Decoding MicroRNA-Guided Antiviral Defense in Cucurbitaceae: Regulatory Networks, RNA Silencing Cross-Talk, and Emerging Strategies for Crop Resilience.

MicroRNAs (miRNAs) are central regulators of gene expression and play pivotal roles in plant antiviral defense. In Cucurbitaceae, a globally important crop family including cucumber, melon, and watermelon, viral pathogens such as CGMMV, CMV, and ZYMV represent major constraints on productivity. However, the regulatory complexity of miRNA-mediated antiviral responses in these species remains incompletely understood. This review provides an integrated overview of recent advances in miRNA-guided antiviral immunity in Cucurbitaceae, highlighting the dynamic reprogramming of small RNA pathways upon

Risk assessment of Panonychus ulmi to global temperate fruit production under climate change.

Climate change is reshaping pest distributions, threatening global fruit production. We present a climate-informed risk assessment for the European red mite, Panonychus ulmi, by integrating its habitat suitability predictions with host crop distributions. Our optimized model identified the mean temperature of the coldest quarter as the primary predictor, highlighting its critical role in P. ulmi overwintering survival. We found that 72.4% of temperate fruit crops are planted in highly suitable habitats for P. ulmi. By the 2070s, this at-risk area is projected to expand to 74.8% under the SSP5-

Harnessing next-generation molecular technologies for precision control of Tuta absoluta and Helicoverpa armigera in tomato.

Tomato (Solanum lycopersicum) is one of the most significant vegetable crops cultivated around the world. Among biotic stresses, production of tomato is severely affected by insect pests mainly by tomato fruit borer (Helicoverpa armigera) and tomato leaf miner (Tuta absoluta). Traditionally to manage the destruction caused by insect pests, farmers mostly used chemical insecticides and by different cultural practices. Use of chemical pesticides is of great concern as it is harmful for the environment and other living beings and with the passage of time, insects develop resistance against pestic

Nitric Oxide and Melatonin Cross Talk on Photosynthetic Machinery.

Nitric oxide (NO) and melatonin (MT) significantly influence photosynthetic processes by modulating redox homeostasis, chlorophyll content, stomatal conductance, and gene expression, particularly under abiotic stress conditions. This review summarizes the intricate crosstalk between NO and melatonin, focusing on their coordinated roles in regulating photosynthetic efficiency. Evidence from various plant species indicates that the application of exogenous NO and melatonin enhances chlorophyll content, photosystem efficiency (particularly PSII), and photosynthetic performance, mitigating stress-

Biochar and nano-hydroxyapatite combined remediation of soil surrounding tailings area: Multi-metal(loid)s fixation and soybean rhizosphere soil microbial improvement.

The soil near tailings areas is relatively barren and contaminated by multi-metal(loid)s, seriously threatening the safety of crop production. Here, biochar and nano-hydroxyapatite (nHAP) were combined to improve the sterilized and unsterilized polymetallic contaminated soil, and soil incubation and soybean pot experiments were designed. Results showed that biochar and nHAP not only increased soil C, N, and P but also effectively reduced multi-metal bioavailability, wherein the combined application of the two amendments had the best effect on metal immobilization. The synergistic effect of the…

Editing of ORF138 restores fertility of Ogura cytoplasmic male sterile broccoli via mitoTALENs.

Cytoplasmic male sterility (CMS), encoded by the mitochondrial open reading frames (ORFs), has long been used to economically produce crop hybrids. However, the utilization of CMS also hinders the exploitation of sterility and fertility variation in the absence of a restorer line, which in turn narrows the genetic background and reduces biodiversity. Here, we used a mitochondrial targeted transcription activator-like effector nuclease (mitoTALENs) to knock out ORF138 from the Ogura CMS broccoli hybrid. The knockout was confirmed by the amplification and re-sequencing read mapping to the mitoch

The analysis of reference genes expression stability in susceptible and resistant Apera spica-venti populations under herbicide treatment.

Weed resistance to herbicides constitutes a serious problem to world crop production. One of the weeds that are significantly threatening the crops' yield and quality is Apera spica-venti. The target-site resistance (TSR) mechanism of A. spica-venti has been widely studied, though, little is known about its non-target-site resistance (NTSR) mechanisms at the molecular level. Molecular examination of NTSR is, to a great extent, based on the expression profiles of selected genes, e.g. those participating in detoxification. However, to obtain reliable results of gene expression analysis, the use …

The Role of Gibberellins in Regulation of Nitrogen Uptake and Physiological Traits in Maize Responding to Nitrogen Availability.

