Cocaine- and amphetamine-regulated transcript: a novel regulator of energy homeostasis expressed in a subpopulation of pancreatic islet cells.

Type 2 diabetes is characterised by chronic hyperglycaemia and its incidence is highly increased by exaggerated food consumption. It results from a lack of insulin action/production, but growing evidence suggests that it might also involve hyperglucagonaemia and impaired control of glucose homeostasis by the brain. In recent years, the cocaine and amphetamine-regulated transcript (CART) peptides have generated a lot of interest in the battle against obesity because, via the brain, they exert anorexic effects and they increase energy expenditure. They are also localised, outside the brain, in d…

Memory and brain-derived neurotrophic factor after subchronic or chronic amphetamine treatment in an animal model of mania.

Progression of bipolar disorder (BD) has been associated with cognitive impairment and changes in neuroplasticity, including a decrease in serum brain-derived neurotrophic factor (BDNF). However, no study could examine BDNF levels directly in different brain regions after repeated mood episodes to date. The proposed animal model was designed to mimic several manic episodes and evaluate whether the performance in memory tasks and BDNF levels in hippocampus, prefrontal cortex, and amygdala would change after repeated amphetamine (AMPH) exposure. Adult male Wistar rats were divided into subchroni…

The cocaine- and amphetamine-regulated transcript, calbindin, calretinin and parvalbumin immunoreactivity in the medial geniculate body of the guinea pig.

The purpose of this study was to describe the distribution and colocalization of cocaine- and amphetamine-regulated transcript (CART) and three calcium-binding proteins (calbindin, calretinin and parvalbumin) in each main division of the medial geniculate body (MGB) in the guinea pig. From low to moderate CART immunoreactivity was observed in all divisions of the MGB, although in most of its length only fibers and neuropil were labeled. A small number of CART immunoreactive somata were observed in the caudal segment of the MGB. The central parts of all divisions contained a distinctly smaller …

Changes of cytosolic calcium and contractility of young rat vas deferens by acute treatment with amphetamine, fluoxetine or sibutramine.

Previous studies conducted in our laboratory indicated that administration of amphetamine, fluoxetine or sibutramine affects the sympathetic nervous system of the rat vas deferens. Therefore, our goal was to verify the role of calcium in vasa deferentia from young rats pretreated with a single dose of these drugs. Young 40-day-old male Wistar rats were pretreated with amphetamine 3 mg/kg, fluoxetine 10 mg/kg or sibutramine 6 mg/kg for 4 h before the experiments. CaCl(2) (10 mM) was used to induce contraction through time-effect curves in calcium-free solution to measure phasic and tonic compon…

Effects of a gastrin-releasing peptide receptor antagonist on D-amphetamine-induced oxidative stress in the rat brain.

Previous studies have suggested that bipolar disorder may be associated with oxidative stress. Administration of D: -amphetamine (AMPH) has been put forward as an animal model of mania, and has shown to increase oxidative stress parameters in the rat brain. Thus, we have used the gastrin-releasing peptide receptor antagonist [D-Tpi(6)Leu(13)psi (CH(2)NH)-Leu(14)] bombesin (RC-3095) as a pharmacological tool to investigate the role of bombesin-like peptides in the redox balance in the hippocampus and cortex of rats treated with AMPH. Rats were given a single 10 ml/kg intraperitoneal (i.p.) inje…

Cocaine and amphetamine regulated transcript (CART) gene in the comorbidity of schizophrenia with alcohol use disorders and nicotine dependence.

Studies have shown a genetic susceptibility to develop schizophrenia, alcohol use disorders and nicotine dependence. Brain areas related to reward and reinforcement show high expression of the cocaine and amphetamine regulated transcript (CART). Nicotine and alcohol are also able to modulate CART expression in the hypothalamic areas. In this study, we evaluated whether CART variants would influence the predisposition of schizophrenia subjects to alcohol use disorders and nicotine dependence. Clinical and genetic data were obtained from 190 unrelated Caucasian schizophrenia subjects collected a…

The reinforcing effects of chronic D-amphetamine and morphine are impaired in a line of memory-deficient mice overexpressing calcineurin.

It has recently emerged that there is a commonality in the molecular mechanisms underlying long-term neuronal changes in drug addiction and … ng-term memory. Here, we investigated the involvement of calcineurin in the motivational effects of amphetamine and morphine using this line of transgenic mice (CN98). Our results showed that amphetamine and morphine did not induce conditioned place preference in calcineurin-mutant mice, whereas food remained an efficient reinforcer. In addition, behavioural …

Individual differences in cocaine- and amphetamine-induced activation of male Sprague-Dawley rats: contribution of the dopamine transporter.

