and then evaluated the beneficial effects of tuna oil on crab TM-induced food allergy in mice. The results demonstrated that tuna oil dose-dependently alleviated allergic symptoms and reduced serum levels of TM-specific IgE, histamine, and cytokines. It also improved intestinal dendritic cell imbalance by reducing CD11c+CD80+/CD86+ cells and restoring CD103+ subsets. In addition, tuna oil enhanced the richness and diversity of gut …

Preliminary Kinetic Assessment of Quality Deterioration and Shelf-Life Estimation in Yellowfin Tuna Under Different Constant Storage Temperatures.

The quality of yellowfin tuna is highly vulnerable to microbial proliferation, which accelerates quality deterioration and spoilage throughout storage. In this study, the dynamic changes in biogenic amines (BAs), total volatile basic nitrogen (TVB-N), and total plate count (TPC) across a different storage temperature range of 0 to 25 °C were studied, followed by a shelf-life prediction based on kinetic models. The TPC, TVB-N, and most BAs increased significantly with increasing storage time and temperature. Correlation analysis was used as an exploratory approach and showed that only histamine…

Tracing mercury sources and transfer pathways in Pacific and Atlantic Ocean tuna and billfish using mercury stable isotopes.

Tuna and billfish indicates Hg and MeHg accumulation through marine food webs, yet the causes of interspecies and interoceanic variation remain unclear. This study analyzed THg, MeHg, and Hg isotopes in seven tuna and billfish species from the Pacific and Atlantic, revealing bioaccumulation pattern in top marine predators. Blue marlin (a kind of billfish) showed significantly higher THg concentrations (1824 ± 1558 ng g-1 ww) and lower MeHg/THg ratios (28.6 ± 17.3 %) compared to other species. BAFs estimated via Monte Carlo simulations revealed species-specific differences, with MeHg values 16-…

Ocean conditions drive interannual variability in juvenile albacore tuna (Thunnus alalunga) muscle energy content in the California Current System.

Juvenile albacore tuna (Thunnus alalunga) undertake long migrations across the North Pacific that they fuel by feeding in the California Current System (CCS) during the summer. The CCS is a highly dynamic system, which can lead to differences in foraging success that may be indicated by differences in body condition. Assessed through stomach content analysis, albacore diet composition by mean proportional weight showed high interannual variability including some years characterized by large increases in consumption of anchovy (2011 = 81%, 2017, 2022) and sardine (Sardinops sagax, 2022). We use…
