Sensory information is encoded throughout the central nervous system by activation of specific groups of neurons. Neurons encoding information from a particular modality are grouped together and constitute an ordered neural representation or "map" of the stimulus. The organization of these representations is not static, but is capable of significant alteration in response to changes in the patterns of inputs delivered to the cortex in the appropriate behavioral context. Therefore, understanding the basic mechanisms that account for discontinuities in cortical representations are important for

The RelB gene product is a member of the nuclear factor (NF)-kappaB family of transcription factors. It has been identified recently within mouse antigen-presenting cells and human monocyte-derived dendritic cells (DC). Disruption of the mouse RelB gene is accompanied, amongst other phenotypes, by abnormalities in the antigen-presenting cell lineages. In order to define RelB expression during human DC differentiation, we have analysed RelB mRNA by reverse transcriptase-polymerase chain reaction and RelB protein by intracellular staining in CD34+ precursors and different types of DC preparation

Chlamydia species are the causative agents of trachoma, various forms of pneumonia, and the most common sexually transmitted diseases. Although the infection cycle has been extensively characterized in epithelial cells, where the Chlamydia entry-vacuoles avoid fusion with host-cell lysosomes, the cellular immune response has received less attention. Moreover, despite the abundant presence of dendritic cells (DC) in the sites of infection, the interaction between Chlamydia and DC has never been studied. We observe that DC kill Chlamydia trachomatis and Chlamydia psittaci. The chlamydiae are int

This paper describes an antibody (mAb 7D6) that specifically recognizes human follicular dendritic cells (FDCs). By expression cloning, a cDNA clone encoding for the long human CR2/ CD21 isoform (CD21L) that contains an additional exon (10a) was isolated. We demonstrated that FDCs selectively express CD21L, while B cells selectively express the short CR2/CD21 lacking exon 10a (CD21S). By screening mouse Ltk- cells transfected with the CD21L cDNA, we further showed that the other two anti-human FDC mAbs DRC-1 and KiM4 also recognize CD21L. Thus, CD21L represents the first characterized human FD

After antigen capture, dendritic cells (DC) migrate into T cell-rich areas of secondary lymphoid organs, where they induce T cell activation, that subsequently drives B cell activation. Here, we investigate whether DC, generated in vitro, can directly modulate B cell responses, using CD40L-transfected L cells as surrogate activated T cells. DC, through the production of soluble mediators, stimulated by 3- to 6-fold the proliferation and subsequent recovery of B cells. Furthermore, after CD40 ligation, DC enhanced by 30-300-fold the secretion of IgG and IgA by sIgD- B cells (essentially memory

The purpose of this study was to investigate the relationship between changes in density and distribution of dendritic cells, both in epidermis and in peritumoral infiltrate, and lymphocyte subset variations in malignant melanomas (MM) of patients belonging to different risk groups. The collective immunoreactive expression of six markers (S100 protein, CD1-a, HLA-DR, CD4, CD8 and CD25) was analyzed in 13 cutaneous malignant melanomas. Changes were observed in density and distribution of Langerhans cells (LC) (S100+, CD1-a+) in the epidermis overlying the tumor, as well as in peritumoral and in

The adenoids have been studied by immunocytochemistry and electron microscopy in children with and without adenoidal enlargement. Compared with normal adenoids, the enlarged ones showed a marked increase in the number of intraepithelial gamma-delta TCR+ lymphocytes and a slight increase in the number of intraepithelial CD8+ lymphocytes. This was accompanied by large amounts of dendritic human lymphocyte antigen (D related) (HLA-DR+) S-100+ accessory cells in the lymphoid tissue underlying the epithelium. By electron microscopy, dead epithelial cells apposed to intraepithelial lymphocytes, and

Dendritic cells (DC) isolated from the lymph nodes or spleens of mice and pulsed with contact sensitizers or protein antigens stimulate primary proliferative responses by syngeneic T cells and responses to alloantigens in the mixed leucocyte reaction (MLR). Using enriched human peripheral blood DC, we attempted to stimulate primary immune responses to contact sensitizers by autologous lymphocytes in vitro. No significant proliferation above background levels or CD69 expression (an early activation antigen on lymphocytes) was detected despite using a wide range of donors, chemicals, antigens an

Many details of the pathogenesis of the human immunodeficiency virus type 1 remain to be elucidated. Details of how the virus gains entry via the mucosal surface upon sexual contact or during breast feeding remain obscure. The means by which the infection travels throughout the body as well as the nature of the major reservoirs of virus infection remains, for the most part, unknown. Recent studies raise the possibility that cells of the Langerhans/dendritic lineage play a central role in human immunodeficiency virus (HIV-1) infection and pathogenesis. It has been known for several years that v

