Mitochondrial myopathy in Senna occidentalis-seed-fed chicken.

Plants of the genus Senna (formerly Cassia) have been recognized as the cause of a natural and experimental syndrome of muscle degeneration frequently leading to death in animals. Histologically, it demonstrated skeletal and cardiac muscle necrosis, with floccular degeneration and proliferation of sarcolemmal nuclei. Recently, it was described as an experimental model of mitochondrial myopathy in hens chronically treated with Senna occidentalis. Currently, skeletal muscles of chicks intoxicated with seeds of the poisonous plant S. occidentalis were studied by histochemistry and electron micros

Crosslinking of telokin to chicken gizzard smooth muscle myosin.

Telokin, which is a candidate for one of the factors stabilizing dephosphorylated myosin filaments in smooth muscle cells, was subjected to a chemical crosslinking study to determine its binding location on the myosin molecule. Telokin labeled with 5-iodoacetamidofluorescein (Fl-telokin) retained the nature of unlabeled telokin: it bound to dephosphorylated gizzard myosin with a stoichiometry of 1 mol/mol myosin and induced filament assembly. The carboxyl groups of Fl-telokin were activated with 1-ethyl-3-(3-dimethyl-amino-propyl) carbodiimide and N-hydroxysuccinimide and then crosslinked to m

The apical caecal diverticulum of the chicken identified as a lymphoid organ.

A peculiar structure, observed as a dome-like protrusion at the apex of the caecum, was investigated macroscopically and histologically in healthy White Leghorn chickens. It was hemispheric or spherical in shape and as it consisted of a lumen with a wall occupied by lymphoid tissue, this structure was designated the apical caecal diverticulum (ACD). ACD were detected in 25.2% of examined chickens and had a mean diameter and height of 1.9 mm and 1.2 mm respectively. Histologically, both the lamina propria mucosae and the submucosa of ACD consisted of well developed aggregated lymphoid nodules. …

Molecular cloning and genomic organization of chicken syndecan-4.

We have cloned and determined the genomic organization of the core protein of the chicken transmembrane proteoglycan, syndecan-4. Identification of the initial cDNA was accomplished using polyclonal antibodies directed against the cytoplasmic domain of murine syndecan-1 core protein. The cDNA for chicken syndecan-4 encodes a putative core protein of 197 amino acids which consists of a 19-amino acid signal peptide, a 125-amino acid ectodomain, a 25-amino acid transmembrane domain, and a 28-amino acid cytoplasmic domain. The predicted molecular mass of the mature core protein is 19,639 daltons.

Indirect excitatory actions of histamine on isolated chicken oesophagus.

The histamine-induced contraction on chicken oesophagus was antagonised non-competitively by atropine, hexamethonium, cocaine, methysergide, indomethacin, theophylline and verapamil. Physostigmine slightly potentiated the excitatory action of histamine. These results indicate that histamine excreted its excitatory action by involving a number of mechanisms as suggested in guinea pig oesophagus.

Effect of acetazolamide on calcium accumulation by the developing chicken embryo.

The injection of 10 mg of acetazolamide into fertile chicken eggs on the 16th or 17th day of incubation resulted in a decreased accumulation of calcium by the developing embryo. A dose of 1 mg of acetazolamide did not affect embryonic calcium content. Although the eggshell serves as the primary source of calcium for the embryo, acetazolamide did not influence eggshell mobilization as measured by the release of 45Ca from labeled eggshells. Acetazolamide did decrease bone formation resulting in a statistically significant reduction in tibia dry weight and calcium content.

The toxicopathology of Prudhoe Bay crude oil in chicken embryos.

Microliter amounts of Prudhoe Bay crude oil were applied to the shell of fertile leghorn chicken eggs on Day 9 of incubation. Gross and microscopic pathological changes were examined in embryos surviving 4 days after exposure. Gross lesions seen consistently were extensive edema, superficial zones of pale hepatic tissue, distension of the heart, and enlargement of the spleen. Histologically, the pale liver tissue corresponded to areas of hepatocyte necrosis. Additional histological lesions were cellular casts and mineralization in the renal tubules, distension of the glomerular capillaries, an

Radioligand binding characteristics of the chicken cardiac muscarinic receptor.

