Mutation of Foxo3 causes adult onset auditory neuropathy and alters cochlear synapse architecture in mice.

Auditory neuropathy is a form of hearing loss in which cochlear inner hair cells fail to correctly encode or transmit acoustic information t … and Myo7a immunoreactivity is reduced. TEM demonstrates apparent afferent degeneration. Strikingly, acoustic stimulation promotes Foxo3 nuclear localization in vivo, implying a connection between cochlear activity and synaptic function maintenance. Together, these findings support a new role for the canonical damage response factor Foxo3 in contributing to the mainten …

Effect of partial neuromuscular blockade on intraoperative electromyography in patients undergoing resection of acoustic neuromas.

Intraoperative electromyographic monitoring of the facial nerve during acoustic neuroma excision provides early detection of nerve injury and improved outcome. To determine whether a useful level of peripheral neuromuscular blockade could be achieved without compromise of facial electromyographic monitoring, we studied 10 patients undergoing resection of acoustic neuroma. Facial nerve monitoring was accomplished by placement of wire electrodes in the orbicularis oris, orbicularis occuli, and mentalis muscles. Peripheral neuromuscular blockade was assessed by recording unprocessed hypothenar co

Effects of stimulus frequency and intensity on c-fos mRNA expression in the adult rat auditory brainstem.

Induction of the cellular fos gene (c-fos) is one of the earliest transcriptional changes observed following neuronal excitation. Although not an activity marker in the strict electrophysiological sense, many neurons in the central nervous system increase their c-fos expression after periods of sustained stimulation at physiological levels of intensity. In the present study, induction of c-fos mRNA expression was examined in the auditory brainstem after 1 hour of continuous free-field acoustic stimulation. Sprague-Dawley rats were exposed to pure tones of 2, 8, 16, or 32 kHz or half-octave noi…

Vocal tremor reduction with deep brain stimulation.

We present a case illustration of the significant effect that deep brain stimulation (DBS) of the thalamus can have on vocal tremor. A 72-year-old female with a history of essential tremor was noted preoperatively to have a moderate vocal tremor (3 on a scale of 1-5). Following bilateral DBS of the thalamus, the vocal tremor rating improved to 1. Acoustic analysis demonstrated her vocal tremor to be affecting the amplitude of her voice at 5.58 Hz preoperatively, at 1. 93 Hz postoperatively with both leads on and at 1.54 Hz with only the left lead on. A videotaped endoscopic view of the patient

[Spectrum analysis of vestibular nystagmus].

A procedure for the spectral analysis of nystagmograms obtained by rotary stimulation with the rate of 180 degrees/s for 20 s has been developed with the aim to reveal the most typical changes in the vestibular nystagmus with one or another form of the cochleovestibular pathology. A signal recorded in the analogous form was processed by computer EC-1022. Periodograms were calculated by the method of the fast Fourier transform and periodogram smoothing by averaging of realization sites. In case of Ménière's disease the third harmonic was not recorded on periodograms when right and left labyrint

The effects of transcutaneous auricular vagus nerve stimulation (taVNS) on cholinergic neural networks in humans: A neurophysiological study.

OBJECTIVE: The mechanisms of actions of transcutaneous auricular vagus nerve stimulation (taVNS) are still unclear, however the activity of the cholinergic system seems to be critical for the induction of VNS-mediated plasticity. Transcranial Magnetic Stimulation (TMS) is a well-suited, non-invasive tool to investigate cortical microcircuits involving different neurotransmitters. Herein, we evaluated the effect of taVNS on short-latency afferent inhibition (SAI), a TMS paradigm specifically measuring cholinergic neurotransmission. METHODS: Fifteen healthy subjects participated in this randomi

Deafness Weakens Interareal Couplings in the Auditory Cortex.

The function of the cerebral cortex essentially depends on the ability to form functional assemblies across different cortical areas serving … y cortex and the secondary posterior auditory field (PAF). Ongoing activity as well as responses to acoustic stimulation (in adult hearing controls) and electric stimulation applied via cochlear implants (in adult hearing controls and CDCs) were analyzed. As functional connectivity measures pairwise phase consistency and Granger causality were used. While the number o …

Effect of Real-Ear Adjusted Stimuli on Vestibular Evoked Myogenic Potential Variability in Children and Young Adults.

OBJECTIVES: There is large variability in cervical and ocular vestibular evoked myogenic potential (c- and oVEMP) amplitudes. One potential source of variability is differences in ear canal shape and size. Real ear-to-coupler difference (RECD) values are used to measure the acoustic environment of an individual's ear canal. RECD may be a useful measure to calibrate air conducted VEMP stimuli, which are elicited at high intensities and may put patients at risk of unsafe sound exposure. A recommendation for avoiding unsafe exposure is to use a 125 dB SPL stimulus for individuals with an equivale

Long-term moderate noise exposure enhances the medial olivocochlear reflex.

OBJECTIVE: To investigate the effects of long-term moderate noise on hearing functions, MOCR, and MEMR. METHODS: Mice were exposed to the moderate noise (11.2 - 22.4 kHz, 80 dB SPL, 6 h/day, 4 weeks). Subsequently, the hearing functions, including threshold and input-output roles of ABR (auditory brainstem response) and cubic (2f1-f2) DPOAEs (distortion product otoacoustic emissions) were evaluated. Also, MEMR and MOCR were assessed shortly after or at four weeks following the termination of exposure to the noise. RESULTS: The mice's acoustic suppression reflex was strengthened, hearing func

Relationship Between Distortion Product - Otoacoustic Emissions (DPOAEs) and High-Frequency Acoustic Immittance Measures.

