N-type voltage-dependent calcium channels mediate the nicotinic enhancement of GABA release in chick brain.

The role of voltage-dependent calcium channels (VDCCs) in the nicotinic acetylcholine receptor (nAChR)-mediated enhancement of spontaneous GABAergic inhibitory postsynaptic currents (IPSCs) was investigated in chick brain slices. Whole cell recordings of neurons in the lateral spiriform (SpL) and ventral lateral geniculate (LGNv) nuclei showed that cadmium chloride (CdCl2) blocked the carbachol-induced increase of spontaneous GABAergic IPSCs, indicating that VDCCs might be involved. To conclusively show a role for VDCCs, the presynaptic effect of carbachol on SpL and LGNv neurons was examined …

Control of glutamate release by calcium channels and kappa-opioid receptors in rodent and primate striatum.

The modulation of depolarization (4-aminopyridine, 2 mM)-evoked endogenous glutamate release by kappa-opioid receptor activation and blockade of voltage-dependent Ca2+ -channels has been investigated in synaptosomes prepared from rat and marmoset striatum. 4-Aminopyridine (4-AP)-stimulated, Ca2+ -dependent glutamate release was inhibited by enadoline, a selective kappa-opioid receptor agonist, in a concentration-dependent and norbinaltorphimine (nor-BNI, selective kappa-opioid receptor antagonist)-sensitive manner in rat (IC50 = 4.4+/-0.4 microM) and marmoset (IC50 = 2.9+/-0.7 microM) striatal

Involvement of dihydropyridine-sensitive calcium channels in human dendritic cell function. Competition by HIV-1 Tat.

The entry of extracellular calcium in leukocytes mediates several cellular processes; however, unlike in excitable tissues, the underlying molecular mechanisms are poorly defined. In this paper we provide phenotypical and biochemical evidence that peripheral blood-derived human dendritic cells express dihydropyridine-sensitive calcium channels. Exposure to the dihydropyridine drug nifedipine, which binds L-type calcium channels blocking calcium influx, prevents two dendritic cell functions that are dependent on extracellular calcium entry: apoptotic body engulfment and interleukin-12 productio…

Properties of single calcium-activated potassium channels of large conductance in rat hippocampal neurons in culture.

Patch-clamp recordings were made on rat hippocampal neurons maintained in culture. In cell-attached and excised inside-out and outside-out patches a large single-channel current was observed. This channel had a conductance of 220 and 100 pS in 140 mM [K+]i/140 mM [K+]o and 140 mM [K+]i/3 mM [K+]o respectively. From the reversal potential the channel was highly selective for K+, the PK+/PNa+ ratio being 50/1. Channel activity was voltage-dependent, the open probability at 100 nM [Ca2+]i increasing by e-fold for a 22 mV depolarization. It was also dependent on [Ca2+]i at both resting and depolar

Halothane effects on human malignant hyperthermia skeletal muscle single calcium-release channels in planar lipid bilayers.

Malignant hyperthermia (MH) may be life-threatening when genetically predisposed individuals are administered triggering anesthetic agents that are believed to produce intracellular calcium release. To test this theory, the effects of halothane on normal and MH human skeletal muscle calcium-release channels were studied. Single calcium-release channels were incorporated from isolated sarcoplasmic reticulum membrane vesicles into a planar lipid bilayer, and halothane effects on the conductance and gating properties were measured by electrophysiologic techniques. Among the subjects studied, seve

Voltage-operated calcium channels in small cell lung carcinoma cell lines: pharmacological, functional, and immunological properties.

Different subtypes of voltage-operated calcium channels (VOCCs) are expressed in different tissues and can be distinguished by functional and pharmacological criteria. One type of high voltage-activated calcium channel, specifically recognized by the peptide neurotoxin omega-conotoxin (omega CTx), is expressed only in neurons. Seven different human small cell lung carcinoma (SCC) cell lines were also found to bind 125I-omega CTx. The binding was specific, saturable, and of high affinity. 125I-omega CTx binding was not antagonized by the calcium channel ligands verapamil, nitrendipine, and dilt…

Calcium translocation during activation of alpha 1-adrenoceptor and voltage-operated channels in smooth muscle cells.

Changes in potential and mechanical activity were measured with the sucrose-gap method and changes in intracellular free calcium were determined from the fluorescence intensity of the calcium-quin 2 complex at 37 degrees C in smooth muscle cells of guinea-pig taenia caecum following alpha 1-adrenoceptor stimulation. The muscle cells responded with a transient hyperpolarization and a transient increase in intracellular free calcium on alpha 1-adrenoceptor stimulation in the absence of extracellular calcium. Sustained hyperpolarization, cessation of spike activity, relaxation and an increase in

Inhibition of calcium channels by harmaline and other harmala alkaloids in vascular and intestinal smooth muscles.

