Facial morphogenesis relies on the coordinated regulation of cellular behaviors that sculpt the face from simple facial prominences to the fully confluent lip and nose. Disruptions to migration of cranial neural crest cells and/or the directed growth of the prominences will lead to abnormalities such as orofacial clefts. Here we review what is known about the roles of the actin cytoskeleton and its key regulators, the small RHO GTPases, during neural crest cell migration and facial development. Although small RHO GTPase signaling has been studied in the context of cancer and vascular pathologi

BACKGROUND: Fibrillins provide a scaffold for elastic fiber formation, which enables lung recoil and aortic compliance. Abnormal fibrillin microfibrils, as in Marfan syndrome, lead to enlarged alveoli, vascular stiffening, and aneurysms. Our earlier studies suggested that fibrillin function depends on O-glucosylation of its epidermal growth factor (EGF)-like repeats by protein O-glucosyltransferase 2 (POGLUT2) and 3 (POGLUT3). Mice lacking both enzymes (Poglut2/3 DKO) die soon after birth. Rare survivors are runted, have digit 2-3 syndactyly, and variable eye defects. Here, we wanted to determ

BACKGROUND: Craniofacial development is a complex developmental process that involves formation of the skeleton, muscle, and tendons. Defects in craniofacial development result in common hereditary disorders. Among genetic factors regulating craniofacial development, dysregulated hedgehog signaling is associated with craniofacial skeletal defects such as orofacial clefting and holoprosencephaly. Here, we characterize craniofacial phenotypes associated with two hedgehog signaling co-receptors, cdon and boc, in zebrafish. Genetic pedigree analyses have previously linked both cdon and boc to micr

BACKGROUND: The zebrafish heart regenerates upon injury. During injury response, fibroblasts and endothelial cells accumulate at the site of damage, and cardiomyocyte cell cycle reentry allows cardiac muscle regrowth. It is relevant to understand how the different cell types communicate with each other to coordinate regeneration.
RESULTS: We present an in silico meta-analysis of ligand-receptor (LR) interactions among periostin b+ fibroblasts, kdlr+ endothelial cells, sox10-derived and rest of ventricular cardiomyocytes, at 7 days post-injury. Using bulk RNA-seq data sets from fluorophore-act

BACKGROUND: Elp1, a subunit of the Elongator complex, is essential for tRNA modification and neuronal development. Mutations in ELP1 underlie familial dysautonomia (FD), a disorder marked by sensory and autonomic neuropathy. While loss of Elp1 disrupts trigeminal ganglion formation and survival, the downstream molecular consequences remain poorly defined.
RESULTS: We performed quantitative proteomic profiling of trigeminal ganglia from Elp1 conditional knockout (CKO) and control embryos at E13.5. Across 5650 detected proteins, 25 were significantly up-regulated and 26 down-regulated in Elp1 C

The olfactory placode (OP) generates a broad array of chemosensory neurons in the nasal region, including olfactory sensory neurons, vomeronasal sensory neurons, neurons of the septal organ, and Grueneberg ganglion. During invagination, the OP also generates migratory neuronal populations, including gonadotropin-releasing hormone-1 (GnRH-1) neurons, somatostatin-expressing neurons, and Prokineticin Receptor 2 (Prokr2) pioneer/terminal nerve (TN) neurons. Despite extensive research, the genetic lineage and molecular identity of many OP-derived neurons remain unclear. GnRH-1 neurons are vital fo

BACKGROUND: In the developing brain, neuronal migration is one of the critical steps in the establishment of an accurate neural network. When neurons reach their final destination, they form layered structures or nuclei. Precerebellar neurons (PCNs) form several nuclei in the pons and medulla and project mossy and climbing fibers to the cerebellum. PCNs originate from the lower rhombic lip in the dorsal hindbrain and migrate tangentially toward their destinations. When PCNs reach their presumptive nucleus-forming region, they change their migration direction from tangential to radial to form t

BACKGROUND: Sexual dimorphism in pelvic morphology has been confirmed at the onset of primary ossification. Since pelvic tilt influences spinal alignment and bipedal locomotion, determining the initial emergence of sex-specific tilt differences is crucial. Herein, we measured pelvic tilt and lumbosacral inclination angles (LSIAs) using magnetic resonance imaging in 72 human fetuses (crown-rump length [CRL], 50-225 mm; 34 males [M] and 38 females [F]).
RESULTS: The cranial-caudal axis was defined from the first to third sacral vertebrae, and sex differences were assessed using multiple regress

BACKGROUND: One of the most characteristic morphogenetic processes in Drosophila is the 360° rotation of the male pupal genital disc. This movement is driven by the myosin Myo1D, whose expression in the genital disc is controlled by the Hox gene Abdominal-B (Abd-B). The rotation takes place in contact and relative to the posterior abdomen, yet the contribution of abdominal tissues has remained unclear.
RESULTS: Here we show that normal genital disc circumrotation requires active remodeling of posterior abdominal larval epidermal cells that contact the rotating terminalia. Preventing apoptosis

BACKGROUND: Transcription factors of the MIT/TFE family-mitfa, mitfb, and tfec-play essential roles in specifying pigment cells derived from neural crest cells in vertebrates. In teleosts, which possess multiple pigment cell types, these factors exhibit partially redundant but largely distinct functions across different pigment lineages. We previously reported that the combined loss of mitfa and mitfb leads to a complete absence of melanophores, xanthophores, and leucophores in medaka.
RESULTS: Here, we describe newly generated hypomorphic alleles of mitfa and mitfb, which result in delayed m

