Viral protein mutations enabling mammalian adaptation in Avian Influenza A viruses: Strategies for zoonotic risk mitigation and future perspectives.

Avian influenza viruses have long been a significant problem for the poultry livestock sector, due to the pathogenicity of some strains and the economic and environmental damage they cause. The close relationship between poultry species of livestock interest and the detection of these viruses in other livestock and pets has prompted the scientific community to investigate the transmission dynamics and the risk of contagion for humans. Research shows how the possible spillover can be driven by some amino acid mutations that regulate the molecular interactions between viruses and different hosts

Fiocruz in Antarctica - health and environmental surveillance facing the challenges of the 21st century.

FioAntar, FIOCRUZ's research project in Antarctica, is based on the One Health approach. FioAntar aims to generate relevant information that will help reduce the risk of future pandemics and improve the search for chemical compounds and new biological molecules. After four expeditions to Antarctica under the scope of PROANTAR, Fiocruz has identified Influenza H11N2 virus in environmental fecal samples, as well as Histoplasma capsulatum and Bacillus cereus in soil samples. In addition, in a prospective virome analysis from different lakes in the South Shetland Islands, six viral orders were des

The place of marker vaccines in control and eradication of animal diseases--aspects of comparative interest.

Smallpox is the first viral infection to have been eradicated world-wide. This remarkable success is due to several factors including the availability of an efficacious vaccine and the absence of a wildlife reservoir. The only animal virus disease sharing these characteristics is rinderpest, for which there are several efficacious vaccines. Other animal viral infections do not share the same characteristics, either due to the lack of an efficacious vaccine (African swine fever), or to the existence of wildlife reservoirs such as the wild boar for classical swine fever, the African buffalo for

Arthropod-borne virus zoonosis surveillance in the Cape Province: 1. Prospective serological investigations for virus activity in the Beaufort West and Middelburg Districts during 1981.

In addition to the routine sero-epidemiological surveillance for arthropod-borne viral zoonoses in the Cape Province carried out by the Department of Medical Microbiology and State Health Department of Virology laboratory, we conducted a prospective serological investigation for virus activity during 1981 in two districts of the Province, namely the Beaufort West and Middelburg districts, which experienced heavy rainfall during the first two months of that year. The approach used was to obtain paired serum samples from identified domestic stock representative of several species from 2-5 months…

A Unified Framework for the Infection Dynamics of Zoonotic Spillover and Spread.

A considerable amount of disease is transmitted from animals to humans and many of these zoonoses are neglected tropical diseases. As outbreaks of SARS, avian influenza and Ebola have demonstrated, however, zoonotic diseases are serious threats to global public health and are not just problems confined to remote regions. There are two fundamental, and poorly studied, stages of zoonotic disease emergence: 'spillover', i.e. transmission of pathogens from animals to humans, and 'stuttering transmission', i.e. when limited human-to-human infections occur, leading to self-limiting chains of transmi

Zoonoses and poverty - a long road to the alleviation of suffering.

Populations living in poverty in the developing world suffer a heavy burden caused by infectious diseases, most of them zoonoses. The poorer populations also tend to be marginalised from the health sector and so are many of the diseases that affect them. The poor in every society, and particularly in developing countries, bear a disproportionately high share of the disease burden. There is a broad range of viral, bacterial, mycotic, chlamydial, rickettsial and parasitic diseases of global and regional importance given their major impact on the health and socio-economic development of many popu

[Viral contamination of food products: a poorly understood public health problem].

Throughout the world there have been several epidemics of food-borne diseases (FBD) about which there is lack of sufficient information for … and small gastroenteric viruses are also important causes of FBD. We also discuss the importance of viral zoonoses, especially hemorrhagic fevers transmitted by contact with rodent feces and tick-borne viral encephalitides (Lassa fever). There is discussion of the controversial mad cow disease and its potential transmission through food products, as well as of d …

Mosquito-independent milk-associated transmission of zoonotic Wesselsbron virus in sheep.

Wesselsbron virus (WSLV) is a zoonotic, mosquito-borne orthoflavivirus endemic to sub-Saharan Africa, causing abortions and stillbirths in small ruminants. The life cycle of WSLV involves Aedes mosquitoes and various wildlife and domestic animals. Seminal studies in the 1950s have shown the zoonotic potential of WSLV, notably in accidental infections of laboratory workers exposed to infected material. More recent epidemiological studies suggest the emergence of clade I WSLV strains in peri-domestic and rural areas of western and eastern Africa. The pathobiology of recent clade I WSLV strains i

Henipaviruses at the threshold: preparedness in the era of recurrent spillover.

Henipaviruses remain high-consequence zoonotic pathogens characterized by recurrent spillover, severe disease, and limited but consequential human-to-human transmission. In this commentary, we frame henipaviruses as "threshold pathogens": agents repeatedly approaching, but not yet consistently crossing, the boundary into sustained community transmission. Expanding viral diversity, widening ecological interfaces, and episodic outbreaks challenge conventional preparedness models, particularly for surveillance, countermeasure evaluation, and regulatory decision-making. We argue that future prepar

The Emergence and Evolution of SARS-CoV-2.

The origin of SARS-CoV-2 has evoked heated debate and strong accusations, yet seemingly little resolution. I review the scientific evidence on the origin of SARS-CoV-2 and its subsequent spread through the human population. The available data clearly point to a natural zoonotic emergence within, or closely linked to, the Huanan Seafood Wholesale Market in Wuhan. There is no direct evidence linking the emergence of SARS-CoV-2 to laboratory work conducted at the Wuhan Institute of Virology. The subsequent global spread of SARS-CoV-2 was characterized by a gradual adaptation to humans, with dual

Conserved HA-peptides expressed along with flagellin in Trichoplusia ni larvae protects chicken against intranasal H7N1 HPAIV challenge.

