Genome Mining and Total Synthesis of Viduamanide, A Non-canonical Prolineless Cycloamanide from Amanita vidua.

The MeOH extract of the newly described deadly mushroom Amanita vidua was dominated by an unknown peptide. Despite the limited fungal material available, the compound was isolated and characterized as a homodetic cyclic hexapeptide by NMR and MS/MS analyses, and its structure was confirmed by high-efficiency solid-phase peptide synthesis. Recently implemented genome mining further validated the structure and provided a broader insight into the cyclopeptide-producing capabilities of this rare mushroom. Named viduamanide, this compound is a non-canonical cycloamanide of the "prolineless" subtype

Lipase ITB 2.1: a structure- and function-validated model for anti-obesity drug screening.

Lipases are key enzymes in lipid metabolism and represent important therapeutic targets for obesity and metabolic disorders. This study integrates sequence analysis, structural modelling, molecular docking, molecular dynamics simulations, and enzymatic assays to elucidate evolutionary conservation and the inhibitory mechanism of orlistat against lipase ITB 2.1. Multiple sequence alignment revealed strong conservation of catalytic serine residue at the nucleophilic elbow across bacterial, fungal, and mammalian lipases, with rare AXSXG motif substitutions in Bacillus and Geobacillus species, ind

Role of the guanine nucleotide exchange factor Rom2 in cell wall integrity maintenance of Aspergillus fumigatus.

Aspergillus fumigatus is a mold and the causal agent of invasive aspergillosis, a systemic disease with high lethality. Recently, we identified and functionally characterized three stress sensors implicated in the cell wall integrity (CWI) signaling of this pathogen, namely, Wsc1, Wsc3, and MidA. Here, we functionally characterize Rom2, a guanine nucleotide exchange factor with essential function for the cell wall integrity of A. fumigatus. A conditional rom2 mutant has severe growth defects under repressive conditions and incorporates all phenotypes of the three cell wall integrity sensor mut

The Saccharomyces killer toxin K62 is a protein of the aerolysin family.

UNLABELLED: K62 is an antifungal killer toxin produced by Saccharomyces paradoxus, encoded by a double-stranded RNA satellite. The toxin exhibits a unique antifungal activity but lacks sequence homology to other killer toxins, and its antifungal mechanism of action remains unknown. To understand the function of K62, its tertiary structure was predicted using AlphaFold, followed by molecular dynamics simulations to create high-confidence molecular models. These analyses revealed that K62 monomers closely resemble the five-beta-strand domain found in pore-forming aerolysin toxins. Models of K62

Using Vibrio natriegens for high-yield production of challenging expression targets and for protein deuteration.

Production of soluble proteins is essential for structure/function studies, however, this usually requires milligram amounts of protein, which can be difficult to obtain with traditional expression systems. Recently, the Gram-negative bacterium Vibrio natriegens appeared as a novel and alternative host platform for production of proteins in high yields. Here, we used a commercial strain derived from V. natriegens (Vmax™ X2) to produce soluble bacterial and fungal proteins in milligram scale, which we struggled to achieve in Escherichia coli. These proteins include the cholera toxin (CT) and N-

Fire Severity Drives Long-Term Divergence of Soil Microbial Communities and Functional Traits in an Alpine Forest.

Increasing wildfire severity under climate change may alter the trajectory of belowground recovery in alpine forests, yet its long-term effects on soil microbial communities remain poorly understood. Here, we examined soil physicochemical properties and microbial communities eight years after fire across unburned, low-, moderate-, and high-severity fire sites in an alpine forest of western Sichuan, China. Fire severity was associated with persistent divergence at depths of 0-10 cm, with significant changes in soil organic carbon (SOC), soil water content (SWC), available nitrogen (AN), and mic

Global proteomic analysis of Cryptococcus neoformans clinical strains reveals significant differences between latent and lethal infection.

