Rat myocardial tissue lipids and their effect on ventricular electrical activity: influence on dietary lipids.

OBJECTIVE: The aim was to investigate the effect of diets with different lipid content on rat myocardial tissue lipid composition and their possible influence on myocardial electrical activity. METHODS: 60 male Sprague-Dawley rats were randomised in three dietary feeding groups. Half the animals were used for the myocardial lipid study and the other half for the ventricular refractory period and ventricular conduction velocity measurements. Synthetic diets of low fat, high fat (predominantly lard fat), and high fat plus marine oil, the last two with cholesterol, were supplied ad libitum for f

Syphilis serology and human immunodeficiency virus positivity in Chandigarh.

AIDS and other sexually transmitted diseases are interlinked. VDRL positivity may indicate that the individual has an increased risk of being HIV positive as the epidemiological risk factors for developing syphilis and AIDS are similar. We analysed 323 (5.8%) VDRL positive serum samples (out of 5592 screened) for HIV positivity. All were HIV negative. While syphilitic infection in India at present is not commonly associated with HIV infection, experience in other countries indicates caution.

Coronary artery vasculitis and myocardial infarction associated with antiphospholipid antibodies in a pregnant woman.

A 28-year-old, 16 week primigravida presented with an acute anteroseptal myocardial infarction and a past history of recurrent venous thromboembolism and primary infertility. Although she lacked other clinical features of systemic lupus erythematosus, she had a circulating 'lupus' anticoagulant, anticardiolipin antibodies, a weakly positive anti-nuclear antibody and thrombocytopenia. She died suddenly despite corticosteroid therapy and autopsy revealed coronary arteritis and thrombosis.

Optic neuritis and myelopathy in systemic lupus erythematosus.

The optic neuritis of systemic lupus erythematosus (S.L.E.) more frequently results in the persistence of a central scotoma or complete blindness after a single attack than demyelinating optic neuritis, although the initial clinical presentations may be identical. A significant number of patients, however, recover normal vision. Optic neuritis may be the presenting symptom of S.L.E. and as myelopathy may also occur in the course of the disease, confusion with multiple sclerosis may result, especially if there are no arthritic, cutaneous nor visceral manifestations. We report a case of lupus op

Serologic response to VDRL in infants with congenital syphilis: ceftriaxone vs. penicillin.

OBJECTIVE: Penicillin remains the only safe and effective drug recommended for treating syphilis in pregnant women and congenital syphilis (CS). Nevertheless, alternative therapies are needed when penicillin is contraindicated, such as in allergic reactions, or unavailable due to supply limitations. This study aimed to compare the serological response to the Venereal Disease Research Laboratory (VDRL) test in infants reported with CS and treated with either ceftriaxone or penicillin. METHOD: A non-competitive cohort study was conducted in three public maternity hospitals in Fortaleza, Ceará,

Antiphospholipid antibodies in the aetiology of ischaemic optic neuropathy.

The involvement of antiphospholipid antibodies in the mediation of acute anterior ischaemic optic neuropathy (AION) was studied, in the light of recent associations between these autoantibodies and other vascular events. A strong association between IgG anticardiolipin antibodies and AION secondary to biopsy proven giant cell arteritis was identified, which did not exist with the 'non-arteritic', biopsy negative form of AION. The possible implications of this finding on the aetiology, diagnosis, and treatment of giant cell arteritis are discussed.

Antiphospholipid antibodies: a risk factor for occlusive ocular vascular disease in systemic lupus erythematosus and the 'primary' antiphospholipid syndrome.

Seven cases of occlusive ocular vascular disease affecting retinal and choroidal vessels were found among 84 consecutive patients with raised levels of anticardiolipin antibodies attending the lupus arthritis clinic at St Thomas's Hospital from 1985 to 1987. Six patients with systemic lupus erythematosus (SLE) and one with a 'primary antiphospholipid syndrome' had occlusive ocular vascular disease affecting a variety of vessels. This gives a prevalence of occlusive ocular vascular disease of 8% in this subgroup of patients, significantly higher than the 0.5-2.0% previously reported in patients

Mitochondrial Quality Control in Degenerative Musculoskeletal Diseases: Signaling Networks and Therapeutic Landscapes.

