Recombinant exosporium protein BclA of Bacillus anthracis is effective as a booster for mice primed with suboptimal amounts of protective antigen.

Bacillus collagen-like protein of anthracis (BclA) is an immunodominant glycoprotein located on the exosporium of Bacillus anthracis. We hypothesized that antibodies to this spore surface antigen are largely responsible for the augmented immunity to anthrax that has been reported for animals vaccinated with inactivated spores and protective antigen (PA) compared to vaccination with PA alone. To test this theory, we first evaluated the capacity of recombinant, histidine-tagged, nonglycosylated BclA (rBclA) given with adjuvant to protect A/J mice against 10 times the 50% lethal dose of Sterne st

No association between neutrophil FcgammaRIIa allelic polymorphism and anti-neutrophil cytoplasmic antibody (ANCA)-positive systemic vasculitis.

ANCA, implicated as having a pathogenic role in systemic vasculitis, can activate tumour necrosis factor-alpha (TNF-alpha)-primed neutrophils by cross-linking surface-expressed ANCA antigens with neutrophil FcgammaRIIa receptors to release reactive oxygen species. The FcgammaRIIa receptor exists as polymorphic variants, R131 and H131, which differ in their ability to ligate human IgG2 and IgG3. Neutrophils homozygous for the FcgammaRIIa-H131 allotype bind more efficiently to IgG3 than the FcgammaRIIa-R131 allotype and are the only human FcgammaR which bind IgG2. Our aim was to determine whethe

Autoimmunity after induction of neonatal tolerance to alloantigens: role of B cell chimerism and F1 donor B cell activation.

BALB/c mice rendered tolerant by the neonatal injection of semiallogeneic (C57BL/6 X BALB/c)F1 spleen cells develop features of autoimmune disease. The possible mechanisms involved in autoantibody production, particularly anti-DNA antibodies, were investigated. In the first 5 wk, there was polyclonal B cell activation, as indicated by marked hypergammaglobulinemia, with a predominance of IgG1 and an increased production of antihapten antibodies. IgG1 anti-SSDNA and anti-DSDNA antibodies were detected with similar kinetics, but at higher titers than the anti-hapten antibodies. Also, there was a

STING Activation in Dendritic Cells Enhances Antigen Presentation by Live Tumor Cells.

The stimulator of interferon genes (STING) pathway is a key regulator of antitumor immunity. Activation of STING by the natural agonist 2'3'-cyclic GMP-AMP (cGAMP) induces type I interferon and promotes CD8+ T cell responses against tumors. However, how type I interferon modulates tumor cell recognition by T cells remains unclear. Here, using a co-culture model and virus-like particles encapsulating cGAMP (cGAMP-VLP), we show that STING activation in dendritic cells enhances CD8+ T cell recognition of live tumor cells through type I interferon signaling. This effect is independent of enhanced

High-affinity interactions between peptides and heat shock protein 70 augment CD8+ T lymphocyte immune responses.

Exogenously delivered antigenic peptides complexed to heat shock proteins (HSPs) are able to enter the endogenous Ag-processing pathway and prime CD8+ CTL. It was determined previously that a hybrid peptide containing a MHC class I-binding epitope and HSP70-binding sequence Javelin (J0) in complex with HSP70 could induce cytotoxic T cell responses in vivo that were more robust than those induced by the minimal epitope complexed with HSP70. The present study introduces a novel, higher-affinity HSP70-binding sequence (J1) that significantly enhances binding of various antigenic peptides to HSP70

Safety and immunogenicity of a multivalent HIV vaccine comprising envelope protein with either DNA or NYVAC vectors (HVTN 096): a phase 1b, double-blind, placebo-controlled trial.

BACKGROUND: Up to now, immunisation regimens that have been assessed for development of HIV vaccines have included purified envelope (Env) protein among the boosting components of the regimen. We postulated that co-administration of Env protein with either a DNA or NYVAC vector during priming would result in early generation of antibody responses to the Env V1/V2 region, which are important markers for effective protection against infection. We aimed to assess the safety and immunogenicity of a multivalent HIV vaccine including either DNA or NYVAC vectors alone or in combination with Env glyco

Melampodium divaricatum (Rich.) DC. extracts exert anti-inflammatory effects via inhibition of the NLRP3 inflammasome.

ETHNOPHARMACOLOGICAL RELEVANCE: Melampodium divaricatum (Rich.) DC. (MelD), commonly known as butter daisy, is a medicinal plant widely distributed in Central and South America. In traditional medicine, particularly in Mexico and other parts of Central America, decoctions prepared from the aerial parts of MelD have been widely used to treat inflammation-associated ailments, including dysentery, fever, infections, and pain. Despite its long-standing ethnomedicinal use, the molecular and immunological mechanisms underlying its anti-inflammatory effects, particularly in the context of innate immu

Adjuvant-dependent protection of SARS-CoV-2 spike vaccines: comparative immunogenicity of human-applicable formulations.

UNLABELLED: Although recombinant vaccines with various adjuvant systems are widely deployed in global coronavirus disease 2019 immunization, their distinct immune profiles have not been fully elucidated. In this study, we evaluated immune responses induced by four clinically validated adjuvants-aluminum hydroxide (Al), two water-in-oil emulsions (Montanide ISA720 and ISA51), and an oil-in-water emulsion (Sepivac SWE)-formulated with the prefusion-stabilized ancestral severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike trimer (S-2P) in mouse models. Safety, effectiveness and immunogenicity of heterologous mRNA-1273 boost after prime with Ad26.COV2.S among healthcare workers in South Africa: The single-arm, open-label, phase 3 SHERPA study.

