Circulating cytokines, chemokines and adhesion molecules in normal pregnancy and preeclampsia determined by multiplex suspension array.

BACKGROUND: Preeclampsia is a severe complication of pregnancy characterized by an excessive maternal systemic inflammatory response with activation of both the innate and adaptive arms of the immune system. Cytokines, chemokines and adhesion molecules are central to innate and adaptive immune processes. The purpose of this study was to determine circulating levels of cytokines, chemokines and adhesion molecules in normal pregnancy and preeclampsia in a comprehensive manner, and to investigate their relationship to the clinical features and laboratory parameters of the study participants, incl

A luminescent attenuated SARS-CoV-2 for the identification and validation of drug-resistant mutants.

UNLABELLED: The emergence of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) variants has necessitated a continuous updating of vaccines. In contrast, antivirals remained effective as they target conserved viral proteins that are essential for the viral life cycle. However, several mutations in SARS-CoV-2 that may affect the efficacy of United States (US) Food and Drug Administration (FDA)-approved antivirals have been recently identified. Detecting drug-resistant SARS-CoV-2 mutants and investigating their escape mechanism(s) are critical to guide the selection of effective antivi

A single-dose intranasal immunization with a novel bat influenza A virus-vectored MERS vaccine provides effective protection against lethal MERS-CoV challenge.

The threat to global health security posed by Middle East respiratory syndrome coronavirus (MERS-CoV) and emerging MERS-like coronaviruses highlights the need to develop safe and efficient vaccines. Viral vector vaccines have been shown to be effective and are widely used to prevent various viral diseases because they mimic natural infection and induce a more comprehensive immune response. Herein, we developed a novel bat influenza A virus-based vaccine vector by replacing the open reading frame of either bat influenza hemagglutinin or neuraminidase with that of the hemagglutinin-esterase-fusi

Personalized external aortic root support: The Dutch experience.

INTRODUCTION: Patients with connective tissue disease and aortic root aneurysm are at risk of dissection and progression of the dilatation. Standard treatment of aortic root aneurysm is either a total root replacement or valve-sparing root replacement. Both procedures have drawbacks, such as the need for anticoagulation and long-term exposure to the chance of reoperation or endocarditis. The ExoVasc personalized external aortic root support (PEARS) is a perivascular stent that has been introduced to reduce these risks while aiming for a similar beneficial outcome on late aortic complications.

HLA and T-Cell Receptor Investigations in Idiopathic and Paraneoplastic Opsoclonus-Myoclonus in Children.

BACKGROUND AND OBJECTIVES: Opsoclonus-myoclonus-ataxia syndrome (OMAS) is a rare pediatric autoimmune neurologic disorder, with 40%-50% of cases occurring as a paraneoplastic syndrome in the setting of neuroblastoma or other neuroblastic tumors. The strong clinical similarities between idiopathic and paraneoplastic OMAS have led to the hypothesis that neuroblastoma may initially be present in idiopathic cases but spontaneously regress before OMAS onset. Both B- and T-cell mechanisms appear to contribute to OMAS autoimmunity, although their precise roles remain unknown. The aim of this study wa

Neurocognitive and neuropsychiatric phenotypes associated with the mutation L238Q of the α-L-iduronidase gene in Hurler-Scheie syndrome.

UNLABELLED: The lysosomal enzyme α-L-iduronidase hydrolyzes terminal iduronic acid from heparan sulfate and dermatan sulfate, and is an essential step in GAG degradation. Mutations of its gene, IDUA, yield a spectrum of mucopolysaccharidosis (MPS) type I clinical disorders. The IDUA mutation, c.712T>A (p.L238Q) was previously noted as a mild mutation. In a longitudinal study of MPS brain structure and function (Lysosomal Disease Network), we found this mutation in 6 of 14 Hurler-Scheie syndrome patients in the age range of 15 to 25 years. We hypothesized that L238Q, when paired with a nonsense

Swallowing therapy--a prospective study on patients with neurogenic dysphagia due to unilateral paresis of the vagal nerve, Avellis' syndrome, Wallenberg's syndrome, posterior fossa tumours and cerebellar hemorrhage.

No studies exist dealing with the outcome of dysphagic patients with posterior fossa (IV. ventricle) tumours (PFT) or cerebellar hemorrhage (CH), and the outcome of patients with Wallenberg's syndrome (WS) after functional swallowing therapy (FST) has so far not been studied in detail. Patients and methods. 208 patients with neurogenic dysphagia (ND) who were consecutively admitted for functional swallowing therapy (FST) over a 3 year period to our hospital were examined clinically, by use of a videofluoroscopic swallowing study (VFSS) and/or fibreoptic evaluation of swallowing (FEES). From vector to allergen: exploring the immunology of tick-triggered α-Gal syndrome.

The α-Gal syndrome (AGS) is an emerging form of food allergy characterized by delayed hypersensitivity reactions to mammalian meat products and mediated by IgE antibodies specific to the carbohydrate galactose-α-1,3-galactose (α-Gal). Although α-Gal-specific IgG, IgM and IgA antibodies are generally present in humans as a consequence of continuous exposure to commensal microbiota and dietary sources, IgE sensitization to α-Gal occurs only in a subset of individuals. Epidemiological and experimental evidence has firmly linked this sensitization to tick bites. Multiple ticks across continents ha

Metabolic Demand, Renal Mass, and Glomerular Filtration Rate Changes in Men and Women: Lessons from Living Kidney Donors and Recipients.

