DdrR acts as a DNA damage-induced cell division inhibitor in Acinetobacter baumannii.

The genus Acinetobacter possesses non-canonical genes and responses to DNA damage. It encodes no LexA homolog to repress the ~150 SOS genes induced after exposure to various DNA-damaging agents, but two SOS regulators, UmuDAb and DdrR, repress both themselves and error-prone polymerases that generate mutations after DNA damage. Further differences include the lack of the Gram-negative cell division inhibitor (CDI), SulA, in members of the Moraxellaceae family to which Acinetobacter belongs. However, ddrR, which lacks homologs outside the Acinetobacter genus, is transcribed divergently from umu

Somatic mutation: a cause of sporadic neurodegenerative diseases?

The major causes of the common neurodegenerative diseases remain unknown. Alzheimer's disease, Parkinson's disease and motor neuron disease occur in both sporadic and familial forms, and mutations are progressively being found in families with these disorders. However, attempts to find causative mutations in blood DNA from the sporadic forms of the diseases have proved fruitless. It is hypothesised that this is because the causative mutations are found only in the cells in the central nervous system that are affected by the disease. These mutations arise in the developing embryo in progenitor

Pelizaeus-Merzbacher disease: cellular hypersensitivity to ultraviolet light.

We report two connatal cases of Pelizaeus-Merzbacher disease (PMD) with cellular ultraviolet (UV)-hypersensitivity. We studied the UV-sensitivity of cultured fibroblast cells derived from these PMD cases, as compared with UV-sensitive Cockayne syndrome (CS) and xeroderma pigmentosum (XP) cells as positive controls. The ability of the PMD cells to form colonies after UV irradiation was intermediate between those of CS cells and normal controls. There were no differences in both colony-forming ability after x-ray irradiation and unscheduled DNA synthesis (UDS) activity after UV irradiation betwe

Tetraplex formation by the progressive myoclonus epilepsy type-1 repeat: implications for instability in the repeat expansion diseases.

The repeat expansion diseases are a group of genetic disorders resulting from an increase in size or expansion of a specific array of tandem repeats. It has been suggested that DNA secondary structures are responsible for this expansion. If this is so, we would expect that all unstable repeats should form such structures. We show here that the unstable repeat that causes progressive myoclonus epilepsy type-1 (EPM1), like the repeats associated with other diseases in this category, forms a variety of secondary structures. However, EPM1 is unique in that tetraplexes are the only structures likel

Dot-and-fleck retinopathy in Alport syndrome caused by a novel mutation in the COL4A5 gene.

PURPOSE: To describe an unusual form of dot-and-fleck retinopathy in a slower progressive form of X-linked Alport syndrome, caused by a novel missense mutation in the COL4A5 gene. METHOD: Ophthalmic examination, polymerase chain reaction, and single-strand conformational polymorphism analysis of genomic DNA were performed in the proband. RESULTS: Ophthalmoscopy revealed classic dot-and-fleck retinopathy but located in an unusual site. A novel COL4A5 gene mutation changing glycine to cysteine at 177 was identified. CONCLUSIONS: Although there is no correlation between mutation site and the r

[Unscheduled DNA synthesis in human peripheral blood lymphocytes undergoing combined exposure to gamma irradiation and methylmethane sulfonate].

The value of the unscheduled DNA synthesis by exposure to combined action of damaging agents--gamma-irradiation and methyl methanesulfonate--does not exceed its value after separate actions of each particular agent in lymphocytes from healthy subjects as well as in those with Xeroderma pigmentosum in form II (XP2LE). This result is due to inhibiting effects of alkylating agents on DNA-polymerases.

Sustainable DNA-polysaccharide hydrogels as recyclable bioplastics.

Traditional petrochemical-derived plastics are challenging to recycle and degrade, and the existing (re)process methods are organic solvent-based and/or energy-intensive, resulting in significant environmental contamination and greenhouse gas emissions. This study presents a sustainable bioplastic material characterized by multi-closed-loop recyclability and water (re)processability. The bioplastics are derived from abundant polysaccharide sources of dextran, alginic acid, carboxymethyl cellulose, and DNA of plant and living organism waste. The process involves chemical oxidation of polysaccha

Mechanisms of fungal pathogenic DNA-activated STING pathway in biofilms and its implication in dental caries onset.

Dental caries, a prevalent oral disease, has long been attributed primarily to bacteria, but emerging evidence highlights the critical role of fungi in its pathogenesis. Fungal biofilms, predominantly Candida albicans, release extracellular DNA (eDNA) and DNA-carrying extracellular vesicles (EVs). Together with bacterial eDNA, these form the biofilm matrix and can activate the host cGAS-STING signaling pathway. This review systematically elaborates on the molecular architecture and biological functions of the cGAS-STING pathway, comparing mechanistic differences in its activation by viral, bac

Assessing the contribution of rare DNA states to cancer mutational signatures using sequence-specific conformational fingerprinting.

