Sertoli Cell-Only Syndrome: Behind the Genetic Scenes.

Sertoli cell-only syndrome is defined by the complete absence of germ cells in testicular tissues and always results in male infertility. The aetiology often remains unknown. In this paper, we have investigated possible causes of Sertoli cell-only syndrome with a special focus on genetic causes. Our results show that, for a large part of the patients (>23% in an unselected group), the sex chromosomes are involved. The majority of patients had a Klinefelter syndrome, followed by patients with Yq microdeletions. Array comparative genomic hybridization in a selected group of "idiopathic patients"

Williams-Beuren syndrome with brain malformation and hypertrophic cardiomyopathy.

Williams-Beuren syndrome (WBS) is a multisystemic genetic disorder caused by a contiguous gene deletion at 7q11.23. We report a severely affected WBS patient with cerebral and cerebellar dysplasia as well as hypertrophic cardiomyopathy. Microarray comparative genomic hybridization (aCGH) detected a deletion on 7q11.23 expanding from RP11-614D7 to RP11-137E8, which is a typical deletion in WBS. To the best of our knowledge, this is the first case report of a WBS patient with severe congenital central nervous system anomaly and progressive hypertrophic cardiomyopathy. The relationship between th

All males do not have 46 xy karyotype: A rare case report.

The sex of an embryo is determined by genetic sex due to presence or absence of Y chromosome, but it may not be true in all. We hereby report an interesting case of a phenotypic male carrying a female karyotype (46 XX). A 26-year-old male presented with bilateral gynecomastia, poor development of secondary sexual characters and azospermia. On evaluation patient had hypergonadotrophic hypogonadism and chromosomal analysis revealed 46 XX karyotype. The ultrasound revealed no Mullerian structures. Fluorescent in situ hybridization (FISH) showed sex determining region of Y chromosome (SRY) gene lo

Left-sided gastroschisis with placenta findings: case report and literature review.

Gastroschisis is a congenital abdominal-wall defect that typically occurs to the right of the umbilicus. Only twenty-one cases of left-sided gastroschisis have been described in the literature. Here we report a large left-sided gastroschisis with pulmonary hypoplasia, scoliosis, ventricular septal defect and absence of gallbladder. Section of placental membranes revealed vacuolization of the amnion, without increased macrophage infiltration of the chorion. Postmortem comparative genomic hybridization micro array did not identify a specific genetic abnormality. Some of the previously reported c

Whole-mount in situ hybridization for analysis of gene expression during Aedes aegypti development.

Blood-feeding mosquitoes, including the dengue and yellow fever vector Aedes aegypti, transmit many of the world's deadliest diseases. Such diseases have resurged in developing countries and pose clear threats for epidemic outbreaks in developed countries. Recent mosquito genome projects have stimulated interest in the potential for arthropod-borne disease control by genetic manipulation of vector insects, and genes that regulate development are of particular interest. This protocol for whole-mount in situ hybridization can be used to analyze gene expression in Ae. aegypti embryos and larvae,

Monozygotic dichorionic twins heterokaryotypic for duplication chromosome 2q13-q23.3.

We present an evaluation of the diagnosis, management and outcome of a pair of heterokaryotypic monozygotic dichorionic twins. The heterokaryotype was an incidental finding from an amniocentesis performed for prenatal diagnosis of beta-thalassaemia major in a pair of dichorionic twins. Monozygocity was revealed by QF-PCR showing identical short tandem repeat markers on chromosomes 21, 18, 13, X and Y. The twins were heterokaryotypic for duplication chromosome 2q13-q23.3, as shown by array comparative genomic hybridization. Selective foeticide was performed. This case demonstrates that heteroka

Gallbladder cancer in a patient with Miller-Dieker syndrome.

UNLABELLED: We report the case of a male patient with Miller-Dieker syndrome (MDS) and gallbladder cancer. Chromosome analysis by fluorescence in situ hybridization revealed a deletion in the 17p13.3 region, an area thought to contain tumour suppressor genes, including the hypermethylated in cancer 1 gene. Considering the rarity of gallbladder cancer in children, we propose that MDS as the genetic background of this patient may have played a role in the occurrence of gallbladder cancer. CONCLUSION: Our present report indicates that the emergence of cancers should be taken into consideration d

Histologically benign metastatic meningioma: morphological and cytogenetic study. Case report.

