Large liposome agglutination technique for the serological detection of syphilis.

In response to tissue invasion by Treponema pallidum, an antibody complex, called reagin, appears in the serum of the syphilitic patients. Reagin has the ability to combine with cardiolipin, therefore, cardiolipin has traditionally been used as the antigen in different test configurations for the serological detection of syphilis: the VDRL (Venereal Disease Research Laboratory) flocculation test and the RPR (rapid plasma reagin) test. Here we introduce a liposome preparation composed of cardiolipin, lecithin and cholesterol and containing a colored dye which upon mixing with syphilitic serum o

[Serologic demonstration of antibodies against Campylobacter jejuni/coli in man].

Antibody response to Campylobacter jejuni/coli infections in man was studied by microagglutination assay against homologues organisms isolated from patients and by complement fixation test against a commercially available group-specific antigen of C. jejuni/coli. Titers of agglutinating antibodies raise within 1-2 weeks post infectionem (p.i.) to an extent of about 320 (reciprocal titer) and persist about 3-4 months. CF-antibody titers raise only to about 40 but they persist for a long time. Because of the low titers in the CF-test and because of a missing seroconversion the CF-test is not sui

Suitability of the microagglutination test for detection of post-infection and post-vaccination Q fever antibodies in human sera.

Serological examination by the microagglutination (MA) and complement-fixation (CF) tests of human sera collected before and after vaccination with Q fever chemovaccine revealed a higher sensitivity of the MA test for detecting both pre-vaccination antibodies reflecting a previous exposure to Q fever and post-vaccination antibodies reflecting vaccine immunogenicity. In persons serologically positive before vaccination the level of post-vaccination MA antibody response was indirectly proportional to the titres of prevaccination MA antibodies.

Identification of the most effective serovars to be included in the MAT antigen panel to optimize the serodiagnosis of Leptospira infection in Northern Italy.

The microscopic agglutination test (MAT) assay is adopted as a world-wide reference test for the serodiagnosis of leptospirosis in humans and animals. One of the main limitations of MAT is the lack of sensitivity and serodiagnostic antigens should be periodically updated with locally circulating serovars in order to optimise its performance. The aim of this study was to determine the need to implement the antigen panel currently adopted in Northern Italy for the diagnosis of Leptospira infection in dogs. For this purpose, a group of 288 dogs with and without clinical signs potentially consiste

New developments in the diagnostic procedures for zoonotic brucellosis in humans.

At present, laboratory diagnosis of human brucellosis is based on isolation of the bacteria from clinical samples followed by standard microbiological tube testing, detection of anti-Brucella antibodies using various serological tests, and the use of molecular methods for the detection of Brucella DNA. None of these diagnostic tools can be used on its own to reliably detect the causative agent. Cultures give a low yield and subsequent phenotypic characterisation is time consuming, meaning that the initiation of adequate antibiotic therapy is frequently delayed. Serological tests seem to be mor

Neutrophil antigens and antibodies in the diagnosis of immune neutropenias.

Neutrophil specific antigens (NA) are expressed exclusively on human neutrophils and were identified using alloantibodies. Neutrophil specific antigens are polymorphic, and several of them (NA1, NA2, NB1, NB2, NC1, ND1, NE1, and 9A), are thought to define genes at different loci. Feto-maternal incompatibility of NA has resulted in alloimmune neonatal neutropenia. Also, NA are the target antigens for autoantibody production in infants and young children with autoimmune neutropenia of infancy and chronic idiopathic neutropenia in adults. Autoimmune neutropenia can occur secondary to several othe

Bovine pneumonic pasteurellosis: experimental induction in vaccinated and nonvaccinated calves.

A study was undertaken to investigate the effects of the immune response induced by combined aerosol and parenteral vaccination on the lung lesions induced in calves by Pasteurella haemolytica AI. Twenty-four calves, twelve of which had been vaccinated with killed P. haemolytica by aerosol and subcutaneous injection in Freund's complete and incomplete adjuvant were challenged by intratracheal inoculation of live P. haemolytica. Serological response to vaccination was not marked but was best measured by the whole cell agglutination test or by indirect bacterial agglutination rather than by the
