[Contribution of sigma B to environmental stress tolerance in Listeria monocytogenes--a review].

The alternative sigma factor Sigma B plays important roles in both virulence and stress tolerance in Listeria monocytogenes. It is now clear that there is a strong link between the virulence potential of Listeria monocytogenes and its ability to tolerate stress. Several studies have identified genes that play important roles in stress tolerance and virulence. For example, genes involved in osmotic stress tolerance, acid and alkaline tolerance, oxidative stress tolerance, extreme temperature tolerance, and bile salt tolerance have all been implicated in the virulence of L. monocytogenes. We rev

Retentive memory of bacteria: Long-term regulation of dehalorespiration in Sulfurospirillum multivorans.

The gram-negative, strictly anaerobic epsilonproteobacterium Sulfurospirillum multivorans is able to gain energy from dehalorespiration with tetrachloroethene (perchloroethylene [PCE]) as a terminal electron acceptor. The organism can also utilize fumarate as an electron acceptor. Prolonged subcultivation of S. multivorans in the absence of PCE with pyruvate as an electron donor and fumarate as an electron acceptor resulted in a decrease of PCE dehalogenase (PceA) activity. Concomitantly, the pceA transcript level equally decreased as shown by reverse transcriptase PCR. After 35 subcultivation

The mucosal expression signatures of g-type lysozyme in turbot (Scophthalmus maximus) following bacterial challenge.

The mucosal surfaces constitute the first line of host defense against infection, and also serve as the dynamic interfaces that simultaneously mediate a diverse array of critical physiological processes, while in constantly contact with a wide range of pathogens. The lysozymes are considered as key components for innate immune response to pathogen infection with their strong antibacterial activities. But their activities in mucosal immune responses were always overlooked, especially for g-type lysozymes, whose expression patterns in mucosal tissues following bacterial challenge are still limit

A snap-shot of Mannheimia hemolytica A1 gene expression during infection in the bovine host.

It is expected that Mannheimia hemolytica A1 expresses a particular collection of genes during infection in the host. The bacterial gene products are produced in the in vivo environment to facilitate growth and survival. Here, we examined gene expression by M. hemolytica A1 in the bovine host after 6 days of infection. Total RNA from M. hemolytica A1 recovered from pneumonic lungs of two animals was used to produce cDNA to screen a custom M. hemolytica A1 microarray. The expression profile was compared to a RNA sample from an in vitro grown culture. The data showed that 44 genes were different

Combinatorial action of regulatory systems generates colistin heteroresistance.

The mechanism by which phenotypic heterogeneity is generated within a bacterial population and leads to colistin heteroresistance has been unclear. Heteroresistance is a form of antibiotic resistance in which a minor subpopulation of resistant cells coexists with a majority susceptible population. Colistin heteroresistance is common among Enterobacter isolates, threatens its utility as a therapeutic, and has become a model to study the fundamental bases of heteroresistance. Here, we identify the sigma factor σE as the source of heterogeneity in colistin resistance. Single-cell tracking reveale

Genetic and functional characterization of the natural transformation system in Streptococcus constellatus.

Streptococcus constellatus is an opportunistic pathogen frequently associated with abscess formation in various body sites. While the species has been shown to acquire exogenous DNA through natural transformation, functional analyses of its underlying mechanisms and optimized genetic editing protocols remain limited. Thus, our aim was to characterize the natural transformation system in S. constellatus and investigate environmental factors coordinating its activation. In addition, we sought to develop an optimized protocol for genome editing. Genomic analysis revealed that 73% of analyzed stra

Vegetation Formation in Staphylococcus Aureus Endocarditis Inversely Correlates With RNAIII and sarA Expression in Invasive Clonal Complex 5 Isolates.

Infective endocarditis (IE) is one of the most feared and lethal diseases caused by Staphylococcus aureus. Once established, the infection is fast-progressing and tissue destructive. S. aureus of the clonal complex 5 (CC5) commonly cause IE yet are severely understudied. IE results from bacterial colonization and formation of tissue biofilms (known as vegetations) on injured or inflamed cardiac endothelium. S. aureus IE is promoted by adhesins, coagulases, and superantigens, with the exotoxins and exoenzymes likely contributing to tissue destruction and dissemination. Expression of the large r

Envelope destabilization by AcrAB2NodT overexpression links antibiotic resistance to metal sensitivity in Caulobacter vibrioides.

Antibiotic (AB) resistance often arises through mutations that alter AB targets, reduce membrane permeability, or increase efflux activity, yet its physiological costs remain poorly understood. In Caulobacter vibrioides, a ΔtipR mutant that constitutively upregulates the RND efflux pump AcrAB2NodT exhibits heightened sensitivity to copper (Cu), revealing a trade-off linked to excessive pump expression. Deleting acrAB2nodT or impairing its transport activity restored cell envelope integrity and, ultimately, Cu resistance, uncoupling the effects of pump abundance from active efflux. The ΔtipR mu

Bacterial heterozygosity promotes survival under multidrug selection.

Although bacterial cells typically contain a single chromosome, some species are naturally polyploid and carry multiple copies of their chromosome. Polyploid chromosomes can be identical or heterogeneous, the latter giving rise to bacterial heterozygosity. Although the benefits of heterozygosity are well studied in eukaryotes, its consequences in bacteria are less understood. Here, we examine this question in the context of antibiotic resistance to understand how bacterial genomic heterozygosity affects bacterial survival. The intrinsic resistome of Klebsiella pneumoniae.