Degenerative musculoskeletal disorders, including osteoarthritis (OA), intervertebral disc degeneration (IVDD), and osteoporosis (OP), are increasingly viewed as disorders of mitochondrial quality control (MQC) rather than passive mechanical wear. The MQC network, comprising mitochondrial dynamics, mitophagy-biogenesis coupling, and inter-organelle communication, organizes the aging-inflammation-metabolism axis. Its collapse releases mtDNA fragments that act as damage-associated molecular patterns (DAMPs) activating innate immunity via cGAS-STING, and metabolic intermediates that remodel the e

Functionalized Ionic Liquids at Bacterial Membrane Interfaces: A Multiscale Perspective from Tensiometry, Microscopy, and Molecular Dynamics.

Recently developed ionic liquids (ILs) are gaining attention for their antibacterial properties in the face of increasing resistance to conventional antibiotics. Their structural tunability enables the design of compounds that balance antimicrobial potency and acceptable toxicity. We investigated two monoterpene-based functionalized ionic liquids (FILs)─[(1R,2S,5R)-(-)-mentoxymethyl]dimethyltetradecylammonium chloride and [(1R,2S,5R)-(-)-mentoxymethyl]tetradecylimidazolium chloride─which differ in cationic core (ammonium vs imidazolium) but share an identical hydrophobic alkyl chain. FtsK with a unique N-terminal extension is involved in coordinating the final steps of chromosome segregation with asymmetric division in mycobacterial cells.

In this study, we identify and functionally characterize a previously unrecognized, conserved N-terminal extension of mycobacterial FtsK (nFtsK) that contributes to the coordination of chromosome segregation with asymmetric cell division. Although FtsK is broadly conserved in bacteria as a late-stage DNA translocase and divisome component, mycobacterial FtsK uniquely contains a long, positively charged, intrinsically disordered N-terminal region. We show that this extension is dispensable for core FtsK functions-including septal localization, DNA translocation, and completion of cell division-

Hydrogen gas with hypothermic machine perfusion induces lipidomic alterations in donation-after-cardiac-death rat livers.

Hypothermic machine perfusion (HMP) combined with hydrogen gas has previously been shown to mitigate ischemia-reperfusion injury (IRI) in rat liver, but without full recovery of function. This study examined how hydrogen gas modulates hepatic lipid metabolism during HMP using donation after cardiac death (DCD) rat livers. Untargeted liquid chromatography-mass spectrometry was employed to perform comprehensive lipidomic profiling of liver samples. The analysis results revealed distinct lipid metabolic alterations across cold storage, machine perfusion (MP), and hydrogen-supplemented perfusion (

Lipidomic Remodeling by HILPDA in Granulosa Cells Impairs Ovarian Reserve Through Disruption of Mitochondrial and Endoplasmic Reticulum Homeostasis.

Diminished ovarian reserve (DOR) is characterized by a decline in oocyte quantity and/or quality and represents a growing cause of female infertility, particularly among young women, yet its pathogenesis remains incompletely understood. Disordered lipid metabolism impairs folliculogenesis and steroidogenesis, thereby contributing to the pathological changes of DOR. Here, we identify hypoxia-inducible lipid droplet-associated protein (HILPDA), a regulator of lipid droplet biogenesis, as a potential contributor to DOR. We found that HILPDA was significantly upregulated in granulosa cells (GCs) f

Case Report: Deletion in the 5' untranslated region of TAFAZZIN in a boy with Barth syndrome.

BACKGROUND: Barth syndrome is an X-linked disorder characterised by cardiomyopathy, growth abnormalities, neutropenia, and 3-methylglutaconic aciduria. It is caused by pathogenic variants in TAFAZZIN, which encodes a mitochondrial protein essential for cardiolipin remodelling. In this study, we describe the case of a patient with Barth syndrome in whom initial research genetic testing missed a 5' untranslated region deletion in TAFAZZIN that was later identified through a phenotype-guided reanalysis of exome sequencing data. CASE PRESENTATION: A male infant presented with dilated cardiomyopat

Loss of STARD7 Impairs Mitochondrial Phospholipid Homeostasis and Contributes to Mitochondrial Myopathy.