UNLABELLED: To predict the outcomes of disseminated fungal disease, a deeper understanding of host-pathogen interactions at the site of infection is needed to identify targets for clinical intervention and diagnostic development. Cryptococcus neoformans is the causative agent of cryptococcosis, the largest infectious killer of individuals living with HIV. Cryptococcal infection begins in the lungs, with loss of immunological control leading to disseminated central nervous system disease and death. Using advanced mass spectrometry-based proteomic techniques, in vivo infection models, and patien

Genetic Diversity and Genomic Characteristics of the Respiratory Virome in Patients With Severe Fungal Infections.

Respiratory tract infections represent a leading cause of morbidity and mortality globally, with viral pathogens accounting for a substantial proportion of these cases. However, research on the human respiratory virome is still in its infancy, and our understanding of this field remains relatively limited. In the present study, viral metagenomic sequencing was conducted on 65 sputum samples obtained from patients with severe fungal infections. We successfully assembled viral genome sequences belonging to four distinct viral families: Anelloviridae, Genomoviridae, Microviridae, and Inoviridae.

Recent research frontiers of heterocycles as antifungal Agents: Insights from the past five years.

This review explores the growing global concern of fungal infections, particularly in immunocompromised individuals, and highlights the critical need for improved antifungal therapies. With the rise of multidrug-resistant strains, such as Candida auris and Aspergillus fumigatus, current antifungal treatments face limitations, including toxicity, low bioavailability, and the development of resistance. Heterocyclic compounds, particularly those targeting key fungal enzymes and pathways, are emerging as promising candidates in the fight against these infections. The review focuses on the structur

One population, multiple lifestyles: Commensalism and pathogenesis in the human mycobiome.

Candida auris and Candida albicans can result in invasive fungal diseases. And yet, these species can stably and asymptomatically colonize human skin and gastrointestinal tracts. To consider these disparate microbial lifestyles, we first review factors shown to influence the underlying microbiome. Structured by the damage response framework, we then consider the molecular mechanisms deployed by C. albicans to switch between commensal and pathogenic lifestyles. Next, we explore this framework with C. auris to highlight how host physiology, immunity, and/or antibiotic receipt are associated with

Transungual Delivery of Ciclopirox Is Increased 3⁻4-Fold by Mechanical Fenestration of Human Nail Plate in an In Vitro Model.

Onychomycosis is a fungal infection of nails that is widespread and difficult to treat because of the impermeable nature of human nails. Topically applied anti-fungal agents cannot penetrate this structure, and treatment regimens often resort to systemic antifungals with concomitant side effects. One recent clinical study suggested that mechanical fenestration of the nail using an intelligent nail drill might be a possible solution to this problem. In this work, an in vitro model of the transungual delivery of antifungal agents is presented, which utilizes real nail tissue and an inline flow s

Impact of Rearing Conditions on the Ambrosia Beetle's Microbiome.

Ambrosia beetles, along with termites and leafcutter ants, are the only fungus-farming lineages within the tree of life. Bacteria harbored by ambrosia beetles may play an essential role in the nutritional symbiotic interactions with their associated fungi; however, little is known about the impact of rearing conditions on the microbiota of ambrosia beetles. We have used culture-independent methods to explore the effect of rearing conditions on the microbiome associated with Xyleborus affinis, Xyleborus bispinatus, and Xyleborus volvulus, evaluating different media in laboratory-controlled cond

[Research progress of the natural small molecular products synergistically with antifungal agents to inhibit drug-resistant fungi].

The incidence of systemic fungal infections have increased dramatically, moreover, drug resistance including either primary (intrinsic) or secondary (acquired) resistance, becomes one of the main reasons accounting for the failure of treating invasive fungal infections in the past decades. Nowadays, clinically available antifungal drugs are limited and their combination in antifungal therapy was not effective. It is expected to be a new strategy to synergistically sensitize antifungal drugs against drug-resistant fungi by using new small molecules. Based on the study in our research group and

Cryptococcus neoformans responds to mannitol by increasing capsule size in vitro and in vivo.