Effects of harmaline and other harmala alkaloids on the contractions induced in the vascular smooth muscle of rabbit aorta and intestinal smooth muscle of taenia isolated from guinea-pig caecum were examined. In rabbit isolated aorta, harmaline inhibited the sustained contraction induced by 65.4 mM K+ with an IC50 (concentration needed for 50% inhibition) of 4.6 X 10(-5) M. This inhibitory effect on high K+-induced contraction was antagonized by raising the concentration of external Ca2+ but not by Bay K 8644, a Ca2+ channel facilitator. Harmaline also inhibited the sustained contraction induc

Prostanoid synthesis by the rat urinary bladder: evidence for stimulation through muscarine receptor-linked calcium channels.

An in vitro model for the study of muscarine receptor-mediated synthesis of prostacyclin (PGI2) and other prostanoids (PGE2 and PGF2 alpha) by the rat urinary bladder is described. PGI2 synthesis was stimulated by parasympathomimetic agents (carbachol greater than methacholine greater than arecoline; McNA 343, nicotine and dimethyl phenyl piperazinium were without effect). Methacholine (3 X 10(-6) mol X l-1)-stimulated PGI2 synthesis was inhibited by muscarinic antagonists (atropine greater than ipratroprium bromide greater than gallamine greater than pirenzepine) and was completely abolished

Calcium channel blocker-associated pulmonary toxicity: epidemiology, mechanisms, and clinical management.

Calcium channel blockers are first-line medications for the treatment of hypertension, angina pectoris, and arrhythmias, and are generally well-tolerated. However, in recent years, there have been reports indicating that CCBs may cause severe pulmonary toxicity, such as non-cardiogenic pulmonary edema and acute respiratory distress syndrome. This article employs a comprehensive review methodology to systematically summarize the clinical types, incidence, risk factors, mechanisms of injury, and management strategies for CCB-related pulmonary toxicity. Current evidence indicates that pulmonary t

Gating effects of a Cav2.3 calcium channel variant linked to developmental and epileptic encephalopathy.

Developmental and epileptic encephalopathies (DEEs) are a group of neurological disorders that primarily affect young children and are characterized by severe seizures. DEEs are challenging to manage, with some patients experiencing severe side effects or not responding to frontline therapies. The CACNA1E gene, which encodes the voltage-gated calcium channel Cav2.3 (R-type), has recently been associated with DEEs. More than fifteen different variants in CACNA1E have been identified in patients with DEEs; however, the mechanisms by which these variants affect channel function and, thus, their r

Cardiovascular effects of Matricaria chamomilla extract: calcium channel modulation and vasorelaxant activity.

This study aimed to investigate the vasorelaxant potential of M. chamomilla extract and its modulatory effects on calcium ion channels. In vitro experiments assessed the extract’s impact on voltage-gated and GPCR-mediated calcium channels in aortic preparations. In vivo, the Tail Cuff method evaluated blood pressure-lowering effects in adrenaline-induced hypertensive rats. Phytochemical profiling was performed via GC-MS, and molecular docking assessed interactions of key compounds with vascular regulation targets (7VFS, 8THK, 3NOS). In vitro, 5 µg/ mL of the extract slightly increased aortic co…

High-dose insulin euglycaemic therapy for a severe calcium channel blocker overdose.

Calcium channel blockers are a common cause of poisoning and are associated with high mortality in affected patients. Treatment is complex. Although poorly researched, high-dose insulin euglycaemic therapy (HIET) appears to be an effective treatment. In this case report, we review a patient with a severe calcium channel blocker overdose who was successfully treated with HIET.

Dual Case Report: Symptomatic Myocardial Bridging Managed with Calcium Channel Blockers.

BACKGROUND: Myocardial bridging (MB) is a congenital coronary artery anomaly in which a segment of an epicardial coronary artery, most commonly the left anterior descending (LAD) artery, courses intramyocardially, and undergoes systolic compression. Although historically regarded as a benign anatomical variant, myocardial bridging has increasingly been recognized as a potential cause of myocardial ischemia, exertional chest pain, and other cardiac symptoms, particularly in patients without obstructive coronary artery disease. Awareness of this condition is important, as its diagnosis and manag

Revisiting the Association Between Calcium Channel Blockers and Parkinson's Disease in the E3N Cohort.

BACKGROUND: Previous studies on calcium channel blockers (CCBs) and Parkinson's disease (PD) risk reached conflicting conclusions. OBJECTIVES: We examined the relationship between CCBs and PD in the E3N cohort of French women followed for 15 years (2004-2018), while taking into account the potential for reverse causation. METHODS: New users of CCBs were identified using drug reimbursement databases. We validated incident PD using several data sources. We described trajectories of CCBs use using mixed logistic models within a nested case-control study. We used Cox proportional hazards models