BACKGROUND Pathologies that alter the impedance of the middle ear may consequently modify the DPOAE amplitude. The aim of this study was to correlate information from 2 different clinical procedures assessing middle ear status. Data from DPOAE responses (both DP-Gram and DP I/O functions) were correlated with data from multi-component tympanometry at 1000 Hz. MATERIAL AND METHODS The subjects were divided into a double-peak group (DPG) and a single-peak group (SPG) depending on 1000 Hz tympanogram pattern. Exclusion criteria (described in the Methods section) were applied to both groups and fi

A connection between the Efferent Auditory System and Noise-Induced Tinnitus Generation. Reduced contralateral suppression of TEOAEs in patients with noise-induced tinnitus.

BACKGROUND: Subjective tinnitus is an auditory perception that is not caused by external stimulation, its source being anywhere in the auditory system. Furthermore, evidence exists that exposure to noise alters cochlear micromechanics, either directly or through complex feed-back mechanisms, involving the medial olivocochlear efferent system. The aim of this study was to assess the role of the efferent auditory system in noise-induced tinnitus generation. MATERIAL/METHODS: Contralateral sound-activated suppression of TEOAEs was performed in a group of 28 subjects with noise-induced tinnitus (

Wideband ipsilateral measurements of middle-ear muscle reflex thresholds in children and adults.

The goals of the current study were to: 1) evaluate the feasibility of a new wideband approach to measuring middle-ear muscle reflex (MEMR) status, and 2) to test the hypothesis that ipsilateral thresholds elicited with 1 or 2 kHz tones and broadband noise activators on a wideband acoustic transfer function (WATF) system are lower than thresholds elicited on a clinical system. Clinical MEMR tests have limitations, including the need for high activator levels to elicit a shift in a narrowband probe (e.g., a 0.226 or 1 kHz tone). Wideband MEMR tests using WATFs may elicit the reflex at lower lev

Acoustic reflexes to Schroeder-phase harmonic complexes in normal-hearing and hearing-impaired individuals.

Harmonic complexes generated with positive or negative Schroeder-phases may result in differences in cochlear excitation, even though their long-term spectra and amplitudes are equal. As a measure of possible differences in cochlear excitation resulting from these harmonic complexes, thresholds and growth of the acoustic reflex were assessed in normal-hearing and hearing-impaired subjects. Harmonic complexes with fundamental frequencies of 50, 100, and 200 Hz were constructed with positive and negative-Schroeder phases. In normal-hearing subjects, acoustic reflex thresholds for the 50- and 100

The multichannel auditory brainstem implant: how many electrodes make sense?

OBJECT: Development of multichannel auditory brainstem implant (ABI) systems has been based in part on the assumption that audiological outcome can be optimized by increasing the number of available electrodes. In this paper the authors critically analyze this assumption on the basis of a retrospective clinical study performed using the Nucleus 22 ABI surface electrode array. METHODS: The perceptual performances of 61 patients with neurofibromatosis Type 2 were tested approximately 6 weeks after an eight-electrode ABI had been implanted. Of eight implanted electrodes 5.57 +/- 2.57 (mean +/- s

Dynamic range of the contralateral stapedius reflex in cochlear implant patients.

The dynamic range of the contralateral Stapedius reflex elicited by analog electrostimulation via cochlear implant was investigated in 8 subjects supplied with a Vienna prosthesis. The study comprised patients fitted with intracochlear devices as well as patients with extracochlear devices. Sinusoidal bursts of 125, 500, 1,000 and 2,000 Hz were used as stimulation signals. Only in one patient was saturation of reflex amplitude observed. The dynamic range of the Stapedius reflex was defined either by the difference in stimulus level for reflex saturation or individual uncomfortable loudness lev

Precise sound characteristics drive plasticity in the primary auditory cortex with VNS-sound pairing.

INTRODUCTION: Repeatedly pairing a tone with vagus nerve stimulation (VNS) alters frequency tuning across the auditory pathway. Pairing VNS with speech sounds selectively enhances the primary auditory cortex response to the paired sounds. It is not yet known how altering the speech sounds paired with VNS alters responses. In this study, we test the hypothesis that the sounds that are presented and paired with VNS will influence the neural plasticity observed following VNS-sound pairing. METHODS: To explore the relationship between acoustic experience and neural plasticity, responses were reco

Vibration Characterization of the Human Knee Joint in Audible Frequencies.

Injuries and disorders affecting the knee joint are very common in athletes and older individuals. Passive and active vibration methods, such as acoustic emissions and modal analysis, are extensively used in both industry and the medical field to diagnose structural faults and disorders. To maximize the diagnostic potential of such vibration methods for knee injuries and disorders, a better understanding of the vibroacoustic characteristics of the knee must be developed. In this study, the linearity and vibration transmissibility of the human knee were investigated based on measurements collec

Fibromyalgia as a disorder of perceptual organization? An analysis of acoustic stimulus processing in patients with widespread pain.

We examined to what extent patients with fibromyalgia differ from painfree control subjects in the perception and processing not only of somatosensory but also of external stimuli. For this purpose the acoustic perception of 30 patients with fibromyalgia was compared with that of 36 generally pain-free age and gender matched subjects. The groups were also controlled for organic disease of pathological dysfunction of the ear and auditory nerves. Thresholds of unpleasantness and hearing thresholds were determined autiometrically for various frequencies. In addition the participants rated their e
