Effect of copper speciation on whole-cell soluble methane monooxygenase activity in Methylosinus trichosporium OB3b.

Soluble methane monooxygenase (sMMO) activity in Methylosinus trichosporium OB3b was found to be more strongly affected as copper-to-biomass ratios changed in a newly developed medium, M2M, which uses pyrophosphate for metal chelation, than in nitrate mineral salts (NMS), which uses EDTA. When M2M medium was amended with EDTA, sMMO activity was similar to that in NMS medium, indicating that EDTA-bound copper had lower bioavailability than pyrophosphate-bound copper. EDTA did not limit the association of copper with the cells; rather, copper was sequestered in a form which did not affect sMMO a

Effects of perphenazine on the metabolism of inositol phospholipids in SK-N-BE(2) human neuroblastoma cells.

Administration of myo-[3H]inositol to SK-N-BE(2) human neuroblastoma cells for 24 hr resulted in equilibrium labelling of phosphatidylinositol (PI), phosphatidylinositol 4-phosphate (PIP) and phosphatidylinositol 4,5-bisphosphate (PIP2), as well as in retention of a large intracellular pool of free myo-[3H]inositol. Equilibrium labelling was no longer observed when cells were treated for 2 hr with 20 microM perphenazine (PPZ) in label-free medium; under these conditions, myo-[3H]inositol from the retained intracellular pool was incorporated into PI and PIP but not into PIP2. Analysis of water-

[The pathophysiologic basis for the incorporation of 99mTc-pyrophosphate into myocardial infarct].

The investigation studied the relation of 99mTc-pyrophosphate incorporation (99mTc-PYP) into experimental, 48-hours old myocardial infarction in dogs to tissue vascular supply and to the extent of necrotic tissue. The experimental myocardial infarction was induced in five animals during an operation by the ligation RIVA. The myocardial blood supply was measured in tissue samples, taken from transverse sections of the infarction by means of 86Rb captation. The extent of myocytolysis was measured by depletion of tissue creatine kinase (CK). In the subendocardial layer of the infarction it became

Technetium pyrophosphate muscle scans in inflammatory muscle disease.

Technetium-99M pyrophosphate (TcPYP) nuclear scans of extremities were performed on 15 patients at 10 minutes and 2 hours after isotope injection. Scans were carried out both to confirm the diagnosis of myositis and to direct subsequent muscle biopsy. Five of six patients with clinical features strongly suggestive of inflammatory muscle disease had positive scans. All muscle biopsies performed at areas of increased isotope uptake showed inflammatory muscle disease. All nine patients not suspected of active inflammatory muscle disease had negative scans. Two of these underwent muscle biopsy wit

Abnormalities of platelet aggregation in sickle cell disease.

In vitro platelet aggregation responses to epinephrine and ADP were significantly lower in patients with sickle cell disease when compared to age- and sex-matched normal subjects. Platelet aggregation responses were reduced in all patients regardless of clinical status. Measurement of circulating platelet aggregates showed normal values in patients with sickle cell disease in steady state or with infection alone, but a significant increase in others with acute vaso-occlusive crises. Recovery from vaso-occlusion was associated with a decline in circulating platelet aggregates. This study sugges

ATP-PPi exchange activity of progesterone receptor.

Progesterone receptor preparations from avian oviduct catalyze a pyrophosphate (PPi)-exchange reaction between ATP and 32P-labeled PPi. The reaction requires ATP exclusively and is Mn++-dependent. This enzyme activity is detectable in receptor preparations that have been purified extensively by chromatography on ATP-Sepharose and DEAE-Sephadex columns. Polyacrylamide gel electrophoresis of purified preparations reveals a comigration of [3H]progesterone-receptor complex and the enzyme activity. The PPi-exchange reaction is inhibited by both o-phenanthroline and rifamycin AF/013, which also bloc

The stiffness of the flagella of impaled bull sperm.

The elastic rigidity (stiffness) of impaled motionless bull sperm flagella has been determined by a manipulatory technique which permitted direct analytical treatment of the experimental system. The effects of external ATP and ADP were measured. It was found that ATP acts as a plasticizing agent, while ADP does not. The stiffness measured for flagella in a medium without ATP was 15 times greater than the value measured with 10 mM ATP present. The rigor-like stiffness measured with no ATP present is reversible with ATP and seems to be correlated to a transition in the state of the contractile s

Roles of poly(ADP-ribose) synthesis in repair and replication in normal human, Cockayne syndrome, and xeroderma pigmentosum fibroblasts after UV irradiation.

Roles of poly(ADP-ribose) in repair and replication were studied in UV-irradiated normal, xeroderma pigmentosum (XP), and Cockayne syndrome (CS) fibroblasts. UV radiation reduced cellular NAD with concurrent synthesis of poly-(ADP-ribose) in a dose-dependent manner in normal and CS cells, but not in XP cells. Enzymatic incision of DNA of UV-irradiated XP cells by T4-endonuclease V activated poly(ADP-ribose) polymerase. Methyl methanesulfonate (MMS) also reduced the cellular NAD in all the above strains. However, inhibitors of poly(ADP-ribose) synthesis did not affect UV-induced unscheduled DNA

Fluorescent sensor for acidic phosphatase based on aggregation induced emission of copper nanoclusters.