Modified gibberellin (GA) signaling leads to semi-dwarfism with low nitrogen (N) use efficiency (NUE) in crops. An understanding of GA-mediated N uptake is essential for the development of crops with improved NUE. The function of GA in modulating N uptake capacity and nitrate (NO3-) transporters (NRTs) was analyzed in the GA synthesis-deficient mutant zmga3ox grown under low (LN) and sufficient (SN) N conditions. LN significantly suppressed the production of GA1, GA3, and GA4, and the zmga3ox plants showed more sensitivity in shoots as well as LN stress. Moreover, the higher anthocyanin accumu

Farming without Glyphosate?

Recent statements from scientific organisations and court decisions have resulted in widespread public interest and concern over the safety of glyphosate, the most popular and effective herbicide used worldwide. Consequently, glyphosate-based products are under intense scrutiny from governments at all levels. Some jurisdictions have already banned or restricted its use, which will adversely impact international trade in bulk grain commmodities if glyphosate residues are detected. The possibility of farming without glyphosate is becoming an important issue facing the agri-food research and deve

Effect of Soil Amendments on Cd Accumulation by Spinach from a Cd-Mineralized Soil.

Cadmium (Cd)-mineralized soils occur in many nations. When these soils are noncalcareous, crops and especially leafy vegetables such as lettuce ( L.) and spinach ( L.) may accumulate levels of Cd in their edible portions that exceed international standards. Vegetable crops grown on Lockwood loam from Monterey County, CA, absorb an excessive amount of Cd into their edible portions. Agronomic or genetic management alternatives are needed to allow the use of these otherwise highly productive soils for spinach. Previous research has shown that zinc (Zn) fertilizer plus limestone incorporation or b

Application of Helminth ova infection dose curve to estimate the risks associated with biosolid application on soil.

Helminth ova (HO) are the main biological concern when reusing sludge for agricultural production. Worldwide sludge regulations consider a permissible range of 0.25-1 HO/gTS. Such limits are unaffordable to most developing countries, due to high helminth ova content in sludge, and the lack of viable technology to inactivate them as needed. The quantitative microbial risk assessment (QMRA) is a useful tool to estimate the risk of treated sludge, considering feasible and viable limits. QMRA, however, has not been applied before for HO because no dose-infection curve was available. Therefore, the

Physiological characterisation of magnesium deficiency in sugar beet: acclimation to low magnesium differentially affects photosystems I and II.

Magnesium deficiency in plants is a widespread problem, affecting productivity and quality in agriculture, yet at a physiological level it has been poorly studied in crop plants. Here, a physiological characterization of Mg deficiency in Beta vulgaris L., an important crop model, is presented. The impact of Mg deficiency on plant growth, mineral profile and photosynthetic activity was studied. The aerial biomass of plants decreased after 24 days of hydroponic culture in Mg-free nutrient solution, whereas the root biomass was unaffected. Analysis of mineral profiles revealed that Mg decreased m

Soybean grain production and nutritional quality responses under elevated CO2, high temperature, and drought.

Soybean (Glycine max L. Merr.) is an important global crop that supplies protein and oil for the food and feed industries. However, its yield and nutritional quality are increasingly affected by climate change. This study combines experimental data with predictive modeling to assess how elevated CO2 (eCO2), high temperature, and drought (either alone or combined) influence soybean grain production and composition. Plants were grown under controlled conditions simulating future climate scenarios, and biochemical traits, including carbohydrates, proteins, lipids, and amino acids, were analyzed.

Efficient Cadmium-Tolerant Phosphate-Solubilizing Bacteria Loaded on Bone-Derived Biochar Effectively Alleviated Cadmium Pollution and Phosphorus Deficiency in Brassica napus L. Plantation.

Both cadmium (Cd) contamination and phosphorus deficiency pose major constraints to sustainable crop production. This study loaded highly efficient Cd-tolerant phosphate-solubilizing bacteria (CdTPSB) onto calcium- and phosphorus-rich pig bone biochar (PBB) to construct a functional composite (MPBB), which was subsequently applied in a Brassica napus L. pot experiment. The results revealed that the CdTPSB possessed strong capabilities in phosphate solubilization (383.67-520.19 mg L-1), Cd immobilization (64.67-66.67%), IAA production (113.24-114.27 μg mL-1), and siderophore activity (65.02-89.…

Nitrate reductase-dependent nitric oxide production mediates nitrate-conferred salt tolerance in rice seedlings.

Soil salinity is a destructive environmental factor that inhibits plant growth and crop yield. Applying nitrogen fertilizer is a practical method to enhance salt tolerance. However, the underlying mechanisms remain largely unknown. Here, we demonstrated that NO3--enhanced salt tolerance in rice (Oryza sativa L.) seedlings is mediated by nitrate reductase (NR)-dependent nitric oxide (NO) production. Seedlings grown in nitrate condition (N) exhibited much greater salt tolerance compared with those grown in ammonium nitrate and ammonium (A) conditions, a pattern also observed in the MADS-box tran