Rare and short-lived DNA conformations are proposed to be key drivers of mutagenesis, yet assessing their contribution to mutational signatures found in human cancers remains challenging. Here, we developed an approach that quantifies the sequence-dependent propensity to form a rare DNA conformation and compares the resulting fingerprint against cancer mutational signatures. Using 19F NMR, we measured the propensity for the anionic Watson-Crick-like G•T- conformation across all sixteen triplet sequence contexts and discovered a striking 50-fold variation driven by suboptimal interactions betwe

Selection of RNA-binding peptides in vivo.

Many priniciples of sequence-specific DNA recognition have been established over the past decade, largely from structural studies of protein-DNA and drug-DNA complexes. On the basis of these principles, it has been possible to design or select variants of known structural motifs, including zinc-fingers and minor groove-binding drugs, that bind desired sequences. Here we describe a strategy, based on transcriptional termination in bacteria, to identify specific RNA-binding peptides using the arginine-rich RNA-binding motif as a framework. Peptides were isolated from two combinatorial libraries

Topoisomerase IIα represses transcription by enforcing promoter-proximal pausing.

Accumulation of topological stress in the form of DNA supercoiling is inherent to the advance of RNA polymerase II (Pol II) and needs to be resolved by DNA topoisomerases to sustain productive transcriptional elongation. Topoisomerases are therefore considered positive facilitators of transcription. Here, we show that, in contrast to this general assumption, human topoisomerase IIα (TOP2A) activity at promoters represses transcription of immediate early genes such as c-FOS, maintaining them under basal repressed conditions. Thus, TOP2A inhibition creates a particular topological context that r

Cytotoxic molecule expression and epithelial cell apoptosis in oral and cutaneous lichen planus.

We evaluated the expression of T cell-restricted intracellular antigen (Tia-1), granzyme B, and perforin by lymphocytes and the degree of epithelial apoptosis in oral and cutaneous lichen planus (LP) in 51 untreated cases, including 27 oral LP (OLP) and 24 cutaneous LP (CLP) cases. The number of total dermal-positive lymphocytes in OLP and CLP was similar, indicating similar activity of the inflammatory process. Intraepithelial Tia-1-positive, perforin-positive, and granzyme B-positive lymphoid cells were more numerous in OLP than in CLP (P < .05). The epithelial cell apoptotic index (AI) was

The carboxyl-terminal region common to lamins A and C contains a DNA binding domain.

Lamins A and C are intermediate filament proteins which polymerize into the nucleus to form the nuclear lamina network. The lamina is apposed to the inner nuclear membrane and functions in tethering chromatin to the nuclear envelope and in maintaining nuclear shape. We have recently characterized a globular domain that adopts an immunoglobulin fold in the carboxyl-terminal tail common to lamins A and C. Using an electrophoretic mobility shift assay (EMSA), we show that a peptide containing this domain interacts in vitro with DNA after dimerization through a disulfide bond, but does not interac

G-Quadruplex DNA Motifs in the Malaria Parasite Plasmodium falciparum and Their Potential as Novel Antimalarial Drug Targets.

G-quadruplexes are DNA or RNA secondary structures that can be formed from guanine-rich nucleic acids. These four-stranded structures, composed of stacked quartets of guanine bases, can be highly stable and have been demonstrated to occur in vivo in the DNA of human cells and other systems, where they play important biological roles, influencing processes such as telomere maintenance, DNA replication and transcription, or, in the case of RNA G-quadruplexes, RNA translation and processing. We report for the first time that DNA G-quadruplexes can be detected in the nuclei of the malaria parasite

Targeted Protein Degradation Strategies in DNA Virus Research.

DNA viruses rely extensively on host cellular machinery, including replication factors and transcriptional systems, to persist after infection. These mechanisms make studying and targeting DNA viral proteins challenging, as they also play key roles in mammalian processes. Traditional strategies include CRISPR-mediated gene disruption and small interfering RNA (siRNA) to target host proteins. However, Proteolysis Targeting Chimeras (PROTACs) offer a novel strategy by enabling the selective and rapid degradation of specific viral or host proteins involved in the DNA viral lifecycle. PROTACs are

Ultrasensitive Photoelectrochemical Assay for DNA Detection Based on a Novel SnS2/Co3O4 Sensitized Structure.

In this work, an ultrasensitive photoelectrochemical (PEC) assay was established for sensitive DNA detection based on a novel SnS2/Co3O4 sensitized structure as a photoactive matrix and benzo-4-chlorohexadienone (4-CD) precipitate as a signal quencher. Noticeably, the photoelectric conversion efficiency of the SnS2/Co3O4 sensitized structure was dramatically enhanced due to the effective sensitization of Co3O4 toward SnS2, thus attaining an intense photocurrent response, which was sixfold higher than that of pristine SnS2. Additionally, with the assistance of Nt.BstNBI enzyme-assisted target c
