Butyrate receptor HCAR2/GPR109A controls imiquimod-induced psoriasis-like skin inflammation.

Psoriasis is a chronic inflammatory skin disorder characterized by aberrant keratinocyte proliferation and immune cell infiltration with upregulation of inflammatory cytokines. Here, we examined the contribution of HCAR2 encoding for the short-chain fatty acid receptor GPR109A. Human and mouse RNA sequencing public datasets reveal elevated HCAR2 gene expression in psoriatic as compared with healthy skin, both in keratinocytes and myeloid cells. Immunostaining and flow cytometry of imiquimod-induced psoriatic-like lesions in Hcar2-mRFP reporter mice showed increased GPR109A expression by kerati

Brain structural differences in cocaine use disorder: Insights from multivariate and neurotransmitter analyses.

Cocaine use disorder (CUD) is a chronic and relapsing neuropsychiatric disorder characterized by structural and functional brain lesions, posing a significant public health challenge. While the disruptive effects of cocaine on neurotransmitter systems (receptors/transporters) have been well established, the patterns of brain structural abnormalities in CUD and its interaction with other factors remain an ongoing topic of investigation. We employed source-based morphometry (SBM), a multivariate approach on 50 CUD participants and 50 matched healthy controls from the public SUDMEX CONN dataset.

Automatic CD30 scoring method for whole slide images of primary cutaneous CD30+ lymphoproliferative diseases.

AIMS: Deep-learning methods for scoring biomarkers are an active research topic. However, the superior performance of many studies relies on large datasets collected from clinical samples. In addition, there are fewer studies on immunohistochemical marker assessment for dermatological diseases. Accordingly, we developed a method for scoring CD30 based on convolutional neural networks for a few primary cutaneous CD30+ lymphoproliferative disorders and used this method to evaluate other biomarkers. METHODS: A multipatch spatial attention mechanism and conditional random field algorithm were use

Prediction of activity and selectivity profiles of human Carbonic Anhydrase inhibitors using machine learning classification models.

The development of selective inhibitors of the clinically relevant human Carbonic Anhydrase (hCA) isoforms IX and XII has become a major topic in drug research, due to their deregulation in several types of cancer. Indeed, the selective inhibition of these two isoforms, especially with respect to the homeostatic isoform II, holds great promise to develop anticancer drugs with limited side effects. Therefore, the development of in silico models able to predict the activity and selectivity against the desired isoform(s) is of central interest. In this work, we have developed a series of machine

Legal judgment prediction via dynamic task graph and keyword-aware contrastive learning.

Legal judgment prediction (LJP) aims to predict judgment results based on the factual descriptions of criminal cases and is gradually becoming a popular research topic in the legal field. Case factual descriptions contain a large amount of keyword information, and there are complex dependencies among the subtasks. For example, law article prediction can guide charge prediction and term of penalty prediction. However, most of the previous methods often failed to take advantage of the effective keywords that are commonly present in case descriptions, or failed to consider the interrelationships

HSFSurv: A hybrid supervision framework at individual and feature levels for multimodal cancer survival analysis.

Multimodal data play a significant role in survival analysis, with pathological images providing morphological information about tumors and genomic data offering molecular insights. Leveraging multimodal data for survival analysis has become a prominent research topic. However, the heterogeneity of data poses significant challenges to multimodal integration. While existing methods consider interactions among features from different modalities, the heterogeneity of feature spaces often hinders performance in survival analysis. In this paper, we propose a hybrid supervised framework for survival

Unraveling DINCH - Induced hepatotoxicity mechanisms via network toxicology and molecular docking with experimental validation.

Phthalates, as a class of known endocrine disruptors, have been controversial because of their potential carcinogenicity and toxicity. Diisononyl cyclohexane-1,2-dicarboxylate (DINCH) is considered to be less toxic and more prone to environmental degradation, and is widely used as a substitute for phthalate. With the increasing use of DINCH in consumer products and industrial materials, the frequency of its detection in the air and human urine has also increased, which has aroused concern about its potential toxicity in food safety. Despite the increasing popularity of DINCH, toxicological stu

Lightweight transformer-based generative adversarial network for acoustic anomaly detection in converter valves.

Anomaly detection of converter valves via acoustic analysis has been a hot topic in the high-voltage direct current research field. However, several factors have significantly hindered the practical deployment of such acoustic anomaly detection methods, including the infrequency of valve anomalies leading to imbalanced distributions of acoustic samples, limitations in computational resources, and inherent class imbalance in acoustic signals. To address these challenges, this paper proposes a novel unsupervised anomaly detection framework, named lightweight transformer-based generative adversar

Understanding the key challenges in tuberculosis drug discovery: what does the future hold?

INTRODUCTION: Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), remains a major global health concern. It spreads through airborne droplets and has a high mortality rate, particularly without treatment. Drug resistance is rising, with treatments against multidrug-resistant TB (MDR-TB) showing poor treatment success rates. The thick, lipid-rich wall of Mtb and its slow growth reduce antibiotic effectiveness, requiring long treatment courses of 4-6 months. Current therapies often fail against drug-resistant strains, highlighting the urgent need for new, short-course treatment, affor

Novel automated non-invasive detection of ocular surface squamous neoplasia using artificial intelligence.

Ocular surface squamous neoplasia (OSSN) is a common eye surface tumour, characterized by the growth of abnormal cells on the ocular surface. OSSN includes invasive squamous cell carcinoma (SCC), in which tumour cells penetrate the basement membrane and infiltrate the stroma, as well as non-invasive conjunctival intraepithelial neoplasia, dysplasia, and SCC in-situ thereby presenting a challenge in early detection and diagnosis. Early identification and precise demarcation of the OSSN border leads to straightforward and curative treatments, such as topical medicines, whereas advanced invasive

Wearable-Sensor-Based Weakly Supervised Parkinson's Disease Assessment with Data Augmentation.

Parkinson's disease (PD) is the second most prevalent dementia in the world. Wearable technology has been useful in the computer-aided diagnosis and long-term monitoring of PD in recent years. The fundamental issue remains how to assess the severity of PD using wearable devices in an efficient and accurate manner. However, in the real-world free-living environment, there are two difficult issues, poor annotation and class imbalance, both of which could potentially impede the automatic assessment of PD. To address these challenges, we propose a novel framework for assessing the severity of PD p