The muscarinic receptor present in chicken cardiac membranes was characterised using a ligand binding approach and compared to the M1, M2 and M3 receptors that can be identified in ligand binding studies at present. [3H]N-methylscopolamine and [3H]pirenzepine appeared to label the same population of muscarinic receptors in chicken cardiac membranes since the density of sites labeled by the two radioligands was similar. Furthermore, affinity estimates of 8 muscarinic receptor antagonists for chicken cardiac muscarinic receptors were the same irrespective of whether [3H]N-methylscopolamine or [3

Inherited changes in concentrations of retinal and serum melatonin in the chicken.

Concentrations of melatonin in the retina, serum, and pineal gland were studied in genetically blind chicks carrying an autosomal recessive mutation, rc, characterized by the degeneration of photoreceptors in the retina after hatching. Blind homozygous (rc/rc) and sighted heterozygous (Rc+/rc) chicks were housed under 12:12 light:dark cycles. They were decapitated at 4, 6, 8, and 10 weeks of age at midlight and middark. Retinas, pineal glands, and serum samples were collected, and the resultant tissue melatonin was extracted and determined by radioimmunoassay. Retinal and pineal melatonin were

Negative cooperativity of chicken ovotransferrin on Al(III)-binding.

Chicken ovotransferrin, an iron binding protein, has two metal binding sites (amino (N) and carboxy (C) terminal sites). It binds Cu(II), Al(III), Co(II), and other metals, as well as Fe(III). In this study, the selectivity and cooperativity of the N and C sites on Al(III), Co(II), and Tb(III) binding were investigated. Metals were classified into two groups according to their site preference. Co(II) and Al(III) bound to the N site more preferably than to the C site, whereas Tb(III) bound to the C site more preferably. On Fe(III) binding, the binding constant of Fe(III) becomes larger when the

Muscarinic and nicotinic synaptic activation of the developing chicken iris.

The development of the mechanical characteristics of contraction and the pharmacology of synaptic activation in chick iris and ciliary body were examined from embryonic day 9 through posthatching. The ciliary ganglion-target muscle system has proven to be a useful model for both in vivo and in vitro studies of neuron-target interactions; one such interaction is involved in neuronal cell death, which in the ciliary ganglion occurs from Stage (St) 34 to 40. To understand the mechanism by which cholinergic blocking agents prevent naturally occurring neuronal death in the chick ciliary ganglion (s

Effects of atriopeptin and chicken heart extract in Gallus domesticus.

Effects of Ser-Leu-Arg-Arg-atriopeptin III (ANP) and chicken heart extract (CHE) were compared during unilateral renal portal infusion in anesthetized Single Comb White Leghorn chickens. The purpose was to determine whether renal effects were glomerular and/or tubular. Both CHE and ANP caused substantial decreases in mean arterial pressure but had different renal actions. ANP caused small but significant increases in both absolute and fractional sodium excretion, but these effects were modest compared with those reported in mammals. Although there was a tendency for higher fractional sodium ex…

Meckel's diverticulum. II. A novel lymphoepithelial organ in the chicken.

We have studied the lymphoid development and structure of Meckel's diverticulum (MD). The lymphoid accumulation began about 2 weeks of age. Between 2 and 5 weeks of age the longitudinal folds were filled with lymphoid tissue. The intensive germinal center formation occurred between 5 and 7 weeks of age. Germinal center formation was associated with the presence of secretory cells. The absence of the secretory cells in the germinal centers was followed by germinal center inactivity which was indicated by the lack of lymphoblasts and the high number of tingible body macrophages. The lymphoid tis

The structural basis of the blood-aqueous barrier in the chicken eye.

In order to identify the structural basis of the blood aqueous barrier in the chicken eye, the morphology of the blood vessels and epitheliu … ior chamber is blocked by very complex zonulae occludentes between the nonpigmented cells. Thus, in chickens as in mammals tight junctions between the nonpigmented cells of the ciliary epithelium represent the structural equivalent of the blood-aqueous barrier.

The natural occurrence of Trypanozoon in domestic chicken in the Ivory Coast.

From a natural infected chicken (Gallus gallus var. domesticus) in the Ivory Coast trypanosomes were isolated using Mastomys natalensis as recipient animal. Trypanosomes were diagnosed as belonging to the subgenus Trypanozoon from its morphology and its infectivity for rodents. The stabilated stock was able to infect a laboratory chicken. The stock proved to be human plasma subresistant and showed electrophoretic patterns of three enzymes (ALAT III, ME I, PEP III) so far only seen in pig and dog originated Trypanozoon stocks from the same region surveyed. The discovery for the first time of ch