Previously we found that outbred male Sprague-Dawley rats can be classified as either low or high cocaine responders (LCRs or HCRs, respectively), based on their open-field locomotor response to acute cocaine (COC; 10 mg/kg, i.p.). Here, we extended this analysis to amphetamine (AMPH; 0.5, 1, and 5 mg/kg, i.p.) and found that the individual differences in behavioral activation were not as pronounced as with COC. This was confirmed with observational analysis of behaviors. Differences in drug-induced activation could involve differential dopamine transporter (DAT) function/trafficking. To addre…

Interaction between the noradrenergic and serotonergic systems in locomotor hyperactivity and striatal expression of Fos induced by amphetamine in rats.

It is classically considered that Amphetamine acts by increasing extracellular dopamine levels. However, some data suggest a relevant role of other neurochemical systems. The striatum is of particular interest to the study of this question. We have investigated the involvement of the noradrenergic and serotonergic systems and their possible interaction in the striatal responses to Amphetamine using a double behavioral and immunohistochemical approach (i.e., changes in locomotor activity and striatal expression of Fos). In normal rats, Amphetamine induced locomotor hyperactivity and striatal ex…

The VMAT2 gene in mice and humans: amphetamine responses, locomotion, cardiac arrhythmias, aging, and vulnerability to dopaminergic toxins.

Monoamine compartmentalization in monoaminergic neurons uses serial action of the plasma membrane and vesicular monoamine (VAMT2) transporters. We can now define the sequences of the genes encoding these transporters in mice and humans, examine influences of deletions of this gene and alteration in its expression levels in transgenic mice, and identify sequence polymorphisms in the human VMAT2 gene. Examination of VMAT2 variants can provide potential insights into roles for allelic variants at these loci in variant drug responses and in diseases linked to monoaminergic systems, including subst

d-Amphetamine-induced hepatotoxicity: possible contribution of catecholamines and hyperthermia to the effect studied in isolated rat hepatocytes.

Amphetamines are indirect-acting sympathomimetic drugs widely abused due to their physical and psychostimulating effects. However, the use of these drugs has been associated with numerous reports of hepatotoxicity. While glutathione depletion induced by amphetamines contributes to the exposure of hepatocytes to oxidative damage, other indirect effects attributed to amphetamines may have a role in cell injury. To examine this possibility, Wistar rats were used for plasma measurements of d-amphetamine and catecholamines (noradrenaline, adrenaline and dopamine) (15 min) after i.p. injection of d-…

D1 and D2 dopamine and opiate receptors are involved in the restraint stress-induced sensitization to the psychostimulant effects of amphetamine.

The time course of the restraint stress-induced sensitization to the stimulant effects of amphetamine (AMPH, 0.5 mg/kg IP) on locomotor activity was investigated for up to 8 days. In a series of separate experiments, the involvement of opioid and dopaminergic mechanisms in the development of acute restraint stress-induced behavioral sensitization were characterized. Both a single restraint session (2 h) and chronic restraint (2 h per day for 7 days) similarly potentiated the effects of AMPH on motor activity. This behavioral sensitization was prevented by the administration of naltrexone (2 mg…

The role of opioid mechanisms in the anorectic effects of stimulants: U50,488H enhances amphetamine inhibition of free feeding in rats.

The influences of the kappa-opiate agonist U50,488H (U50; 4 mg/kg IP), the neuroleptic haloperidol (HAL; 0.3 mg/kg IP), and MK-801 (0.2 mg/kg IP), a noncompetitive antagonist for NMDA receptors, were compared on the effects of nine days of d-amphetamine (AMPH) treatment (3 mg/kg IP) on food and water intake and urine output. AMPH prevented feeding stimulation produced by U50 during the first 2 h, whereas U50 inhibited the hyperphagic phase that rats showed between 2 and 5 h after AMPH administration. Tolerance did not develop to the first 2-h suppression of feeding; in contrast, the late hyper…

D-amphetamine-like stimulus properties are produced by morphine injections into the ventral tegmental area but not into the nucleus accumbens.

We investigated whether injections of morphine into the ventral tegmental area (VTA) or the nucleus accumbens (NAS) could produce amphetamine-like stimulus properties and locomotor stimulant effects. Rats were trained to discriminate 1.0 mg/kg D-amphetamine from saline using both VI-30 and FR-16 reinforcement schedules and they were then tested following bilateral injections of morphine sulfate (2.5, 5.0 and 10.0 micrograms/side) either into the VTA or the NAS. Posttraining effects of amphetamine, chlorpromazine, ketamine, and scopolamine on the acquisition and extinction of the rabbit's conditioned nictitating membrane response.

Four experiments examined whether posttraining deliveries of drugs modified the performance of the rabbit's conditioned nictitating membrane response (NMR) during acquisition and extinction. The results show that ketamine accelerated, but that amphetamine, chlorpromazine, and scopolamine retarded, conditioning when the drugs were injected immediately after the completion of daily training sessions. However, all conditioning effects of the drugs were lost when their deliveries were delayed by 2 hr after the end of training. During extinction, the only drug that altered conditioned performance w…
