Periodontal disease immunology: 'double indemnity' in protecting the host.

During the last two to three decades our understanding of the immunobiology of periodontal disease has increased exponentially, both with respect to the microbial agents triggering the disease process and the molecular mechanisms of the host engagement maintaining homeostasis or leading to collateral tissue damage. These foundational scientific findings have laid the groundwork for translating cell phenotype, receptor engagement, intracellular signaling pathways and effector functions into a 'picture' of the periodontium as the host responds to the 'danger signals' of the microbial ecology to

An Integrative Computational Approach for the Prediction of Human-Plasmodium Protein-Protein Interactions.

Host-pathogen molecular cross-talks are critical in determining the pathophysiology of a specific infection. Most of these cross-talks are mediated via protein-protein interactions between the host and the pathogen (HP-PPI). Thus, it is essential to know how some pathogens interact with their hosts to understand the mechanism of infections. Malaria is a life-threatening disease caused by an obligate intracellular parasite belonging to the Plasmodium genus, of which P. falciparum is the most prevalent. Several previous studies predicted human-plasmodium protein-protein interactions using comput

The microbiota-host metabolic axis in cervical cancer: from homeostatic disruption to mechanisms of therapy resistance.

Persistent infection with high-risk human papillomavirus (HR-HPV) is a prerequisite for cervical carcinogenesis. In this process, the vaginal microbiota, as a key component of the cervical microenvironment, plays a significant role in facilitating viral persistence and malignant transformation. Moving beyond the conventional focus on compositional shifts, this review systematically examines the dynamic remodeling of the host-microbiota co-metabolic network during the progression from health to invasive cervical cancer. It explores in depth the interactive mechanisms through which metabolites d

Co-occurrence patterns of internal parasitic nematodes and coccidia in wild bighorn sheep depend on host geography and environment.

Heterogeneity of internal parasite communities amongst host animals is shaped by processes at multiple scales, including landscape distribution of parasites, individual host suitability, and parasite interactions within a host. As a result, pathogen co-occurrence patterns vary across space and time and among host populations. Often the challenges of wildlife sampling limit understanding of the drivers of pathogen community heterogeneity in host animals. In this study, we explore patterns and drivers of parasite community heterogeneity amongst wild bighorn sheep (Ovis canadensis) hosts over tim

Protein Interactions Network of Hepatitis E Virus RNA and Polymerase With Host Proteins.

Host-pathogen interactions are crucial for the successful propagation of pathogens inside the host cell. Knowledge of interactions between host proteins and viral proteins or viral RNA may provide clues for developing novel antiviral strategies. Hepatitis E virus (HEV), a water-borne pathogen that causes acute hepatitis in humans, is responsible for epidemics in developing countries. HEV pathology and molecular biology have been poorly explored due to the lack of efficient culture systems. A contemporary approach, to better understand the viral infection cycle at the molecular level, is the us…

Effector-host interactome map links type III secretion systems in healthy gut microbiomes to immune modulation.

Pseudomonadota (formerly Proteobacteria) are prevalent in the commensal human gut microbiota, but also include many pathogens that rely on secretion systems to support pathogenicity by injecting proteins into host cells. Here we show that 80% of Pseudomonadota from healthy gut microbiomes also have intact type III secretion systems (T3SS). Candidate effectors predicted by machine learning display sequence and structural features that are distinct from those of pathogen effectors. Towards a systems-level functional understanding, we experimentally constructed a protein-protein meta-interactome

Microdissection approaches in tuberculosis research.

Mycobacterium tuberculosis, a globally significant pathogen, results in active or latent tuberculosis. The granuloma is the characteristic lesion that offers insight into host-pathogen interactions in these distinct states. Microdissection provides a way to isolate and consequently investigate specific tissue sections. We review various techniques available and in use.

Mapping the social network: tracking lice in a wild primate (Microcebus rufus) population to infer social contacts and vector potential.

BACKGROUND: Studies of host-parasite interactions have the potential to provide insights into the ecology of both organisms involved. We monitored the movement of sucking lice (Lemurpediculus verruculosus), parasites that require direct host-host contact to be transferred, in their host population of wild mouse lemurs (Microcebus rufus). These lemurs live in the rainforests of Madagascar, are small (40 g), arboreal, nocturnal, solitary foraging primates for which data on population-wide interactions are difficult to obtain. We developed a simple, cost effective method exploiting the intimate r

A comprehensive two-hybrid analysis to explore the Legionella pneumophila effector-effector interactome.

UNLABELLED: Legionella pneumophila uses over 300 translocated effector proteins to rewire host cells during infection and create a replicative niche for intracellular growth. To date, several studies have identified L. pneumophila effectors that indirectly and directly regulate the activity of other effectors, providing an additional layer of regulatory complexity. Among these are "metaeffectors," a special class of effectors that regulate the activity of other effectors once inside the host. A defining feature of metaeffectors is direct, physical interaction with a target effector. Metaeffect

Recent Progress in Innate Immune Responses to Enterovirus A71 and Viral Evasion Strategies.

