Upconversion fluorescent aptasensor for bisphenol A and 17β-estradiol based on a nanohybrid composed of black phosphorus and gold, and making use of signal amplification via DNA tetrahedrons.

This study describes an upconversion fluorescent aptasensor based on black phosphorus nanohybrids and self-assembled DNA tetrahedrons dual-amplification strategy for rapid detection of the environmental estrogens bisphenol A (BPA) and 17β-estradiol (E2). Tetrahedron complementary DNAs (T-cDNAs) were self-assembled in an oriented fashion on a 2D nanohybrid composed of black phosphorus (BP) and gold to give a materials of architecture BP-Au@T-cDNAs. In parallel, core-shell upconversion nanoparticles were modified with aptamers (UCNPs@apts) and used as capture probes. On complementary pairing, th

Passivation of black phosphorus as organic-phase enzyme platform for bisphenol A determination.

Black phosphorus (BP) has a high charge-carrier mobility (∼1000 cm2 V-1 s-1), but the bare BP degrades rapidly in the presence of oxygen and water which limits the application of the BP. In this study, a simple, non-covalent passivation strategy is developed by modifying of the BP with hexamethylendiamine (HA). The functionalized BP exhibits good stability over 4 weeks. The organic phase interdigital electrode which is constructed by stable HA/BP and tyrosinase displays lowest noise signal (0.025 nA) and relatively low detection limit (10 nmol L-1) for bisphenol A. This work provides a new str

Low-dose exposure to Bisphenol A during development has limited effects on male reproduction in midpubertal and aging Fischer 344 rats.

Low doses of Bisphenol A (BPA) during development may affect reproduction. In this study, Fischer 344 rats were exposed to 0.5 or 50 μg BPA/kg bw/day via drinking water from gestational day 3.5 to postnatal day 22. Anogenital distance, organ weight, histopathology of reproductive organs, hormone analysis and sperm morphology were evaluated in male offspring. In this study no major effects of BPA on male reproduction in midpubertal (postnatal day 35) or adult (12-month-old) rats were revealed, apart from a higher prevalence of mild inflammatory cell infiltrate in cauda epididymis in adult rats

Effects of bisphenol A treatment during pregnancy on kidney development in mice: a stereological and histopathological study.

Bisphenol A (BPA) is a chemical found in plastics that resembles oestrogen in organisms. Developmental exposure to endocrine-disrupting chemicals, such as BPA, increases the susceptibility to type 2 diabetes (T2DM) and cardiovascular diseases. Animal studies have reported a nephron deficit in offspring exposed to maternal diabetes. The aim of this study was to investigate the prenatal BPA exposure effects on nephrogenesis in a mouse model that was predisposed to T2DM. This study quantitatively evaluated the renal structural changes using stereology and histomorphometry methods. The OF1 pregnan

Thyroid hormone disrupting potentials of bisphenol A and its analogues - in vitro comparison study employing rat pituitary (GH3) and thyroid follicular (FRTL-5) cells.

As adverse health effects of bisphenol A (BPA) become a growing public health concern, the chemicals substituting BPA have been increasingly used in everyday lives. BPA substitutes have been frequently detected in both environment and biota in increasing levels. However, very limited toxicological information is available for these chemicals. In the present study, thyroid disrupting effects of nine structural analogues of BPA were evaluated along with BPA, using rat pituitary (GH3) and thyroid follicular (FRTL-5) cells. Similar to BPA, its analogues caused significant down-regulation of tshβ,

Evaluation of the potential health risks of substances migrating from polycarbonate replacement baby bottles.

Since the European Commission prohibited the use of bisphenol A in the production of polycarbonate (PC) baby bottles, many other materials have replaced PC for the manufacture of this type of food contact materials. In the present study, the potential migration risks associated with these alternative materials were investigated. First, all substances were evaluated for endocrine disruptive (ED) activity by using different existing lists of (suspected) ED chemicals. Next, the potential non-ED risks were assessed. A distinction was made between migrants listed in Annex I of European Regulation 1

Lifestyle and IVF Outcomes.

Whereas much has been written about the prognostic factors associated with outcomes of in vitro fertilization (IVF) such as female age, diagnosis, and ovarian reserve, relatively little attention has been devoted to patient-oriented lifestyles that may influence IVF outcomes. Patients are particularly interested in this topic because many patients wish to partner with their physicians and want to know specific behaviors to improve their chances of IVF success. This brief review is not intended as an exhaustive literature search of all possible lifestyles that may influence assisted reproductiv

Reproduction Symposium: developmental programming of reproductive and metabolic health.