Molecular epidemiology studies aiming at understanding the acquisition of antimicrobial resistance by clinical isolates of Klebsiella pneumoniae are regularly published; however, information on the genes that contribute to its characteristic phenotype of resistance to antibiotics (intrinsic resistome) is scarce. To fill this gap, a K. pneumoniae transposon mutant library was screened and 171 mutants presenting changes in their susceptibility to antibiotics were selected, in which the transposon insertion site was determined in 75. Twenty-seven mutants for which insertion points had been previo

The intrinsic cephalosporin resistome of Listeria monocytogenes in the context of stress response, gene regulation, pathogenesis and therapeutics.

Intrinsic resistance to antibiotics is a serious therapeutic problem in the case of many bacterial species. The Gram-positive human pathogen Listeria monocytogenes is intrinsically resistant to broad spectrum cephalosporin antibiotics, which are commonly used in therapy of bacterial infections. Besides three penicillin-binding proteins the intrinsic cephalosporin resistome of L. monocytogenes includes multidrug resistance transporter transporters, proteins involved in peptidoglycan biosynthesis and modification, cell envelope proteins with structural or general detoxification function, cytopla

Calcium causes multimerization of the large adhesin LapF and modulates biofilm formation by Pseudomonas putida.

LapF is a large secreted protein involved in microcolony formation and biofilm maturation in Pseudomonas putida. Its C-terminal domain shows the characteristics of proteins secreted through a type I secretion system and includes a predicted calcium binding motif. We provide experimental evidence of specific binding of Ca(2+) to the purified C-terminal domain of LapF (CLapF). Calcium promotes the formation of large aggregates, which disappear in the presence of the calcium chelator EGTA. Immunolocalization of LapF also shows the tendency of this protein to accumulate in vivo in certain extracel

Characterization of the general stress response in Bartonella henselae.

Bacteria utilize a general stress response system to combat stresses from their surrounding environments. In alpha-proteobacteria, the general stress response uses an alternate sigma factor as the main regulator and incorporates it with a two-component system into a unique regulatory circuit. This system has been described in several alpha-proteobacterial species, including the pathogens Bartonella quintana and Brucella abortus. Most of the studies have focused on characterizing the PhyR anti-anti-sigma factor, the NepR anti-sigma factor, and the alternate sigma factor. However, not enough att

Long Non-coding RNA LincRNA-EPS Inhibits Host Defense Against Listeria monocytogenes Infection.

Long non-coding RNAs (lncRNAs) have emerged as key regulators of gene expression in several biological systems. The long intergenic RNA-erythroid pro-survival (lincRNA-EPS) has been shown to play a critical role in restraining inflammatory gene expression. However, the function of lincRNA-EPS during bacterial infections remains unknown. Here, we demonstrate that following infection with the intracellular bacterium Listeria monocytogenes, both mouse macrophages and dendritic cells lacking lincRNA-EPS exhibit an enhanced expression of proinflammatory cytokine genes, as well as an increased expre

Metabolic rewiring and transcriptional interference underlie the biocontrol efficacy of Burkholderia arboris BB417 against Ralstonia solanacearum species complex.

Ralstonia solanacearum species complex (RSSC) is a globally destructive plant pathogen with high ecological persistence, making bacterial wilt management a long-standing challenge. In this study, we identified Burkholderia arboris BB417 as a potent biocontrol agent and systematically elucidated its multilevel antagonistic mechanisms against RSSC. BB417 exhibited broad-spectrum inhibition against diverse RSSC strains and achieved greenhouse control efficacy comparable to chemical pesticides within the 10-day observation period, while also promoting plant growth through nitrogen fixation, phosph

An insoluble de novo protein enables survival of Escherichia coli by sequestering a gene repressor.

De novo proteins that share no ancestry with natural sequences can serve as additions to the evolved proteomes of living cells. Upon expression in cells, these novel proteins can provide biological functions that alter cell viability and growth. To isolate such proteins, we searched a combinatorial library of novel sequences by selecting for sequences that sustain the growth of Escherichia coli under conditions where the recipient cell would otherwise be inviable. This led to the identification of Resc4 (Rescuer 4), a de novo protein that sustains growth on minimal medium of an E. coli strain

Vibrio cholerae virulence and its suppression through the quorum-sensing system.

Vibrio cholerae is a cholera-causing pathogen known to instigate severe contagious diarrhea that affects millions globally. Survival of vibrios depend on a combination of multicellular responses and adapt to changes that prevail in the environment. This process is achieved through a strong communication at the cellular level, the process has been recognized as quorum sensing (QS). The severity of infection is highly dependent on the QS of vibrios in the gut milieu. The quorum may exist in a low/high cell density (LCD/HCD) state to exert a positive or negative response to control the regulatory

Novel gene similar to nitrite reductase (NO forming) plays potentially important role in the latency of tuberculosis.

The development of the latent phenotype of Mycobacterium tuberculosis (Mtb) in the human lungs is the major hurdle to eradicate Tuberculosis. We recently reported that exposure to nitrite (10 mM) for six days under in vitro aerobic conditions completely transforms the bacilli into a viable but non-cultivable phenotype. Herein, we show that nitrite (beyond 5 mM) treated Mtb produces nitric oxide (NO) within the cell in a dose-dependent manner. Our search for the conserved sequence of NO synthesizing enzyme in the bacterial system identified MRA2164 and MRA0854 genes, of which the former was fou