Copper nanoclusters (CuNCs) that display aggregation-induced emission (AIE) properties has promising applications in biosensing areas due to their good optical properties and good biocompatibility. But common AIE inducing strategies such as solvent effects and domain confinement effects lack specificity or require complex design, which limits their practical biosensing applications. To address these issues, this study designed a fluorescent biosensor based on the complexation of CuNCs with adenosine triphosphate (ATP) for the efficient detection of acid phosphatase (ACP). ATP induces CuNCs agg

The Arabidopsis thaliana PATL1 functions downstream of CaM3 to mediate vesicle trafficking under the heat shock pathway.

Although vesicle trafficking is highly responsive to various environmental triggers, its role in thermotolerance remains unclear. Arabidopsis thaliana Patellin 1 (PATL1) plays a crucial role in vesicle trafficking. To elucidate the role of PATL1-mediated vesicle trafficking in thermotolerance, we investigated PATL1 function under heat shock (HS) conditions and found PATL1 inhibited vesicle trafficking, leading to a decreased accumulation of HS proteins (HSPs), indicating a negative correlation with thermotolerance. Previously, we reported that the calmodulin (CaM) isoform, CaM3, is involved in

Pyrimidinergic calcium signaling links tubular metabolism to fibrosis in kidney disease.

Chronic kidney disease (CKD) is a major global health problem, with substantial associated morbidity and mortality. Fibrosis is the final common pathway of organ damage in CKD, so understanding how this arises during kidney injury is critical for building a holistic picture of the pathogenesis. Here, using gene expression data, intravital microscopy in mice and realistic cell models, we uncover evidence of a signaling pathway linking tubular pyrimidine metabolism and injury-evoked extracellular uridine diphosphate (UDP) release to activation of the P2Y6 receptor (P2Y6R) in surrounding fibrobla

Engineering a Thiamine-Dependent Benzoylformate Decarboxylase for Stereodivergent Radical C(sp3)-C(sp3) Bond Formation.

The stereodivergent construction of C(sp3)-C(sp3) bonds between two prochiral radicals remains a formidable challenge. Here, we report the engineering of a thiamine-diphosphate-dependent radical enzyme to catalyze redox-neutral and stereodivergent cross-coupling of two electrophilic C(sp3) precursors. This new-to-nature enzyme reactivity was achieved via light-accelerated single-electron transfer from an unprecedented enzymatic vinyl Breslow intermediate, generating two distinct prochiral radicals whose stereoselective recombination forges the new C(sp3)-C(sp3) bond. Through systematic protein

Cryo-EM structures of Cdr1 reveal snapshots of substrate transport and diverse inhibitor recognition.

In Candida albicans-a World Health Organization fungal priority pathogen-overexpression of the adenosine triphosphate (ATP)-binding cassette transporter Cdr1 drives multidrug resistance. We present seven cryo-electron microscopy structures capturing substrate entry and expulsion. An inward-facing transmembrane channel with three on-off substrate binding sites defines a proposed entry pathway for a single substrate molecule. Coordinated ATP binding to both nucleotide-binding domains induces transmembrane domain closure, driving the substrate expulsion; adenosine diphosphate release following AT

A modular, reusable biocatalytic flow system for UDP-GlcNAc production.

We report here the continuous flow synthesis of a high-value sugar nucleotide. Immobilisation of enzymes onto solid carriers permitted transfer of the biocatalysts into packed bed reactors to realise a continuous biocatalytic platform for the synthesis of uridine diphosphate N-acetylglucosamine (UDP-GlcNAc) on 100 mg scale, with capacity for multiple reuses. The modular continuous flow approach described here represents a significant, up to 11-fold, improvement in space time yield (STY) when compared to batch studies, along with preventing product induced enzyme inhibition, reducing the need f

Inhibition of the Parkinson's Disease-Related Protein DJ-1 by Endogenous Neurotoxins of the 1,2,3,4-Tetrahydroisoquinoline Family.

The protein DJ-1 appears to play a protective role in the development of Parkinson's disease (PD). Here, we show that endogenous neurotoxins of the 1,2,3,4-tetrahydroisoquinoline family (TIQs), formed upon reaction of various aldehydes such as methylglyoxal (MGO) with the neurotransmitter dopamine, act as irreversible inhibitors of the esterase activity of human DJ-1, with IC50 values between 15 and 57 μM. The presence of a catechol function appears to be essential for these inhibitory effects, which may be at the origin of the oxidation of cysteine 106, a crucial residue in the DJ-1 active si

Alpha-synuclein amyloids catalyze the degradation of ATP and other nucleotides.

Intracellular accumulation of alpha-synuclein amyloids is a main pathological hallmark in a subgroup of human neurodegenerative diseases called synucleinopathies. Cell death of energy-deprived dopaminergic neurons causes decreased dopamine levels, which underly many of the neurological symptoms in the most prevalent synucleinopathy, Parkinson's disease. Amyloid-mediated toxicity can proceed via gain-of-function through diverse pathways. In this work, we report that alpha-synuclein amyloids can degrade adenosine triphosphate in a catalytic fashion, producing adenosine diphosphate and adenosine