Limited studies have been conducted on the safety and effectiveness of heterologous COVID-19 vaccine boosting in lower income settings, especially those with high-HIV prevalence., The Sisonke Heterologous mRNA-1273 boost after prime with Ad26.COV2.S (SHERPA) trial evaluated a mRNA-1273 boost after Ad26.COV2.S priming in South Africa. SHERPA was a single-arm, open-label, phase 3 study nested in the Sisonke implementation trial of 500000 healthcare workers (HCWs). Sisonke participants were offered mRNA-1273 boosters between May and November 2022, when Omicron sub-lineages were circulating. Adver

A multi-epitope pan-betacoronavirus vaccine construct predicted to induce broad-spectrum and durable immune responses: an immunoinformatics approach.

INTRODUCTION: Recurrent zoonotic spillovers and continuous antigenic evolution among betacoronaviruses, including SARS-CoV, MERS-CoV, and SARS-CoV-2, highlight the urgent need for a broad-spectrum vaccine capable of eliciting cross-protective immunity. Conventional vaccines, although effective against specific strains, may be limited by antigenic mismatch and waning immunity. This study aimed to design a multi-epitope pan-betacoronavirus vaccine targeting conserved regions within the receptor-binding domain (RBD) using an integrated immunoinformatics and reverse vaccinology framework. Carbohydrate-to-fat ratio is positively associated with comprehensive frailty across protein-intake levels in older adults.

PURPOSE: Diet influences frailty, and protein intake has been a prime focus of prevention. However, frailty remains prevalent, even among older adults with relatively high protein intake, suggesting the influence of remaining energy distribution between carbohydrates and fats. We investigated the association between the carbohydrate-to-fat energy ratio (CFr) and comprehensive frailty in older Japanese adults and whether this association differed by protein intake. METHODS: Cross-sectional baseline data from 5679 community-dwelling adults aged ≥ 65 years in the Kyoto-Kameoka Study were analyze

Molecular Insights into the Conformational Dynamics of the Novel Dispersion Sensor Histidine Kinase DspS.

Dispersion sensor (DspS) is a membrane-bound histidine kinase in Pseudomonas aeruginosa that senses cis-2-decenoic acid (CDA), a fatty acid signal regulating biofilm dispersion, motility, and virulence. However, its structural mechanism of signal transduction remains poorly defined. Here, we combined AlphaFold-Multimer modeling, multiple-replica accelerated molecular dynamics simulations, and Markov state modeling (MSM) to investigate the conformational dynamics of full-length dimeric DspS. Simulations focused on the cyclases/histidine kinase-associated sensory extracellular domain and transme

Targeting gut-brain-immune axis in amyotrophic lateral sclerosis.

Amyotrophic lateral sclerosis (ALS) is a fatal motor neuron neurodegenerative disorder with a median survival of only 3-5 years. The heterogeneity of the disease and lack of effective therapies highlight the importance of identifying novel pathogenic mechanisms. We hypothesize that dysbiosis of gut microbiota enhances ALS by disrupting intestinal barrier function and altering metabolite profiles to drive systemic inflammation and neuronal stress. Precisely, the decrease in health-promoting bacteria (e.g., Akkermansia muciniphila, Bifidobacterium and Lactobacillus spp.) in ALS can reduce neurop

Environmental assessment of land-use transformation and peri-urban resource loss post-february 2023 earthquakes: a case study in Antakya, Türkiye.

Large-scale disasters fundamentally alter urban spatial structures, often triggering rapid and unsustainable land-use changes during the reconstruction phase. This study quantifies the spatial extent and nature of urban transformation in Antakya, Türkiye, following the devastating February 6, 2023, earthquakes, by analyzing Land Use/Land Cover (LULC) change between the pre-disaster (2022) and post-reconstruction (2025) periods. To achieve high-accuracy mapping in a spectrally complex post-disaster landscape, we implement an Object-Based Image Analysis (OBIA) framework coupled with a Support Ve

Periodontitis and Alzheimer's disease, common pathways in down syndrome.

Down Syndrome (DS) involves widespread systemic issues, including a near-universal development of early-onset Alzheimer's Disease, driven by factors beyond Amyloid Precursor Protein (APP) over-expression, such as chronic neuroinflammation and endosomal dysfunction. High prevalence of periodontitis in this population acts as a significant source of peripheral inflammation that may accelerate this cognitive decline, necessitating further study into the bidirectional, pro-inflammatory links between systemic health and neurodegeneration in DS. This review analyzes the common pathophysiological pat

Integrated genomic analysis of Salmonella Typhi from Pakistan: Antimicrobial resistance determinants, pangenome structure, and reverse vaccinology-driven vaccine target discovery.

Salmonella enterica serovar Typhi (S. Typhi) is the aetiologic agent of typhoid fever in humans. The burden of typhoid is highest in low- and middle-income countries such as Pakistan. Moreover, the increasing emergence of antibiotic-resistant and hypervirulent S. Typhi strains highlights the need for deeper genomic understanding as current treatment regimens become progressively less effective. Therefore, this study analyzed publicly available S. Typhi genomes from Pakistan to characterize genomic diversity, sequence types, plasmid content, and antimicrobial resistance and virulence profiles.

Soil microbial legacy mediated by buckwheat flavonoids enhances cabbage resistance to clubroot disease.

BACKGROUND: The legacy of plant growth significantly impacts the health of subsequent plants, yet the mechanisms by which soil legacies in crop rotation systems influence disease resistance through rhizosphere plant-microbiome interactions remain unclear. Using a buckwheat-cabbage rotation model, we investigated how microbial soil legacies shape cabbage growth and clubroot disease resistance. RESULTS: Three-year field trials revealed that buckwheat rotation sustainably reduced clubroot severity by 67%-97%, regardless of pathogen load. Soil sterilization eliminated this suppression, implicatin