Enterovirus A71 (EV-A71) is a major pathogen causing hand, foot, and mouth disease (HFMD) in children worldwide. It can lead to severe gastrointestinal, pulmonary, and neurological complications. The innate immune system, which rapidly detects pathogens via pathogen-associated molecular patterns or pathogen-encoded effectors, serves as the first defensive line against EV-A71 infection. Concurrently, the virus has developed various sophisticated strategies to evade host antiviral responses and establish productive infection. Thus, the virus-host interactions and conflicts, as well as the abilit

Simultaneous profiling of sexually transmitted bacterial pathogens, microbiome, and concordant host response in cervical samples using whole transcriptome sequencing analysis.

Pelvic inflammatory disease (PID) is a female upper genital tract inflammatory disorder that arises after sexually transmitted bacterial infections (STI). Factors modulating risk for reproductive sequelae include co-infection, microbiota, host genetics and physiology. In a pilot study of cervical samples obtained from women at high risk for STIs, we examined the potential for unbiased characterization of host, pathogen and microbiome interactions using whole transcriptome sequencing analysis of ribosomal RNA-depleted total RNAs (Total RNA-Seq). Only samples from women with STI infection contai

Host-derived CEACAM-laden vesicles engage enterotoxigenic Escherichia coli for elimination and toxin neutralization.

Enterotoxigenic Escherichia coli (ETEC) cause hundreds of millions of diarrheal illnesses annually ranging from mildly symptomatic cases to severe, life-threatening cholera-like diarrhea. Although ETEC are associated with long-term sequelae including malnutrition, the acute diarrheal illness is largely self-limited. Recent studies indicate that in addition to causing diarrhea, the ETEC heat-labile toxin (LT) modulates the expression of many genes in intestinal epithelia, including carcinoembryonic cell adhesion molecules (CEACAMs) which ETEC exploit as receptors, enabling toxin delivery. Here,

Challenging Mycobacterium tuberculosis dormancy mechanisms and their immunodiagnostic potential.

Mycobacterium tuberculosis is the etiologic agent of tuberculosis, one of the world's greatest cause of morbidity and mortality due to infectious disease. Many evolutionary mechanisms have contributed to its high level of adaptation as a host pathogen. Prior to become dormant, a group of about 50 genes related to metabolic changes are transcribed by the DosR regulon, one of the most complex and important systems of host-pathogen interaction. This genetic mechanism allows the mycobacteria to persist during long time periods, establishing the so-called latent infection. Even in the presence of a

Basic Methods for Examining Neisseria gonorrhoeae Interactions with Host Cells In Vitro.

The obligate human pathogen Neisseria gonorrhoeae colonizes primarily the mucosal columnar epithelium of the male urethra and the female endocervix. In addition, gonococci can infect the anorectal, pharyngeal, and gingival mucosae and epithelial cells of the conjunctiva. More rarely, the organism can disseminate through the bloodstream, which can involve interactions with other host cell types, including blood vessel endothelial cells and innate immune cells such as dendritic cells, macrophages, and neutrophils. "Disseminated gonococcal infection" is a serious condition with various manifestat

Encrypting the message: Bacterium turns host RNA into a target.

Pathogenic bacteria are remarkably inventive when it comes to manipulating their hosts. In this issue of Cell Host & Microbe, McCaffrey et al. identify a previously uncharacterized bacterial effector from Legionella pneumophila that modifies not host proteins but messenger ribonucleic acid (mRNA) to dampen the host inflammatory response.

Non-coding RNAs in dermatophytosis: Key regulators of host-fungal interactions and pathogenesis-A review.

Dermatophytosis, a prevalent superficial fungal infection caused by fungi such as Trichophyton rubrum, Trichophyton mentagrophytes, and Microsporum canis, develops through the dynamic interaction between fungal virulence factors - including adhesion, secretion of keratinolytic proteases, and hyphal invasion - and the host's multifaceted immune response, which involves antimicrobial peptides, pattern recognition receptors, and cellular immunity. Recent evidence identifies non-coding RNAs (ncRNAs), including miRNAs, lncRNAs, circRNAs, and tRFs, as key regulators of this host-fungal crosstalk. Th

Immune Memory-like Responses of Akoya Pearl Oyster to Pinctada Birnavirus.

Among accumulating knowledge of invertebrate immune-like responses, antiviral mechanisms in mollusks remain poorly understood. Pinctada birnavirus (PiBV) infects the mantle epithelial cells of the pearl oyster (Pinctada fucata), causing mass mortality of juveniles during high-temperature periods. Here, we examined immune memory-like responses of pearl oysters to PiBV reinfection. At a high temperature (24-25 °C), experimental infection caused ~80% mortality, whereas mortality remained <30% and did not differ significantly from uninfected controls at a lower temperature (18-20 °C). Juveniles th

Breaking Through to the Other Side: How Plant Viruses Modify Plasmodesmata and the Plant Cell Wall.

Viruses are obligate intracellular parasites that rely on their hosts for the cellular machinery necessary for their replication and transmission to new hosts. Viruses are therefore invaluable for illuminating numerous facets of their hosts' biology. Plant viruses take advantage of the cell-to-cell connectivity provided by plasmodesmata for their movement from sites of replication to the vasculature for eventual systemic dissemination in their plant hosts, a strategy distinct from viruses that use animal and fungal hosts. Given their reliance on plasmodesmata for successful infection and sprea
