Genital yeast infections.

Genital yeast infection was studied in 533 women seen in a department of venereology. Yeasts were recovered in culture from 138 patients (26% of the total). Candida albicans accounted for 112 (81%) of the isolates and Torulopsis glabrata for 22 (16%); other yeasts were uncommon. There was no evidence that the presence of yeasts was related to age. 32% of the women who were taking an oral contraceptive harboured yeasts, compared with 18% of those who were not.The symptoms and signs of the women with yeast infections were compared with those with vaginal trichomoniasis and those with no evidence…

Transcriptomic responses of kveik yeasts to heat and ethanol stress.

Kveik strains are a group of Saccharomyces cerevisiae strains notable for their exceptional tolerance to environmental stress. To investigate the molecular basis of this robustness, the transcriptomes of three kveik strains and a conventional ale strain were compared under non-stress and stress conditions using RNA-seq. Under non-stress conditions, kveik strains showed elevated basal expression of environmental stress response genes, suggesting that part of the stress-responsive transcriptional program is active irrespective of stress exposure. Catalase activity was elevated, providing physiol

Ergosterol from Yeasts Promotes Helicobacter pylori Internalization into Candida albicans.

Helicobacter pylori is a cholesterol-auxotrophic pathogen capable of internalizing into Candida albicans, where the yeast may serve as a protective intracellular niche under adverse environmental conditions. Because ergosterol is an analog of cholesterol, this study investigated its contribution to the interaction between H. pylori J99 and C. albicans ATCC 90028. Ergosterol was extracted from the wild-type strain ATCC 90028 and from Δerg4, Δerg5, and Δerg6 mutants, identified and quantified by UHPLC-MS/MS, and evaluated for its effects on bacterial viability, growth, chemotaxis, and intracellu

[Yeasts in domestic animals: species identification and susceptibility to antifungals].

Yeasts frequently colonize various kinds of domestic animals, but may also cause serious diseases. The aim of this study was to identify yeast isolates collected from dogs, cows and pigs, and to determine their in vitro antifungal susceptibility. Fifty-six yeast isolates from dogs (n = 24), cows (n = 20), and pigs (n = 12) were investigated. Appearance of colonies grown on Sabouraud agar, micromorphology on rice agar, as well as assimilation and fermentation of various carbon and nitrogen sources were evaluated. Susceptibility to six antifungals (flucytosine, amphotericin B, miconazole, ketoco…

A synthetic peptide selectively kills only virulent Paracoccidioides brasiliensis yeasts.

This work was conducted to identify virulence biomarkers for Paracoccidioides brasiliensis (Pb), the fungus responsible for Paracoccidioidomycosis (PCM), a systemic disease endemic in Latin America. Measurement of mortality showed that all B10.A mice were killed after 250 days by the virulent Pb18 isolate while only one of the mice that received the attenuated counterpart died. Also, number of lung CFUs from virulent Pb18 inoculated mice were much higher when these isolates were compared. Phage display methodology allowed selection of three phages that specifically bound to virulent Pb18. Vari

[Antifungal susceptibility of yeasts by Etest. Comparison of 3 media].

BACKGROUND: The increasing frequency of systemic fungal infections and the emergency of secondary resistance to antifungals in the lasts years, has stimulated the use of methods for antifungal susceptibility testing. Etest is an easily performed quantitative method that has a good agreement with the broth microdilution reference method (NCCLS), if appropriate media are used. AIM: To compare the susceptibility to Amphotericin B (AmB) and Fluconazole (Flu) of 22 opportunistic yeast isolates (C albicans (7), C tropicalis (9), C parapsilosis (3) and Cryptococcus neoformans (3) by Etest, using thr

Cyanide-resistant respiration, a very frequent metabolic pathway in yeasts.

It has recently been shown that cyanide-resistant respiration (CRR) is very common in Crabtree-negative yeasts (incapable of aerobic fermentation) and in non-fermentative yeasts. It is conferred by a salicylhydroxamic acid-sensitive alternative oxidase that transfers electrons from ubiquinol to oxygen, bypassing the cytochrome chain. An interesting finding is that, in general, whenever CRR is present, complex I is also present. In this article we briefly review the occurrence of CRR, the biochemistry and molecular biology of the alternative oxidase, and summarise the putative functions that ha…

Yeast for virus research.

