Slow flow in regenerating C-fibers.

An identical rate of slow flow was measured in 3 different populations of regenerating C-fibers which have very different growth abilities as expressed by a 6-fold difference in their elongation velocities. At 21 degrees C, this rate of slow flow is 3.5 times faster than the rate measured in intact nerves and is identical to the elongation velocity of the most rapidly regenerating fibers. Slow flow may constitute the upper limit of the elongation velocity of a regenerating axon but other factors determined by the perikaryon (such as protein synthesis) might prevent the neuron from reaching the

Retinotopic analysis of fiber pathways is amphibians. I. The adult newt Cynops pyrrhogaster.

The possibility that pathways of retinal fibers within the optic tract and the tectum of the adult newt are retinotopic was examined by selective labeling of the retinal fibers with horseradish peroxidase. Within the optic tract fibers from the ventral, temporal and dorsal retinal quadrants were ordered from the dorsal to ventral edges of th optic tract. The nasal retinal fibers exhibited two different pathways. The fibers from the dorsonasal retina ran along the ventral edge of the optic tract, while the fibers from the ventronasal retina ran along the dorsal edge of the optic tract. Segregat

Identification of cells of origin of tectopontine fibers in the cat superior colliculus: an experimental study with the horseradish peroxidase method.

The pontine projections from the superior colliculus in the cat have been studied by means of retrograde axonal transport of horseradish peroxidase (HRP). Following injections of HRP in the dorsolateral pontine nucleus, where the tectopontine fibers terminate, a fair number of labeled cells are found throughout the rostrocaudal extent of the ipsilateral superior colliculus. Relatively few of the labeled cells are of medium size (25-40 micron in diameter), more than 80% are small (10-25 micron), but no large cells are labeled. The cell bodies giving rise to tectopontine fibers are distributed i…

Mapping sensory circuits by anterograde transsynaptic transfer of recombinant rabies virus.

Primary sensory neurons convey information from the external world to relay circuits within the CNS, but the identity and organization of the neurons that process incoming sensory information remains sketchy. Within the CNS, viral tracing techniques that rely on retrograde transsynaptic transfer provide a powerful tool for delineating circuit organization. Viral tracing of the circuits engaged by primary sensory neurons has, however, been hampered by the absence of a genetically tractable anterograde transfer system. In this study, we demonstrate that rabies virus can infect sensory neurons in

Identification of spinal electromotoneurons in the ray Raja clavata (Rajidae).

Retrograde transport of horseradish peroxidase injected ipsilaterally in several parts of the electric organ and light microscopy were employed to study motoneurons and electromotoneurons of the spinal cord in the weakly electric ray Raja clavata (Rajidae). The horseradish peroxidase-labelled electromotoneurons form a chain of cells located ventrolaterally in the gray matter of the spinal cord segments adjacent to the electric organ. The diameters of cells in the electromotor nucleus vary from 70 to 110 microns. When comparing other motoneurons of the spinal cord with electromotoneurons, the l

The nervus terminalis also exists in cyclostomes and birds.

The terminal nerve has been described in all vertebrate classes, with the exception of cyclostomes and birds. With regard to this question, we have examined representatives of these two classes using tracer techniques, and found a terminal nerve in larval lampreys and young domestic mallards. Horseradish peroxidase or cobaltous lysine was injected into the olfactory mucosa, which is known to be innervated by peripheral branches of the terminal nerve. The brains were then searched for labeled, centrally directed fibers of the terminal nerve that project further caudally than the glomerular laye

Further observations on the olivocerebellar projection in the monkey.

The distribution of retrogradely labeled cells in the inferior olive was studied following cerebellar injections of horseradish peroxidase in four experiments in macaque monkeys. The flocculus was found to receive its main projection from the dorsal cap and a minor projection from the medial accessory olive. The uvula receives fibers from the nucleus beta, the dorsomedial cell column and from an area of the medial accessory olive. The paramedian lobule is supplied from the dorsal and ventral lamella of the principal olive and from an area in each of the accessory olives. The findings are consi

The efferent connections of the ventromedial nucleus of the hypothalamus of the rat.

The efferent connections of the ventromedial nucleus of the hypothalamus (VMH) of the rat have been examined using the autoradiographic method. Following injections of small amounts (0.4-2.0 muCi) of tritium labeled amino acids, fibers from the VMH can be traced forward through the periventricular region, the medial hypothalamus and the medial forebrain bundle to the preoptic and thalamic periventricular nuclei, to the medial and lateral preoptic areas, to the bed nucleus of the stria terminalis and to the ventral part of the lateral septum. Some labeled axons continue through the bed nucleus

Peripheral neuropathy in mitochondrial disease.