The authors report on a 75-year-old man with histologically benign fibroblastic meningioma metastasizing to the lung, liver, spleen, and kidney. The original tumor exhibited a complex karyotype involving different structural and numerical anomalies associated with monosomy of chromosome 22. The implication of chromosome 1p36 was confirmed by fluorescence in situ hybridization in most interphase nuclei. Metastases occurred 4 months after incomplete resection with prior therapeutic embolization. The recurrent tumor in turn displayed anaplastic features and an increased Ki-67 labeling index. Gene

A mutation in a case of early onset narcolepsy and a generalized absence of hypocretin peptides in human narcoleptic brains.

We explored the role of hypocretins in human narcolepsy through histopathology of six narcolepsy brains and mutation screening of Hcrt, Hcrtr1 and Hcrtr2 in 74 patients of various human leukocyte antigen and family history status. One Hcrt mutation, impairing peptide trafficking and processing, was found in a single case with early onset narcolepsy. In situ hybridization of the perifornical area and peptide radioimmunoassays indicated global loss of hypocretins, without gliosis or signs of inflammation in all human cases examined. Although hypocretin loci do not contribute significantly to gen

[New diagnostic possibilities in dermatologic diseases].

The author reviews the latest developments regarding the most important immunological methods for the diagnosis of dermatological diseases. The salt-split-skin, fluorescence overlay antigen mapping and immunoblot techniques are useful methods for the differentiation of autoimmune bullous skin diseases. The use of recombinant fusion products of antigenic epitopes in patients with pemphigus and bullous pemphigoid affords a possibility for the differentiation or diagnosis of the disease by means of a simple ELISA technique. Polymorphic microsatellite markers demonstrate hybridization and interspecific gene flow between lumbricid earthworm species, Eisenia andrei and E. fetida.

The lumbricid earthworms Eisenia andrei (Ea) and E. fetida (Ef) have been used as model organisms for studies on hybridization. Previously they have been identified by species specific sequences of the mitochondrial COI gene of maternal origin ('a' or 'f') and the nuclear 28S gene of maternal/paternal origin ('A' or 'F'). In experimental crosses, these hermaphroditic species produce progeny of genotypes Ea (aAA), Ef (fFF) and hybrids (aAF and fFA) originating by self-fertilization or cross-fertilization. To facilitate studies on new aspects of the breeding biology and hybridization of earthwor

Metabolite profiling characterises chemotypes of Musa diploids and triploids at juvenile and pre-flowering growth stages.

Bananas (Musa spp.) are consumed worldwide as dessert and cooking types. Edible banana varieties are for the most part seedless and sterile and therefore vegetatively propagated. This confers difficulties for breeding approaches against pressing biotic and abiotic threats and for the nutritional enhancement of banana pulp. A panel of banana accessions, representative of the diversity of wild and cultivated bananas, was analysed to assess the range of chemotypes available globally. The focus of this assessment was banana leaves at two growth stages (juvenile and pre-flowering), to see when duri

[Studies of F1 of transgenic allotetraploid hybrids of Carassius auratus red var. (Female) x Cyprinus carpio (Male)].

The tetraploid fish has been developed by assortative breeding the hybrids of Carassius auratus red var. (Female) x cyprinus carpio (Male), which has the stable genetic characters and can reproduce themselves. An "all-fish" recombinant DNA construct (pCAgcGHc) containing common carp beta-actin gene promoter and cDNA for grass carp (Ctenopharyngodon idella) growth hormone gene was introduced into fertilized eggs of the allotetroploid fish through microinjection as soon as artificial insemination was done. Artificial insemination was carried out between the female and the male transgenic allotet

Bridging markers defining the map position of X linked hypophosphataemic rickets.