The polysaccharide capsule of the fungus Cryptococcus neoformans is its main virulence factor. In this study, we determined the effects of mannitol and glucose on the capsule and exopolysaccharide production. Growth in mannitol significantly increased capsular volume compared with cultivation in glucose. However, cells grown in glucose concentrations higher than 62.5 mM produced more exopolysaccharide than cells grown in mannitol. The fibre lengths and glycosyl composition of capsular polysaccharide from yeast grown in mannitol was structurally different from that of yeast grown in glucose. Fu

Discovery and Biosynthesis of a Novel Acorane-Type Sesquiterpene from Rhynchosporium agropyri.

Acoranes represent an important family of sesquiterpenes featuring a unique spiro[4.5]decane skeleton with remarkable structural diversity and extensive biological activities. In this study, genome mining of the plant pathogenic fungus Rhynchosporium agropyri enabled the identification of a small biosynthetic gene cluster (rhy) encoding sesquiterpene synthase RhyA and cytochrome P450 RhyB, which together catalyze the formation of a novel acorane-type sesquiterpene, rhyagracorone (1). Furthermore, by elucidating the biosynthetic pathway of 1, we confirmed that RhyA acts as an acora-3(4),7(8)-di

Structural Diversification and Biosynthesis of Pesticidal Pyrroline-Derived Alkaloid from Penicillium citrinum SCSIO DF147.

The pyrroline-derived alkaloid N-(2-methyl-3-oxodec-8-enoyl)-2-pyrroline (MOEP, 1) is a potential pesticide that remains to be structurally diversified for target screening and biosynthesis studies. Bioactivity-guided investigation of the marine-derived fungus Penicillium citrinum SCSIO DF147 yielded five classes of pyrroline-derived alkaloids (1-15), including five new compounds (2-4, 14, and 15). These compounds were first evaluated for chitinase (OfChtII and OfChi-h) inhibition, revealing that both the 3-pyrroline ring and the Δ8',9' double bond are key functional groups. Pesticidal and fun

Dendritic Antifungal Peptides as Potent Agents against Drug-Resistant Candida albicans and Biofilm.

Candida albicans infection is a major public health problem, exacerbated by the emergence of drug-resistant fungi with the widespread use of antifungal drugs. Therefore, the development of novel antifungal drugs for drug-resistant C. albicans infections is crucial. We constructed a series of dendritic antifungal peptides (AFPs) with different chain lengths of fatty acids as hydrophobic ends and 2 or 3 protease-stable repeats (Arg-Pro) as dendritic peptide branches. Among them, C4-3RP exhibited excellent antidrug-resistant fungal and biofilm activity (GMall = 5.04 μM) and was nontoxic. Furtherm

Azole-triphenylphosphonium conjugates combat antifungal resistance and alleviate the development of drug-resistance.

Azole antifungals are commonly used to treat fungal infections but have resulted in the occurrence of drug resistance. Therefore, developing azole derivatives (AZDs) that can both combat established drug-resistant fungal strains and evade drug resistance is of great importance. In this study, we synthesized a series of AZDs with a fluconazole (FLC) skeleton conjugated with a mitochondria-targeting triphenylphosphonium cation (TPP+). These AZDs displayed potent activity against both azole-sensitive and azole-resistant Candida strains without eliciting obvious resistance. Moreover, two represent

Spiroaxillarone A, a Symmetric Spirobisnaphthalene with an Original Skeleton from Cyanotis axillaris.

The first representative of a new type of spirobisnaphthalene with a previously unknown skeletal system, Spiroaxillarone A (1), was isolated from Cyanotis axillaris. The planar structure was determined through analysis of spectroscopic evidence, while electronic circular dichroism (ECD) calculations were used to determine its absolute configuration. The structure of 1 was further confirmed through X-ray diffraction studies of its tetrabromobenzoate derivative (1a). Spiroaxillarone A exhibited significant antimalarial activity against resistant Plasmodium falciparum (IC50 = 2.32 μM).