BACKGROUND: The notochord is a midline structure essential for vertebrate embryogenesis, contributing to the development of the nervous system, digestive tract, and vertebral column. In particular, notochord signaling is indispensable for proper patterning and coordinated development of alternating vertebrae and intervertebral discs (IVDs). Later, notochordal cells (NCs) mature and adopt a characteristic vacuolated morphology before giving rise to the core of the forming IVD, the nucleus pulposus (NP). Postnatally, NCs play a pivotal role in maintaining disc integrity through the secretion of

BACKGROUND: Histone post-translational modification (PTM) is an important epigenomic regulation content and an essential process regulating gene expression. Histone lysine lactylation is the newly identified histone PTM that utilizes the lactyl moiety for its modification. Although histone lysine lactylation is considered an essential outcome of the Wardburg effects and the interconnection between cellular metabolism and gene regulation, the developmental contexts involving this PTM are largely unknown. In this study, we comprehensively observed histone lysine lactylation during Drosophila oog

BACKGROUND: The pancreas exhibits diverse structures and roles across vertebrates. The pancreas has evolved to include both endocrine and exocrine cells, a change that occurred during the transition from fish to amphibian. This event emphasizes the evolutionary significance of amphibians. However, research has focused predominantly on anuran amphibians, with urodeles, such as newts, remaining underexplored. In this study, we investigated the development of the pancreas using Pleurodeles waltl as a model species of urodele.
RESULTS: The newt pancreas consists of a single organ with exocrine ti

Maternal effects, encompassing both genetic (maternally expressed gene products) and non-genetic (maternal state) influences, are powerful determinants of offspring phenotype, yet their RNA-level mechanisms remain incompletely resolved. In parallel, epitranscriptomics, an emerging field centered on chemical modifications to RNA, has revealed new layers of gene regulation with implications for cell fate, plasticity, and response to environmental cues. In this perspective article, a conceptual link is proposed between maternal effects and epitranscriptomic mechanisms, focusing on how maternal en

BACKGROUND: Phosphatidylserine synthase (PSS), localized in the mitochondrial membrane, synthesizes phosphatidylserine. In humans, mutations in Pss lead to Lenz-Majewski hyperostotic dwarfism, a disorder affecting growth and development. The effects of Pss mutations on the growth of Drosophila melanogaster are not fully known. Hence, this study was conducted to investigate the effects of Pss knockdown on the growth and development of D. melanogaster.
RESULTS: Enterocyte (EC)-specific Pss knockdown resulted in reduced cell size in the gut via reduced Akt signaling. EC-specific Pss knockdown wa

BACKGROUND: Dentitions have diversified enormously during vertebrate evolution, involving reductions, modifications, or allocations to prey seizing and processing regions. A combination of ancient and novel features related to dental and oropharyngeal apparatuses is found in extant lineages of non-teleost fishes, such as the gars. While relevant to evolutionary-developmental studies, gars have largely been overlooked regarding how their dentition arises, thus leaving our comprehension of the evolutionary history of vertebrate dentitions incomplete.
RESULTS: Here, we complement this knowledge

BACKGROUND: Actin filament organization in cardiomyocytes critically depends on the formin Fhod3, but a role for Fhod3 in skeletal muscle development has not yet been described.
RESULTS: We demonstrate here that in zebrafish mutated for one of two fhod3 paralog genes, fhod3a, skeletal muscle of the trunk appears normal through 2 days post-fertilization, but afterward exhibits myofibril damage, including gaps between myofibrils and myofibril fragmentation. Despite the progressive nature of the myofibril damage, fhod3a mutants differ from muscular dystrophy models in that damage is exacerbated

BACKGROUND: Sporadic venous malformation (VM) is associated with the hyperactivating p.L914F mutation in TIE2, a receptor tyrosine kinase essential for vascular development. This mutation is not found in hereditary VM, suggesting incompatibility with life when expressed during early vascular development. Therefore, we utilized a genetic mouse model that expresses TIE2 p.L914F to determine its phenotypical effects during development.
RESULTS: B6-Tg(Rosa26-TIE2L914F)EBos (TIE2L914F) mice were generated and then validated for the presence of the transgene. The constitutive endothelial-specific T

BACKGROUND: Previously, we explored the role of Sox9 in the second heart field (SHF) in atrioventricular septation. For that study, we created a SHF-specific Sox9 knockout mouse. In addition to the presence of primary atrial septal defects in half of the offspring, we found that virtually all specimens also developed a ventricular septal defect. Histological analysis suggested that the ventricular septal defects resulted from developmental perturbation of the mesenchymal structures within the outflow tract. In the current study, we investigated the role of Sox9 in the SHF in the development of

BACKGROUND: Robinow syndrome is a rare developmental syndrome caused by variants in genes in Wnt signaling pathways. We previously showed that expression of patient variants in Dishevelled 1 (DVL1) in Drosophila and chicken models disrupts the balance of canonical and non-canonical Wnt signaling.
RESULTS: In this study, we further examine morphological changes that occur due to expression of DVL11519ΔT, which serves as a prototype for other pathogenic variants. We show that epithelial imaginal disc development is disrupted in legs and wings and accompanied by increased cell death, without cha
