Increased energy expenditure through intermittent cold exposure does not enhance tirzepatide-induced weight loss in obese mice.

Obesity results from an imbalance between energy intake and energy expenditure. Although pharmacological approaches such as tirzepatide, a dual gastric inhibitory protein/glucagon-like protein-1 receptor agonist, reduce food intake and body weight, enhancing energy expenditure via intermittent cold exposure (ICE)-induced activation of thermogenesis may provide complementary benefits. Intermittent cold exposure increases energy expenditure and improves glucose homeostasis independent of weight loss. The objective of this study was to investigate the individual and combined effects of tirzepatid

Smoking and pursuit of thinness in schoolgirls in London and Ottawa.

It has been proposed that teenage girls often smoke cigarettes to protect themselves from the impulse to binge eat, with its feared weight-gain consequences, particularly when other measures such as greater dietary restraint have failed. The present study looked at the relationship between body mass index and standardised questionnaire responses concerning smoking, alcohol consumption, moods, weight changes, attitudes to body weight and shape, dietary patterns and menstruation in 1936 British (London) and 832 Canadian (Ottawa) schoolgirls. Data analysis revealed links between cigarette smoking

Gut microbial diversity at baseline conditions the clinical, microbiome, and metabolic response to paraprobiotic Lactiplantibacillus plantarum LRCC5282 in overweight adults.

The gut microbiota is increasingly recognized as a target for obesity management; however, whether baseline gut microbial diversity conditions responsiveness to microbiota-targeted interventions remains unclear. We aimed to investigate whether baseline gut microbial diversity is associated with responsiveness to a paraprobiotic derived from Lactiplantibacillus plantarum LRCC5282 (LP5282-P) in overweight adults. In a 12-week, randomized, double-blind, placebo-controlled, multicenter trial of 120 overweight adults, LP5282-P produced no significant between-group differences in any clinical outcom

Hormonal and metabolic effects of olanzapine and clozapine related to body weight in rodents.

OBJECTIVE: To characterize a model of atypical antipsychotic drug-induced obesity and evaluate its mechanism. RESEARCH METHODS AND PROCEDURES: Chronically, olanzapine or clozapine was self-administered via cookie dough to rodents (Sprague-Dawley or Wistar rats; C57Bl/6J or A/J mice). Chronic studies measured food intake, body weight, adiponectin, active ghrelin, leptin, insulin, tissue wet weights, glucose, clinical chemistry endpoints, and brain dopaminergic D2 receptor density. Acute studies examined food intake, ghrelin, leptin, and glucose tolerance. RESULTS: Olanzapine (1 to 8 mg/kg), b

Influence of body composition and cardiorespiratory fitness on plasma HSP72, norepinephrine, insulin, and glucose responses to an acute aerobic exercise bout performed in the fed state.

Being overweight is already considered a metabolic risk factor, which can be overcome by increasing cardiorespiratory fitness (CRF). Acute exercise is known to induce changes in plasma hormones and heat shock proteins release. However, there is a lack of studies investigating the impact of body composition and CRF on these variables following acute aerobic exercise. To assess the influence of body composition and cardiorespiratory fitness on plasma heat shock protein 72 kDa (HSP72), norepinephrine (NE), insulin, and glucose responses to an acute aerobic exercise bout in the fed state. Effect of low-dose ethanol on NLRP3 inflammasome in diabetes-induced lung injury.

To observe the changes in NLR family pyrin domain containing 3 (NLRP3) inflammasome in a rat model of diabetes-induced lung injury, and investigate the effect of low-dose ethanol on the production of NLRP3 inflammasome. The type I diabetic mellitus (DM) rat model was established, and the rats were divided into four groups: normal control group (CON group), low-dose ethanol group (EtOH group), diabetes group (DM group) and DM+EtOH group. The rats were fed for 6 and 12 weeks, respectively. The ratio of lung wet weight/body weight (lung/body coefficient) was calculated, and the changes of pulmona

[The effect of water soluble trichloromonofluoromethane freon on the weight and proteins of the heart muscle in Japanese quail].

An experiment was conducted to study the effects of water-soluble trichloromonofluoromethane freon (F-11, CC13F) on the weight of heart and on the level of freely extractable proteins in the heart of Japanese quail (Coturnix coturnix japonica). After three-week sublethal exposure of Japanese quail to the effect of freon the heart weight of the tested birds was found to have markedly decreased (however, the total body weight of the birds did not decrease). The tested quail also showed pronounced changes in the concentration of two protein fractions (an increase in one, a decline in the other),

Weight change patterns among breast cancer survivors: results from the Shanghai breast cancer survival study.

In a population-based cohort study of 5,014 women with stage 0-III breast cancer, we evaluated weight change patterns from diagnosis to 6, 18, and 36 months post-diagnosis. Patients were recruited to the study approximately 6 months after cancer diagnosis between 2002 and 2006 and followed through 36 months post-diagnosis. The medians of weight change from diagnosis to 6, 18, and 36 months post-diagnosis were 1.0, 2.0, and 1.0 kg, respectively. Approximately, 26% of survivors gained > or =5% of their at-diagnosis body weight during the first 6 months after diagnosis, while 37% and 33% of women

Body weight predicts denosumab pharmacodynamic duration: a real-world longitudinal cohort study.