Budding yeast (Saccharomyces cerevisiae) and fission yeast (Schizosaccharomyces pombe) are two popular model organisms for virus research. They are natural hosts for viruses as they carry their own indigenous viruses. Both yeasts have been used for studies of plant, animal and human viruses. Many positive sense (+) RNA viruses and some DNA viruses replicate with various levels in yeasts, thus allowing study of those viral activities during viral life cycle. Yeasts are single cell eukaryotic organisms. Hence, many of the fundamental cellular functions such as cell cycle regulation or programed …

Identification of Yeast and Yeast-Like Fungi at a Tertiary Care Center: A Retrospective Study.

Background Fungal infections pose a significant global health challenge. Despite their substantial impact, these ubiquitous fungi can become pathogenic but have not received adequate attention in public health, leading to infections that are often underestimated by the general public and healthcare professionals. Candida species and Cryptococcus species play a key role in these infections, with emerging multidrug resistance in Candida species posing considerable challenges. This study aimed to understand the prevalence of yeast and yeast-like infections, particularly in the COVID-19 era, and t

Hybrid yeasts as dynamic platforms for robust and scalable industrial bioprocessing.

Hybrid yeasts represent a promising strategy for developing robust microbial platforms capable of sustaining industrial bioprocesses under multifactorial stress conditions, including high ethanol concentrations, osmotic pressure, temperature fluctuations, and inhibitory compounds. This review examines yeast hybridization as an integrative approach that combines complementary genetic and physiological traits from distinct parental lineages to expand metabolic capacity and improve process performance. Evidence indicates that hybridization, particularly when coupled with adaptive evolution, can e…

Harnessing yeasts for sustainable methanol valorization: Metabolic and fermentation engineering strategies.

Methanol has emerged as a sustainable C1 feedstock owing to its compatibility with existing infrastructure and the potential for renewable production from CO₂ and green hydrogen. Methylotrophic yeasts, including Komagataella phaffii (also known as Pichia pastoris) and Ogataea polymorpha, can natively assimilate methanol and therefore represent attractive hosts for biomanufacturing. However, industrial application of methanol-based processes remains constrained by cytotoxicity, redox imbalance, and limited productivity compared to sugar-based fermentations. To address these challenges, extensiv…

Cross-reactivity of cryptococcal antigen lateral flow assay with basidiomycetous yeasts.

The cryptococcal antigen lateral flow assay (CrAg LFA) is a widely available point-of-care test for the diagnosis of cryptococcal meningitis. Although there have been some reports of cross-reactivity with Trichosporon species, the extent of cross-reactivity among basidiomycetous yeasts has not been widely explored. We tested a range of basidiomycete yeast isolates using a CrAg LFA to examine potential cross-reactivity, with positive results for all isolates belonging to Trichosporon and Naganishia species, and for some isolates of Cutaneotrichosporon, Filobasidium and Ustilago species. While t…

Antifungal resistance in yeasts from One Health perspective: A Brazilian study.

Environmental exposure to fungicides can induce cross-resistance in filamentous fungi, but little is known about their effects on medically important yeasts. This multicenter study investigated the resistance in Candida isolated from different sources and evaluated the influence of clinical and environmental antifungals. Different yeasts were collected from patients, hospital environments, swine, poultry and their surroundings in four different states of Brazil. A total of 571 isolates were identified, of which 82.5 % belonged to the genus Candida. Antifungal susceptibility testing revealed th…

The effects of tyrosol on yeasts: an overview of current knowledge.

Quorum sensing is a cell density-dependent microbial communication form, which can regulate several microbial properties, including virulence, biofilm formation and cell-cell competence. The phenomenon of fungal quorum sensing was first uncovered nearly 25 years ago, following the identification of farnesol and tyrosol as two key signalling molecules. Although the major roles of these regulatory molecules were elucidated, several questions primarily regarding tyrosol-mediated effects remain to be addressed, particularly with regard to molecular events influenced by tyrosol. Based on available