BACKGROUND: The determinants of glomerular filtration rate (GFR) and its changes over time are multiple and diverse. Nephron endowment is the starting point for GFR. Low renal endowment due to maternal undernutrition or premature birth as well as the loss of renal mass because of surgical procedures have a major impact on GFR. Eventually low renal mass may lead to organ damage when combined with metabolic syndrome and obesity or diabetes. The kidney has a major role in maintaining homeostasis. Changes in metabolic demand have a major influence in renal function. Increments in metabolic demand

From lobotomy to connectomics: a critical review of contemporary psychosurgery.

Contemporary psychosurgery is increasingly regarded less as an invasive surgical discipline and more as a circuit-based medicine. This narrative critical review traces the evolution from the extensive frontal lesions of the early twentieth century to contemporary techniques, in which focal ablation and reversible neuromodulation form part of a shared therapeutic armamentarium. Building on the classical history-from trephinations and lobotomies to stereotaxy-we outline a functional neuroanatomical framework that integrates thelimbic system, basal ganglia, prefrontal cortex, and monoaminergic sy

Addressing SARS-CoV-2 evolution: neutralization of emerging variants of concern by the AVX/COVID-12 'Patria' vaccine based on HexaPro-S ancestral Wuhan spike or its updated BA.2.75.2 version.

INTRODUCTION: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) remains a global health challenge, causing severe morbidity and mortality, particularly in vulnerable groups such as the elderly, immunocompromised individuals, and those with comorbidities. In low- and middle-income countries (LMICs), vaccine access is hindered by high costs and inequitable distribution. To tackle these issues, Mexico developed the AVX/COVID-12 (V-Wu) vaccine, a recombinant Newcastle disease virus (NDV)-based platform expressing a stabilized ancestral Wuhan spike protein (HexaPro-S). Locally manufactur

Efficacy of a non-recombinant NADC34-like PRRSV-2 vaccine candidate against a recombinant NADC34-like PRRSV-2 isolate.

NADC34-like porcine reproductive and respiratory syndrome virus 2 (NADC34-like PRRSV-2) has become prevalent in Chinese swine herds. However, current commercial vaccines only provide limited cross-protection against NADC34-like PRRSV-2 infection. In our previous study, we developed the infectious clone (rBJ1805-2) of a non-recombinant NADC34-like PRRSV-2 BJ1805-2 isolate and identified the GP2a 91/97/98 amino acid substitutions (from "TMF" to "VVL" pattern) as a sufficient and necessary determinant for its Marc-145 cell tropism. In this study, the non-recombinant Marc-145-adapted NADC34-like m

[Follow-up in patients with testicular germ cell tumors-an update].

Systematic follow-up (FU) examinations in patients with testicular germ cell tumors (GCTs) came into practice in the 1980s. The goal is to detect recurrent disease as early as possible to facilitate successful treatment. Cross-sectional imaging and tumor marker measurements (AFP, bHCG) represent the mainstay of FU examinations. Due to the biological and clinical diversity of GCTs, there is no uniform general pattern of examinations that would suit all GCT patients. Therefore, FU should be performed in a risk-adapted way; the rationale of which is to tailor examinations to the extent needed, wh

Development of highly adaptable RT-PCR methods for identifying Delta and BA.1 variants in inactivated COVID-19 vaccines.

Background The emergence and rapid spread of coronavirus disease 2019 (COVID-19), caused by severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2), poses a significant threat to human health and public safety. While next-generation sequencing (NGS) is capable of detecting and tracking new COVID-19 variants for disease diagnosis and prevention, its high cost and time-consuming nature limit its widespread use. In this study, our aim was to develop a highly adaptable and accurate RT-PCR method for identifying the Delta or BA.1 variants in inactivated COVID-19 vaccine. We devised three two-p

Strong T-cell activation in response to COVID-19 vaccination in multiple sclerosis patients receiving B-cell depleting therapies.

Immunocompromised individuals, including multiple sclerosis (MS) patients on certain immunotherapy treatments, are considered susceptible to complications from severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection and specific vaccination regimens have been recommended for suitable protection. MS patients receiving anti-CD20 therapy (aCD20-MS) are considered especially vulnerable due to acquired B-cell depletion and impaired antibody production in response to virus infection and COVID-19 vaccination. Here, the humoral and cellular responses are analyzed in a group of aCD20-MS

Reciprocal Regulation of MAGED2 and HIF-1α Augments Their Expression under Hypoxia: Role of cAMP and PKA Type II.

Hypoxia stabilizes the transcription factor HIF-1α, which promotes the transcription of many genes essential to adapt to reduced oxygen levels. Besides proline hydroxylation, expression of HIF-1α is also regulated by a range of other posttranslational modifications including phosphorylation by cAMP-dependent protein kinase A (PKA), which stabilizes HIF-1α. We recently demonstrated that MAGED2 is required for cAMP generation under hypoxia and proposed that this regulation may explain the transient nature of antenatal Bartter syndrome (aBS) due to MAGED2 mutations. Consequently, we sought to det