UNLABELLED: Denosumab is administered as a fixed 60-mg subcutaneous injection every 6 months, regardless of body weight. In a retrospective cohort of 2123 patients with osteoporosis, the GAM-predicted duration of CTX reduction after each injection was about two months shorter at 80 kg than at 40 kg, but still exceeded the 6-month interval across the evaluated weight range. These findings support body-weight-aware CTX monitoring in heavier patients, not a change to the approved dosing interval. PURPOSE: Denosumab is administered as a fixed 60-mg subcutaneous injection every 6 months, but the d

Different impact of sleeve gastrectomy and Roux-en-Y gastric bypass on thyroid function and levothyroxine requirements.

PURPOSE: Bariatric surgery might impact the thyroid function test and, in hypothyroid patients, the dose of levothyroxine (LT4), but data are not univocal. We evaluated changes in thyroid function during the first year following bariatric surgery in patients without pre-existing thyroid disease, as well as adjustments in LT4 dosage in those with thyroid disorders, comparing the effects of Roux-en-Y gastric bypass (RYGB) and sleeve gastrectomy (SG). METHODS: A retrospective observational study including 69 patients on LT4 therapy (Tr) and 85 patients with normal thyroid function (Nt) at pre-su

Urine Output Calculated Using Actual Body Weight May Result in Overestimation of Acute Kidney Injury for Obese Patients.

GOAL: The derived hourly urine output (UO) indexed by body weight is one of the major criteria for the diagnosis of acute kidney injury (AKI). However, it is unclear whether actual body weight (ABW) or ideal body weight (IBW) should be used. This study aims to explore whether UO calculation based on ABW might lead to overestimation of AKI. METHOD: AKI patients identified in the Medical Information Mart for Intensive Care III database by different components of the Kidney Disease Improving Global Outcomes guidelines and different definitions of body weight were retrospectively studied. Effect of spontaneous breathing on umbilical venous blood flow and placental transfusion during delayed cord clamping in preterm lambs.

INTRODUCTION: During delayed umbilical cord clamping, the factors underpinning placental transfusion remain unknown. We hypothesised that reductions in thoracic pressure during inspiration would enhance placental transfusion in spontaneously breathing preterm lambs. OBJECTIVE: Investigate the effect of spontaneous breathing on umbilical venous flow and body weight in preterm lambs. METHODS: Pregnant sheep were instrumented at 132-133 days gestational age to measure fetal common umbilical venous, pulmonary and cerebral blood flows as well as arterial and intrapleural (IP) pressures. Fetotoxic response of technical quinalphos in rats.

Technical Quinalphos was administered po to pregnant rats through day 6-20 of gestation at doses of 0.5 or 1.5 mg/kg body weight. Quinalphos produced enzymatic changes in liver and serum of dams associated with mild pathomorphological changes in liver and brain. Fetotoxic effects were not observed at the tested dose levels as determined by total number of implantations, percentage resorption, live fetuses, crown rump length and fetal weight. A few insignificant skeletal deformities were noticed. A significant inhibition of acetylcholinesterase activity in fetal brain and placenta indicated pos

When the body changes faster than the brain: a narrative and theoretical review of body schema updating after bariatric surgery.

As indicated by the guidelines of the National Institutes of Health (NIH) and the Italian Society of Obesity Surgery and Metabolic Diseases (SICOB), bariatric surgery is the treatment of choice for severe obesity due to its effectiveness in achieving long-term weight loss and improving associated comorbidities. Many individuals with obesity exhibit profound body dissatisfaction, which often represents a primary motivation for seeking bariatric surgery, sometimes outweighing health-related reasons. Following surgery, a significant reduction in body weight is typically observed; however, perceiv

Efficacy of opioid antagonist in patients with binge eating behavior: A systemic review and meta-analysis.

BACKGROUND: Binge eating (BE) involves consuming unusually large amounts of food within a short period and feeling a loss of control. Neurobiological mechanisms underlying BE involve dysregulation of reward and inhibitory control systems, with the endogenous opioid system playing a key role. Opioid antagonists and the combination therapies with bupropion may have potential to reduce BE by affecting the reward system, but evidence regarding their effectiveness remains inconclusive. This meta-analysis reviewed randomized controlled trials (RCTs) exploring the effects of opioid antagonists on BE

Effects of pramlintide on energy intake and food preference in rats given a choice diet.

Amylin is a peptide hormone involved in the control of energy balance, making the amylin system a potential target for pharmacotherapies to treat obesity. Pramlintide, an amylin analogue, is an FDA-approved medication for the treatment of diabetes that also has food intake- and body weight-suppressive effects. However, it is unknown whether pramlintide may preferentially reduce intake of highly palatable, energy dense food, the overconsumption of which is thought to play a role in the etiology of obesity. Here, we investigate the effects of pramlintide on food intake and body weight in rats gi
