Progress of genome-wide association studies of ankylosing spondylitis.

Ankylosing spondylitis (AS) is an immune-mediated arthritis which primarily affects the spine and sacroiliac joints. Significant progress has been made in discovery of genetic associations with AS by genome-wide association studies (GWAS) over past decade. These findings have uncovered novel pathways involved pathogenesis of the disease and have led to introduction of novel therapeutic treatments for AS. In this Review, we discuss the genetic variations associated with AS identified by GWAS, the major pathways revealed by these AS-associated variations and critical cell types involved in AS de

Potential crosstalk between sonic hedgehog-WNT signaling and neurovascular molecules: Implications for blood-brain barrier integrity in autism spectrum disorder.

Autism Spectrum Disorder (ASD) is a neurodevelopmental disease originating from combined genetic and environmental factors. Post-mortem human studies and some animal ASD models have shown brain neuroinflammation, oxidative stress, and changes in blood-brain barrier (BBB) integrity. However, the signaling pathways leading to these inflammatory findings and vascular alterations are currently unclear. The BBB plays a critical role in controlling brain homeostasis and immune response. Its dysfunction can result from developmental genetic abnormalities or neuroinflammatory processes. In this review

Metabolome alterations in severe critical illness and vitamin D status.

BACKGROUND: Metabolic homeostasis is substantially disrupted in critical illness. Given the pleiotropic effects of vitamin D, we hypothesized that metabolic profiles differ between critically ill patients relative to their vitamin D status. METHODS: We performed a metabolomics study on biorepository samples collected from a single academic medical center on 65 adults with systemic inflammatory response syndrome or sepsis treated in a 20-bed medical ICU between 2008 and 2010. To identify key metabolites and metabolic pathways related to vitamin D status in critical illness, we first generated

Disruption of cysteine metabolism leads to synthetic lethality and in vivo fitness impairment in Acinetobacter baumannii.

Acinetobacter baumannii is a critical nosocomial pathogen that causes infections in diverse host niches, including the respiratory tract, bloodstream, and urinary tract. Considering the contemporary landscape of antibiotic resistance, alongside targeting resistance determinants, there is a need to identify novel and alternative targets for therapeutic intervention. Central metabolism plays a major role in the success of a pathogen and represents a pool of plausible drug targets. However, metabolic networks are inherently complex and often employ redundant pathways to ensure the supply of essen

Signal transduction to hypoxia-inducible factor 1.

Hypoxia-inducible factor 1 (HIF-1) is a transcriptional activator that functions as a master regulator of O2 homeostasis. HIF-1 target genes encode proteins that increase O2 delivery and mediate adaptive responses to O2 deprivation. HIF-1 activity is regulated by the cellular O2 concentration and by the major growth factor-stimulated signal transduction pathways. In human cancer cells, both intratumoral hypoxia and genetic alterations affecting signal transduction pathways lead to increased HIF-1 activity, which promotes angiogenesis, metabolic adaptation, and other critical aspects of tumor p

Visceral artery syndromes and reconstructions.

Acute and chronic visceral ischemia are now well recognized clinical entities and extensive progress has been made in both the evaluation and treatment of patients with these lesions. The visceral vessels and the nature of their collateral pathways can also be of critical importance when dealing with other intra-abdominal pathology. Poor anastomotic healing, or the formation of strictures and intestinal fistulas may result when the mesenteric circulation is slowly compromised. Hence, it is imperative that the visceral arteries, their lesions, and their collateral pathways be understood by all

Integrated omics reveals the regulatory role of PKCα in Sertoli cell proliferation and apoptosis through the MAPK/ERK signaling pathway in goose testis.

Testicular development is essential for reproductive performance in geese, as the testes are the primary organs for sperm production and play a pivotal role in egg-laying physiology. Despite their importance, genes, proteins, and pathways regulating goose testicular development are poorly understood. This study employed integrative transcriptomic and proteomic analysis methods to identify critical regulators of testicular development in geese across three reproductive periods. Additionally, the role of PKCα in Sertoli cell proliferation via the MAPK/ERK pathway was evaluated at the cellular le

Innate immune signaling and functions in astrocytes.

Astrocytes, long considered supportive cells of the central nervous system (CNS), have critical roles in innate immunity. This Review explores immune signaling pathways in astrocytes, including pattern recognition through Toll-like receptors, nucleic acid sensors and inflammasomes. These pathways enable the detection of danger signals and initiate protective responses and endogenous innate immune functions. Downstream signaling pathways, including the interferon, NF-κB and STAT3 pathways, mediate astrocyte reactivity and drive cytokine secretion, antiviral responses, phagocytosis and many othe

V1-bypassing thalamo-cortical visual circuits in blindsight and developmental dyslexia.

Vision rests on computations that primarily rely on the parvocellular and magnocellular geniculate relay of retinal signals to V1. Secondary pathways involving superior colliculus, koniocellular lateral geniculate nucleus and pulvinar and their V1-bypassing projections to higher order cortex are known to exist. While they may form an evolutionary old visual system, their contribution to perception and visually guided behaviour remain largely obscure. Recent developments in tract tracing and circuit manipulation technologies provide new insights. Here we discuss how secondary visual pathways me

EEG monitoring during therapeutic hypothermia in neonates, children, and adults.

Therapeutic hypothermia is being utilized as a neuroprotective strategy in neonates, children, and adults. The most common indications are hypoxic ischemic encephalopathy in neonates and post cardiac arrest in adults. Electroencephalographic monitoring use is increasing in critical care units, and is sometimes a component of therapeutic hypothermia clinical pathways. Monitoring may detect non-convulsive seizures or non-convulsive status epilepticus, and it may provide prognostic information. We review data regarding indications for therapeutic hypothermia and electroencephalographic monitoring

TrkB Activation during a Critical Period Mimics the Protective Effects of Early Visual Experience on Perception and the Stability of Receptive Fields in Adult Superior Colliculus.

During a critical period in development, spontaneous and evoked retinal activity shape visual pathways in an adaptive fashion. Interestingly, spontaneous activity is sufficient for spatial refinement of visual receptive fields (RFs) in superior colliculus (SC) and visual cortex (V1), but early visual experience is necessary to maintain inhibitory synapses and stabilize RFs in adulthood (Carrasco et al., 2005, 2011; Carrasco and Pallas, 2006; Balmer and Pallas, 2015a). In V1, BDNF and its high-affinity receptor TrkB are important for development of visual acuity, inhibition, and regulation of t

Emerging Roles of Synapse Organizers in the Regulation of Critical Periods.

Experience remodels cortical connectivity during developmental windows called critical periods. Experience-dependent regulation of synaptic strength during these periods establishes circuit functions that are stabilized as critical period plasticity wanes. These processes have been extensively studied in the developing visual cortex, where critical period opening and closure are orchestrated by the assembly, maturation, and strengthening of distinct synapse types. The synaptic specificity of these processes points towards the involvement of distinct molecular pathways. Attractive candidates ar

Regulation of mast cell survival and function by tuberous sclerosis complex 1.

Mast cells play critical roles in allergic disorders and asthma. The importance of tuberous sclerosis complex 1/2-mammalian target of rapamycin (TSC1/2-mTOR) signaling in mast cells is unknown. Here, we report that TSC1 is a critical regulator for mTOR signaling in mast cells downstream of FcεRI and c-Kit, and differentially controls mast cell degranulation and cytokine production. TSC1-deficiency results in impaired mast cell degranulation, but enhanced cytokine production in vitro and in vivo after FcεRI engagement. Furthermore, TSC1 is critical for mast cell survival through multiple pathwa