Inappropriate programming of the reproductive system by developmental exposure to excess steroid hormones is of concern. Sheep are well suited for investigating developmental origin of reproductive and metabolic disorders. The developmental time line of female sheep (approximately 5 mo gestation and approximately 7 mo to puberty) is ideal for conducting sequential studies of the progression of metabolic and/or reproductive disruption from the developmental insult to manifestation of adult consequences. Major benefits of using sheep include knowledge of established critical periods to target ad

Bisphenol A: an endocrine and metabolic disruptor.

Bisphenol A (BPA), initially designed, like diethylstilbestrol, as a synthetic estrogen, has been rapidly and widely used for its cross-linking properties in the manufacture of polycarbonate plastics and epoxy resins. Because of incomplete polymerization and degradation of the polymers by exposure to higher than usual temperatures, BPA leaches out from food and beverage containers, as well as from dental sealants. In humans, free active unconjugated BPA is metabolized by rapid glucurono- or sulfo-conjugation and eliminated via renal clearance. However, exposure to environmental nanomolar conce

Exploring the mechanism link between endocrine disrupting chemicals and metabolic diseases: Observational and GWAS evidence.

Endocrine disrupting chemicals (EDCs) are widespread environmental contaminants that may contribute to the development of metabolic diseases. However, their causal roles and biological mechanisms remain unclear. This study aimed to investigate the associations between EDC exposure and metabolic disorders by integrating epidemiological and genetic approaches. A pooled analysis of 53 observational studies, comprising 109394 participants, demonstrated that EDC exposure significantly increased the risk of metabolic diseases (OR=1.38, 95 %CI=1.28-1.48, I2=82.3 %, P < 0.01). Positive associations we

Bisphenol S exposure induces cardiac remodeling and aggravates high-fat diet-induced cardiac hypertrophy in mice.

Bisphenol S (BPS) is widely used in the manufacture products and increase the risk of cardiovascular diseases. The effect of the association between obesity and BPS on cardiac outcomes is still unknown. Male C57BL/6 mice were divided into standard chow diet (SC; 15 kJ/g), standard chow diet + BPS (SCB), high-fat diet (HF; 21 kJ/g), and high-fat diet + BPS (HFB). Over 12 weeks, the groups were exposed to BPS through drinking water (dose: 25 μg/kg/day) and/or a HF diet. We evaluated: body mass (BM), total cholesterol, systolic blood pressure (SBP), left ventricle (LV) mass, and cardiac remodelin

Zebrafish (Danio rerio) TRβ- and TTR-based electrochemical biosensors: Construction and application for the evaluation of thyroid-disrupting activity of bisphenols.

Thyroid-disrupting chemicals (TDCs) have received increasing concerns because of their negative health impacts on both wildlife and humans. This study aimed to develop in vitro screening assays for TDCs based on thyroid hormone receptor β (TRβ) and transthyretin (TTR) proteins. Firstly, the recombinant ligand-binding domain of TRβ (TRβ-LBD) and TTR proteins of zebrafish were produced by eukaryotic expression system and then used as bio-recognition components to construct electrochemical biosensors. In the biosensors, the supported bilayer lipid membrane (s-BLM) was used as a matrix to immobili

Unveiling the presence of endocrine disrupting chemicals in northern French soils: Land cover variability and implications.

Endocrine disrupting chemicals (EDCs) are chemicals that can be found in the environment and have adverse effects on human health by mimicking, perturbing and blocking the function of hormones. They are commonly studied in water surfaces, rarely in soils, although it can be an important source of their presence in the environment. Their detection in soils is analytically challenging to quantify, hence the lack of known background concentrations found in the literature. This scientific research aimed to detect EDCs in soils by analyzing 240 soil samples using an optimized protocol of double ext

Organic and metallic pollutants in water treatment and natural wetlands: a review.

A literature review shows that more than 500 compounds occur in wetlands, and also that wetlands are suitable for removing these compounds. There are, however, obvious pitfalls for treatment wetlands, the most important being the maintenance of the hydraulic capacity and the detention time. Treatment wetlands should have an adapted design to target specific compounds. Aquatic plants and soils are suitable for wastewater treatment with a high capacity of removing nutrients and other substances through uptake, sorption and microbiological degradation. The heavy metals Cd, Cu, Fe, Ni and Pb were