In this study, we have investigated by light and electron microscopy the presence, distribution, and inner structure of CD36(OKM5)+ dendritic cells (DC) in the lamina propria and epithelium of the oral mucosa of HIV- and HIV+ subjects; in the latter, both clinically healthy areas and areas of hairy leukoplakia (HL) were studied. Perivascular CD36+ DC were present in the lamina propria of all the specimens studied. They were also found in small numbers in the epithelium of clinically healthy mucosa of HIV- and HIV+ subjects, but were practically absent from the epithelium of HL. CD36+ DC seemed

The expression of HLA-DR and OKT6 antigens by epidermal cells in pemphigus was examined by immunohistochemical methods using skin biopsy specimens from human patients with pemphigus vulgaris. Changes in the distribution and subpopulations of Langerhans cells were observed in lesional (involved) skin. HLA-DR-positive, OKT6-negative dendritic cells were numerous and generally confined to the margins of epidermal clefts. OKT6-positive dendritic cells were rare or absent in lesional skin. The HLA-DR-positive, OKT6-negative dendritic cells associated with intraepidermal clefts are most likely funct

A collection of new and established FDC-reactive MoAb has been used in an immunohistological study designed to throw light on (a) the nature of FDC (which are of unknown lineage) as judged by their sharing of antigens with other cell types and (b) the reason for the strong expression of some B cell antigens on FDC. The MoAb were tested on: (1) sections of tonsil, (2) sections of lymph nodes from four cases of non-Hodgkin's lymphoma, (3) peripheral blood cells and (4) cells of cultured haemopoietic cell lines. Only one of the ten new MoAb bound to FDC and no other component of the tissues scree

Full thickness skin biopsies were examined from 12 untreated leprosy patients and included five borderline tuberculoid (BT leprosy), five borderline lepromatous (BL leprosy) and two subpolar lepromatous leprosy cases. The non-lymphoid mononuclear cells present in the dermal infiltrates were analysed with immunohistological techniques using monoclonal antibodies (MoAb) which in normal tissues identify subpopulations of macrophage-like cells in tissue sections; RFD2 (recognizing all and monocytes/macrophages), RFD1 (recognizing interdigitating cells), NA1/34 (recognizing Langerhans cells) and RF

Golgi studies of three cortical biopsies from patients with SSPE reveal an abnormal pyramidal dendritic domain, spine-loss, and reactive gliosis. These findings support results obtained in a previous ultrastructural study of this disease, and are in keeping with the pathological features observed in other situations in which mental changes occur.

In the central nervous system, the majority of excitatory synapses exist on small actin-enriched structures protruding from dendrites, known as dendritic spines. Actin cytoskeletal rearrangements drive dynamic changes in spine shape, size, and density depending on developmental stage and synaptic activity. Spines initially emerge from the dendritic shaft as dynamic protrusions known as filipodia-like spines, which serve as precursors to mature dendritic spines. The formation and maturation of dendritic spines facilitate information transfer in neural circuits, underlying cognitive processes, s…

Antigen-presenting cells shape the efficacy of cancer immunotherapies by providing essential signals required for antitumor T-cell response. Dendritic cells (DCs) are central to oncolytic virus (OV)-induced antitumor immunity because they couple tumor-antigen acquisition to cross-presentation and T-cell priming in tumor-draining lymph nodes. In tumors, however, DC recruitment, maturation, migration, and antigen-processing functions are frequently impaired by immunosuppressive and metabolic constraints. OVs can partially reprogram this axis by selectively infecting and lysing tumor cells, incre

Dendritic cells (DCs) are crucial in processing and cross-presenting tumor-associated antigens via MHC I molecules to prime CD8+ T lymphocytes (CTLs) for antitumor immunity. However, the moderate regulation of antigen endocytic escape from lysosomes into the cytoplasm hinders this cross-presentation process due to the lack of finely tunable tools at the organelle-specific level. Magneto-mechanical modulation technology provides a promising approach to regulate lysosomal membrane permeability (LMP) and promote antigen leakage for efficient cross-presentation. This approach relies on magnetic na

Abnormal deposition of the microtubule-associated protein tau has long been associated with spine loss and neuronal death in neurodegenerative diseases. Elucidating how pathological tau affects synaptic activity in vivo and whether individual synaptic properties dictate the survival fate of dendritic spines is central to understanding disease progression. Here we examined the visual response properties of layer 2/3 primary visual cortical dendrites and spines, using longitudinal two-photon calcium imaging in the P301S mouse model of tauopathy. Neuronal outputs in tau mutant mice were hyperacti…
