Genetic surveillance of Plasmodium falciparum populations following treatment policy revisions in the Greater Mekong Subregion.

Genetic surveillance of Plasmodium falciparum (Pf) can track antimalarial-resistant strains, to inform decision-making by National Malaria Control Programmes (NMCPs). The GenRe-Mekong project prospectively collected 5982 samples in the Greater Mekong Subregion (GMS) between 2017 and 2022, genotyping drug resistance markers, and barcodes that recapitulate genetic variation. Genotypes were analyzed with the grcMalaria R package, first described in this paper, to translate genetic epidemiology data into actionable visual information. Since 2020, Pf incidences decreased rapidly, accompanied by a d

Genetic Diversity and Phylogenetic Relatedness of Cameroonian Plasmodium falciparum Isolates and Comparative Analysis with Global Populations.

PURPOSE: Plasmodium falciparum (Pf) is the deadliest malaria parasite species, and its genetic diversity is a significant obstacle to its effective control. Here, we analyzed the genetic diversity and phylogeny of Pf isolates collected in Cameroon using merozoite surface proteins 1 and 2 (msp1/2) markers. METHODS: Samples were collected in three malaria epidemiological facets of three regions of Cameroon (Littoral, North, Far North). The msp1 block 2 and msp2 block 3 were genotyped using polymerase chain reaction and Sanger sequencing. The structural organization the msp1/2 allelic families (

Molecular detection and population genetic diversity analysis of Theileria ovis in partial regions of Xinjiang, China.

Ovine theileriosis is a disease caused by the genus Theileria (e.g., T. ovis, T. lestoquardi), preventing the sheep farming industry from developing, particularly in regions reliant on sheep for milk, meat, and associated economic benefits. However, there is limited information available on the epidemiological data and genetic diversity of T. ovis in Xinjiang. This study was conducted in May 2024 to investigate the molecular prevalence of T. ovis in sheep from five counties (Shaya, Wensu, Aketao, Keping, Awati) in Xinjiang. A total of 357 blood samples were screened for the presence of Theiler

Immunogenetic polymorphism of human platelet antigens in the Saudi population: Insights into genetic diversity and alloimmune risk.

BACKGROUND AND OBJECTIVES: Population study on immunogenetic polymorphism of human platelet antigens (HPAs) may aid in predicting the risk of alloimmune thrombocytopaenia syndromes. We aimed to characterize the diversity of HPAs in Saudis and estimate the alloimmunization risk. MATERIALS AND METHODS: We assessed the genotype and allele frequencies of HPA-1 to -11 and HPA-15 in 118 Saudi blood donors using a real-time polymerase chain reaction assay. We estimated the mismatch risk associated with blood transfusions and pregnancies for each individual allele. Furthermore, we evaluated the misma

Returning genetic risk information for hereditary cancers to participants in a population-based cohort study in Japan.

Large-scale population cohort studies that collect genomic information are tasked with returning an assessment of genetic risk for hereditary cancers to participants. While several studies have applied to return identified genetic risks to participants, comprehensive surveys of participants' understanding, feelings, and behaviors toward cancer risk remain to be conducted. Here, we report our experience and surveys of returning genetic risks to 100 carriers of pathogenic variants for hereditary cancers identified through whole genome sequencing of 50 000 individuals from the Tohoku Medical Mega

X-chromosome association study reveals genetic susceptibility loci of hypospadias in southern Chinese population.

PURPOSE: X-chromosome variants contribute significantly to hypospadias risk but have not been fully elucidated in the Chinese population. Here we aim to assess how X-chromosome variants contribute to hypospadias susceptibility in the Chinese population. METHODS: We recruited 1,073 boys with hypospadias and 5,150 controls in a southern Chinese population. Single-variant and gene/pathway-based association analyses were conducted for the distal and proximal hypospadias. Haplotype analysis was performed on top susceptibility genes. Additionally, we performed a multi-ancestral comparison between t

Genetic diversity and population structure of Oncomelania hupensis in Sichuan Province, China: implications for schistosomiasis control.

Schistosomiasis, caused by the infection with Schistosoma japonicum, remains a significant public health concern in China. As the sole intermediate host of S. japonicum, the breeding and spread of Oncomelania hupensis contribute significantly to the potential risk of disease occurrence and transmission. Exploring the population genetics of the snail vector is conducive to better understanding its distribution and dispersal patterns, and provides more data for future snail surveillance and control from a molecular perspective. The genetic diversity and population structure of O. hupensis in Sic

Unraveling the genetic association between autoimmune thyroid diseases and idiopathic inflammatory myopathies in the European population.

While clinical observations suggest potential links between thyroid autoimmunity disorders (specifically Hashimoto thyroiditis and Graves' disease) and idiopathic inflammatory myopathies (notably dermatomyositis and polymyositis), the underlying causative mechanisms remain incompletely understood. We implemented a Mendelian randomization approach to elucidate potential causal pathways through which thyroid autoimmunity might influence the development of idiopathic inflammatory myopathies. Comprehensive genetic association data regarding thyroid autoimmunity and idiopathic inflammatory myopathi

Detection and genetic diversity of Wolbachia and its associated prophage WO in mosquito populations from Ethiopia.

Arboviral diseases transmitted by mosquitoes, including dengue, Chikungunya, Zika, yellow fever, West Nile virus (WNV), and Rift Valley fever (RVF), pose a significant public health challenge globally, particularly impacting populations in low and middle-income countries. Conventional mosquito control methods, which primarily rely on insecticides, face critical challenges, including the development of insecticide resistance and environmental concerns. In this context, Wolbachia, an endosymbiotic bacterium, presents an alternative strategy due to its ability to manipulate mosquito reproduction

Genetic risk accentuates dietary effects on hepatic steatosis, inflammation and fibrosis in a population-based cohort.

BACKGROUND & AIMS: Steatotic liver disease (SLD), characterized by elevated liver fat content (LFC), is influenced by genetics and diet. However, whether diet has a differential effect based on genetic risk is not well-characterized. We aimed to determine how genetic factors interact with diet to affect SLD in a large national biobank. METHODS: We included UK Biobank participants with dietary intake measured by 24-hour recall and genotyping. The primary predictors were dietary pattern, PNPLA3-rs738409-G, TM6SF2-rs58542926-T, a 16-variant hepatic steatosis polygenic risk score (PRS), and gene-

Population Genetic Structure of the Rubber Tree Powdery Mildew Pathogen (Erysiphe quercicola) from China.

In order to manage agricultural pathogens, it is crucial to understand the population structure underlying epidemics. Rubber tree powdery mildew, caused by Erysiphe quercicola, is a serious threat to rubber plantations worldwide, especially in subtropical environments including all rubber tree-growing regions in China. However, the population structure of the pathogen is uncertain. In this study, 16 polymorphic microsatellite markers were used to genotype powdery mildew samples from the main rubber tree-growing regions including Yunnan (YN), Hainan (HN), western Guangdong (WG), and eastern Gua