Mitochondria are composed of phospholipid bilayers rich in phosphatidylcholine (PC). StAR-related lipid transfer domain-containing protein 7 (STARD7) functions as a lipid transfer protein that plays a crucial role in maintaining mitochondrial PC homeostasis. In this study, we investigated the physiological role of STARD7 in skeletal muscle using muscle-specific knockout (mKO) mice. STARD7 expression was markedly higher in the soleus, a mitochondria-dense slow-twitch muscle, compared with fast-twitch fibers. Although muscle fibers from mKO mice exhibited no apparent structural abnormalities, th

Lysyl-Phosphatidylglycerol: A Lipid Involved in the Resistance of Staphylococcus aureus to Antimicrobial Peptide Activity.

Lysyl-phosphatidylglycerol (lysyl-PG) is one of the major lipids found in bacterial membranes; it is synthesized by attaching lysine to the headgroup of phosphatidylglycerol. First identified in Staphylococcus aureus in 1964, lysyl-PG is now recognized as a virulence factor that protects Staphylococcus aureus from antimicrobial agents, such as cationic antimicrobial peptides and phospholipase A2 type IIA. Under normal growth conditions, Staphylococcus aureus membranes are negatively charged due to a high proportion of anionic lipids, such as phosphatidylglycerol and cardiolipin. This intrinsic

Metabolic profiling of therapy-induced senescent cancer cells via TPEF, MALDI-MS, and RNA-sequencing.

Despite advances in cancer therapies, treatment failure from resistance and recurrence remains a major clinical challenge. Therapy-induced senescence (TIS), a state of stable cell cycle arrest with sustained metabolic activity, has emerged as a driver of inflammation, tumor persistence, and relapse. However, the heterogeneity of TIS complicates its detection and targeting. Here, we applied a multi-modal strategy to characterize metabolic alterations in senescent cancer cells induced by doxorubicin or γ-irradiation across three tumor cell lines: MCF7, HeLa, and TPC-1. Mitochondrial dysfunction

Anti-alcoholism drug disulfiram inhibits PANoptosis by blocking mitochondrial permeabilization in macrophages.

INTRODUCTION: PANoptosis is a form of inflammatory cell death that exhibits simultaneous activation of pyroptosis, apoptosis and necroptosis signaling. Disulfiram is a clinically used anti-alcoholism drug and can inhibit NLRP3 inflammasome activation and pyroptosis. However, it is unknown whether and how disulfiram interferes with PANoptosis and related inflammatory diseases. METHODS: PANoptosis was induced in murine macrophages and related protein levels were assayed by immunoblotting. The effects of disulfiram on PANoptosis were assessed both in macrophages in vitro and in a mouse model of

Onion-Mitochondria Inhibit Lipopolysaccharide-Induced Acute Lung Injury by Shaping Lung Macrophage Mitochondrial Function.

Mitochondrial dysfunction contributes to various inflammatory-related diseases by triggering the release of inflammatory molecules. Targeting mitochondrial dysfunction is emerging as a promising avenue for treating inflammatory diseases. Here, it is demonstrated that dietary plant-derived mitochondria (P-Mit) are capable of rescuing the lung macrophage mitochondrial (M-Mit) dysfunction in lipopolysaccharide (LPS)-induced mouse acute lung injury (ALI). Specifically, oral administration of dietary onion-derived mitochondria (O-Mit) can travel from the gut to the lungs in ALI mice, where preferen

Pretransplant natural antibody levels identify a subset of deceased donor kidney transplant recipients that benefit from infliximab induction.

Targeting peritransplant inflammation via tumor necrosis factor alpha blockade failed to improve kidney transplantation outcomes in the CTOT-19 trial that tested infliximab (IFX) induction. As natural antibodies (nAbs) to cardiolipin and phosphatidylethanolamine promote graft injury, we hypothesized that CTOT-19 outcomes were confounded by nAb levels. Pretransplant plasma anticardiolipin (aCL) and antiphosphatidylethanolamine (aPE) immunoglobulin M/immunoglobulin G (IgM/IgG) were measured in 177 CTOT-19 subjects and analyzed in relation to delayed graft function (DGF), 2-year estimated glomeru