The immunization of poultry where H5 and H7 influenza viruses (IVs) are endemic is one of the strategies to prevent unexpected zoonoses. Our group has been focused on conserved HA-epitopes as potential vaccine candidates to obtain multivalent immune responses against distinct IV subtypes. In this study, two conserved epitopes (NG-34 and CS-17) fused to flagellin were produced in a Baculovirus platform based on Trichoplusia ni larvae as living biofactories. Soluble extracts obtained from larvae expressing "flagellin-NG34/CS17 antigen" were used to immunize chickens and the efficacy of the vacci

Poxviruses in bats … so what?

Poxviruses are important pathogens of man and numerous domestic and wild animal species. Cross species (including zoonotic) poxvirus infections can have drastic consequences for the recipient host. Bats are a diverse order of mammals known to carry lethal viral zoonoses such as Rabies, Hendra, Nipah, and SARS. Consequent targeted research is revealing bats to be infected with a rich diversity of novel viruses. Poxviruses were recently identified in bats and the settings in which they were found were dramatically different. Here, we review the natural history of poxviruses in bats and highlight…

Lassa Virus Genetics.

In a pattern repeated across a range of ecological niches, arenaviruses have evolved a compact four-gene genome to orchestrate a complex life cycle in a narrow range of susceptible hosts. A number of mammalian arenaviruses cross-infect humans, often causing a life-threatening viral hemorrhagic fever. Among this group of geographically bound zoonoses, Lassa virus has evolved a unique niche that leads to significant and sustained human morbidity and mortality. As a biosafety level 4 pathogen, direct study of the pathogenesis of Lassa virus is limited by the sparse availability, high operating co

Next-generation diagnostics: virus capture facilitates a sensitive viral diagnosis for epizootic and zoonotic pathogens including SARS-CoV-2.

BACKGROUND: The detection of pathogens in clinical and environmental samples using high-throughput sequencing (HTS) is often hampered by large amounts of background information, which is especially true for viruses with small genomes. Enormous sequencing depth can be necessary to compile sufficient information for identification of a certain pathogen. Generic HTS combining with in-solution capture enrichment can markedly increase the sensitivity for virus detection in complex diagnostic samples. METHODS: A virus panel based on the principle of biotinylated RNA baits was developed for specific

Lassa fever or lassa hemorrhagic fever risk to humans from rodent-borne zoonoses.

Viral hemorrhagic fevers (VHFs) typically manifest as rapidly progressing acute febrile syndromes with profound hemorrhagic manifestations and very high fatality rates. Lassa fever, an acute hemorrhagic fever characterized by fever, muscle aches, sore throat, nausea, vomiting, diarrhea and chest and abdominal pain. Rodents are important reservoirs of rodent-borne zoonosis worldwide. Transmission rodents to humans occur by aerosol spread, either from the genus Mastomys rodents' excreta (multimammate rat) or through the close contact with infected patients (nosocomial infection). Other rodents o

Marine mammal zoonoses: a review of disease manifestations.

Marine mammals evoke strong public affection as well as considerable scientific interest. However, the resultant close contact with marine wildlife poses human health risks, including traumatic injury and zoonotic disease transmission. The majority of zoonotic marine mammal diseases result in localized skin infections in man that resolve spontaneously or with appropriate medical therapy. However, other marine mammal zoonoses, if left untreated, induce life-threatening systemic diseases that could pose public health risks. As the number of zoonotic diseases rises, the diagnosis of and treatment

One Health Genomic Surveillance at Human-Animal Interfaces in Rural Ghana Reveals Underreported Viruses of Zoonotic and Economic Concern.

Under a One Health framework, viruses of veterinary and zoonotic importance pose significant threats to animal and human health, food security, and livelihoods, particularly in regions with intense human-animal interactions. In West Africa, despite recent advances in surveillance programs, important gaps remain in understanding viral diversity and cross-species transmission at wildlife-livestock interfaces. We conducted metagenomic surveillance to characterize viruses circulating across livestock, domestic animals, and wildlife in rural Ghana in 165 animals sampled across five regions. Viral R

Encephalomyocarditis virus (EMCV): An overlooked threat to primates.

Many emerging zoonotic viruses pose major risks to animal and human health, with most recent epidemics of viral origin. This review focuses on encephalomyocarditis virus (EMCV), a member of the Picornaviridae family and the Cardiovirus genus with significant but largely underrecognized zoonotic potential. While EMCV has not yet caused major public health crises, its broad host range and rodent reservoir suggest a wider ecological impact. It is typically detected only during high-fatality outbreaks, particularly in domestic animals and non-human primates (NHPs), which are among the most vulnera

Distribution and richness of Orthohantavirus reservoir hosts: Implications for zoonotic risk in southern South America.

Orthohantaviruses represent a major zoonotic threat in Southern South America (SSA), where 13 native rodent species are recognized reservoir hosts of 15 different viral genotypes. The information on their geographic distribution remains scattered, limiting regional assessments of exposure risk. We compiled and curated occurrence records for the Orthohantavirus reservoir hosts in SSA and used ecological niche modeling to estimate their potential distributions. We integrated binary presence predictions to map reservoir and genotype richness as well as reservoir assemblages. The richness maps wer