Hypophosphataemic rickets is commonly an X linked dominant hereditary disorder associated with a renal tubular defect in phosphate transport and bone deformities. The gene causing this disorder has been mapped to Xp22.31----p21.3 by using cloned human X chromosome sequences identifying restriction fragment length polymorphisms (RFLPs) in linkage studies of affected families. The hypophosphataemic rickets gene locus (HPDR) was previously mapped distal to the X linked polymorphic locus DXS41 (99.6) but its position in relation to the distal loci DXS43 (D2) and DXS85 (782) was not established. Crossing selected lines vs. selecting from a cross in mice.

Eight generations of selection were practiced for post-weaning gain in body weight and for litter size at birth in two outbred stocks of mice and in a stock derived by crossing these outbreds. At each generation, the selected outbred stocks were crossed, and offspring of the cross compared to those from selection in the cross stock. Selection from the cross stock gave greater response than crossing selected stocks in post-weaning gain, a trait where inheritance was primarily by additive gene action. For litter size, a trait where dominance and epistatic interactions were of greater importance,

Comparative transcriptome analysis reveals putative transcription factors components of the regulatory network for cannabidiol biosynthesis in Cannabis sativa L.

BACKGROUND: Cannabidiol (CBD), a unique secondary metabolite of Cannabis sativa L. (cannabis), is widely used in medicine, food, cosmetics, and forage. Elucidation of the transcriptional regulation mechanism can provide a theoretical basis for molecular breeding with higher CBD content. However, the regulatory network for cannabidiol biosynthesis in cannabis is largely unknown. METHODS: Transcriptome analysis of four cannabis tissues was conducted to screen differentially expressed genes. Short time-series expression miner (STEM) analysis combined with strain-specific expression correlation a

Comparative nutritional analysis of newly developed interspecific citrus hybrids: multivariate insights for crop improvement.

Enhancing the quality and nutrient composition of citrus fruits is essential for ensuring food and nutritional security. In the present study, twenty-four newly developed interspecific citrus scion hybrids (16 Orangelos and 8 Tangelos) from 'Pummelo' × 'Mosambi' sweet orange and 'Pummelo' × 'Dancy' tangerine crosses, along with their parental genotypes, were evaluated for fruit physico-chemical attributes, bioactive composition, and mineral nutrient profiles. Significant variation was observed due to heterozygosity-driven transgressive segregation in the interspecific hybrid progenies. Fruit p

Multitarget Fluorescence In Situ Hybridization Diagnostic Applications in Tumors.

Multitarget fluorescence in situ hybridization (mFISH) is a technique that allows the detection of multiple target sequences on the same sample using spectrally distinct fluorophore labels. The mFISH approach is currently a useful assay in the oncologic field for the detection of predictive, prognostic, and diagnostic biomarkers. In this chapter, we summarize the application of mFISH in the identification of target genetic aberrations in formalin-fixed, paraffin-embedded (FFPE) tissue samples of several tumor types. We discuss the mFISH protocols in FFPE samples, the innovative multitarget pro

Clinical Manifestations of 22q11.2 Deletion Syndrome.

DiGeorge syndrome (DGS), also known as "22q11.2 deletion syndrome" (22q11DS) (MIM # 192430 # 188400), is a genetic disorder caused by hemizygous microdeletion of the long arm of chromosome 22. In the last decades, the introduction of fluorescence in situ hybridization assays, and in selected cases the use of multiplex ligation-dependent probe amplification, has allowed the detection of chromosomal microdeletions that could not be previously identified using standard karyotype analysis. The aim of this review is to address cardiovascular and systemic involvement in children with DGS, provide ge

Twin anemia-polycythemia sequence with blood chimerism in monochorionic dizygotic opposite-sex twins.

We report a case of twin anemia-polycythemia sequence with blood chimerism in monochorionic dizygotic twins born to a 30-year-old woman who conceived via ovulation induction. The neonates developed twin anemia-polycythemia sequence; the female twin had anemia and the male had polycythemia. We detected blood chimerism using fluorescence in situ hybridization (FISH). Twin anemia-polycythemia sequence carries not only perinatal risks, but also genetic and immunological implications due to blood chimerism. Although previous reports have described twin-to-twin transfusion syndrome in monochorionic