Mitochondria are essential for the health and viability of both motor and sensory neurons and their axons. Processes that disrupt their normal distribution and transport along axons will likely cause peripheral neuropathies. Similarly, mutations in mtDNA or nuclear encoded genes result in neuropathies that either stand alone or are part of multisystem disorders. This chapter focuses on the more common genetic forms and characteristic clinical phenotypes of "mitochondrial" peripheral neuropathies. We also explain how these various mitochondrial abnormalities cause peripheral neuropathy. In a pa

Modelling the pathogenesis of X-linked distal hereditary motor neuropathy using patient-derived iPSCs.

ATP7A encodes a copper-transporting P-type ATPase and is one of 23 genes in which mutations produce distal hereditary motor neuropathy (dHMN), a group of diseases characterized by length-dependent axonal degeneration of motor neurons. We have generated induced pluripotent stem cell (iPSC)-derived motor neurons from a patient with the p.T994I ATP7A gene mutation as an in vitro model for X-linked dHMN (dHMNX). Patient motor neurons show a marked reduction of ATP7A protein levels in the soma when compared to control motor neurons and failed to upregulate expression of ATP7A under copper-loading c

Chronic toxic neuropathy after organophosphorus poisoning in quails (Coturnix coturnix japponica).

Peripheral neuropathy was induced by the long-term administration of organo-phosphorus compounds (phtalimid/phosmet) in quails (Coturnix coturnix japponica). After 4 weeks, the first symptoms of organophosphorus (OPC) poisoning (apathy, diarrhea) were present. During the second month of a daily administration of the toxic substance using the probe, an apparent clinical autonomic and peripheral neuropathy with ataxia had developed. Toxic disturbance of the nervous system was confirmed by the examination of spinal and cortical somatosensory evoked potentials (SEP) after tibial nerve stimulation.

Optic nerve axoplasm and papilledema.

A detailed review of optic nerve axoplasm is presented. A number of hypotheses have been postulated for the pathogenesis of papilledema associated with increased intracranial pressure. These hypotheses, mechanical and nonmechanical, are critically evaluated in relation to five essential features of papilledema. Theories, as well as clinical and experimental studies, of axonal transport are reviewed, and a new hypothesis is proposed: Papilledema is primarily a mechanical, nonvascular phenomenon in which an excess amount of extracellular fluid is present in the prelaminar region of the optic dis…

Nifuroxazide rescues the deleterious effects due to CHCHD10-associated MICOS defects in disease models.

The identification of a point mutation (p.Ser59Leu) in the CHCHD10 gene was the first genetic evidence that mitochondrial dysfunction can trigger motor neuron disease. Since then, we have shown that this mutation leads to the disorganization of the MItochondrial contact site and Cristae Organizing System (MICOS) complex that maintains the mitochondrial cristae structure. Here, we generated yeast mutant strains mimicking MICOS instability and used them to test the ability of more than 1600 compounds from two repurposed libraries to rescue the growth defect of those cells. Among the hits identif

Pridopidine rescues BDNF/TrkB trafficking dynamics and synapse homeostasis in a Huntington disease brain-on-a-chip model.

Huntington disease (HD) is a neurodegenerative disorder caused by polyglutamine-encoding CAG repeat expansion in the huntingtin (HTT) gene. HTT is involved in the axonal transport of vesicles containing brain-derived neurotrophic factor (BDNF). In HD, diminished BDNF transport leads to reduced BDNF delivery to the striatum, contributing to striatal and cortical neuronal death. Pridopidine is a selective and potent sigma-1 receptor (S1R) agonist currently in clinical development for HD. The S1R is located at the endoplasmic reticulum (ER)-mitochondria interface, where it regulates key cellular …

Axonal regeneration from CNS neurons in the cerebellum and brainstem of adult rats: correlation with the patterns of expression and distribution of messenger RNAs for L1, CHL1, c-jun and growth-associated protein-43.

Some neurons in the brain and spinal cord will regenerate axons into a living peripheral nerve graft inserted at the site of injury, others will not. We have examined the patterns of expression of four molecules thought to be involved in developmental and regenerative axonal growth, in the cerebellum and brainstem of adult rats, following the implantation into the cerebellum of peripheral nerve grafts. We also determined how the expression patterns observed correlate with the abilities of neurons in these regions to regenerate axons. Three days to 16 weeks after insertion of living tibial nerv
