From Farms to Pharma: A "Natural" History of Vaccine Production and Vaccine Skepticism.

In the era of synthetic biology, vaccine skeptics have made claims that vaccines are "unnatural," that the technology used to develop them is risky and untested, and that "naturally acquired" immunity is superior to vaccination. Public health practitioners and physicians alike have attempted to respond to these concerns by reminding patients and the public that vaccines generate a "natural" immune response. These negotiations over the language to describe vaccines are nothing new. This article puts the relationship between vaccines and concepts like "nature" and "natural" in historical perspec

Transplantation in miniature swine. VI. Factors influencing survival of renal allografts.

Renal allografts were performed between and among animals from three herds of miniature swine that were selectively inbred to homozygosity at the major histocompatibility complex, MSLA. The results suggest several genetic factors which influence the survival of renal allografts in these animals. As expected, the major histocompatibility complex (MHC) was of dominant importance, and all MSLA-mismatched grafts were rejected promptly (12 +/- 3.7 days). Some MSLA-matched grafts were also rejected (30 +/- 15.0 days), indicating that non-MSLA loci also determine antigens which can lead to kidney rej

Effectiveness of a BHV-1/BEFV bivalent vaccine against bovine herpesvirus type 1 infection in cattle.

Bovine herpesvirus type 1 (BHV-1) causes acute febrile respiratory diseases (infectious bovine rhinotracheitis, IBR), decreased milk production, weight loss and abortion. Bovine ephemeral fever virus (BEFV) causes acute febrile respiratory disease, with pulmonary emphysema and pulmonary edema as the main signs. These viruses infect domesticated herds and lead to significant economic losses. In our previous study, an inactivated BHV-1 and BEFV bivalent vaccine was formulated with water-in-oil-in-water adjuvant, and vaccine efficacy was evaluated in guinea pigs. In this study, we evaluated the e

Changes in childhood vaccine hesitancy during the COVID-19 pandemic: a retrospective study based on data from Turkey.

BACKGROUND: Vaccine refusal is a growing public health concern, threatening herd immunity and the control of vaccine-preventable diseases. The COVID-19 pandemic has influenced healthcare delivery and vaccine refusal behavior as a proxy for vaccine hesitancy. This study aimed to assess changes in childhood vaccine refusal behavior during the pandemic in Turkey. METHODS: We conducted a retrospective observational study including 2,786 children born between 2017 and 2022 who reached 24 months of age and had complete vaccination records from primary healthcare. Children were categorized as pre-pan

Moving Beyond the Female-Only Paradigm: Arguments for Accelerating the Global Gender-Neutral Human Papillomavirus Vaccination.

Since its adoption into public health programmes in 2006, the Human Papillomavirus (HPV) vaccine has proven its powerful effect in preventing cervical cancer, the most prevalent HPV-associated cancer. Nevertheless, a considerable burden of HPV infection is associated with non-cervical cancers and conditions that, due to a historically gendered narrative around HPV vaccination, are still partially neglected in most preventive agendas. Gender-neutral vaccination (GNV) programmes have been steadily adopted in an increasing number of countries, but global endorsement of this strategy is still bein

A mathematical model for Zika virus disease: Intervention methods and control of affected pregnancies.

Zika virus is spread to human populations primarily by Aedes aegypti mosquitoes, and Zika virus disease has been linked to a number of developmental abnormalities and miscarriages, generally coinciding with infection during early pregnancy. In this paper, we propose a new mathematical model for the transmission of Zika and study a range of control strategies to reduce the incidence of affected pregnancies in an outbreak. While most infectious disease models primarily focus on measures of the spread of the disease, our model is formulated to estimate the number of affected pregnancies throughou

Optimization of a bovine whole-blood cytokine recall assay for the detection of interferon-gamma and interleukin 10 following stimulation with Coxiella burnetii antigens.

The intracellular bacterium, Coxiella burnetii, causes Q fever in humans and coxiellosis in animals. Cell-mediated immunity (CMI) has been shown to be important in Q fever; however, few studies have investigated the role of CMI in coxiellosis in cattle. Therefore, we aimed to optimize a whole-blood cytokine recall assay (CRA) and to evaluate C. burnetii-specific interferon-gamma (IFNγ) and interleukin 10 (IL10) responses in naturally infected cattle. Duplicate blood samples were collected from cows in an endemically infected dairy herd (as determined by serologic and molecular testing as well

A regional genomic surveillance program is implemented to monitor the occurrence and emergence of SARS-CoV-2 variants in Yubei District, China.

BACKGROUND: In December 2022, Chongqing experienced a significant surge in coronavirus disease 2019 (COVID-19) epidemic after adjusting control measures in China. Given the widespread immunization of the population with the BA.5 variant, it is crucial to actively monitor severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) variant evolution in Chongqing's Yubei district. METHODS: In this retrospective study based on whole genome sequencing, we collected oropharyngeal and nasal swab of native COVID-19 cases from Yubei district between January to May 2023, along with imported cases from

Potent neutralization by a receptor binding domain monoclonal antibody with broad specificity for SARS-CoV-2 JN.1 and other variants.

SARS-CoV-2 continues to be a public health burden, driven in-part by its continued antigenic diversification and resulting emergence of new variants. While increasing herd immunity, current vaccines, and therapeutics have improved outcomes for some; prophylactic and treatment interventions that are not compromised by viral evolution of the Spike protein are still needed. Using a rationally designed SARS-CoV-2 Receptor Binding Domain (RBD) - ACE2 fusion protein and differential selection process with native Omicron RBD protein, we developed a recombinant human monoclonal antibody (hmAb) from a

Attitudes towards Receiving Monkeypox Vaccination: A Systematic Review and Meta-Analysis.

BACKGROUND: The public's attitude towards Mpox vaccination is a critical factor in the success of immunisation programmes. Understanding the factors contributing to vaccine acceptance or hesitancy is critical for developing effective health communication strategies. This systematic review and meta-analysis aims to bring together evidence from observational studies on attitudes towards Mpox vaccination, including willingness and rejection. METHODS: From this review's inception until June 2023, a comprehensive search was conducted across four major electronic databases: PubMed, Web of Science,

Does poverty increase COVID-19 in Africa? A cross-country analysis.

BACKGROUND: Most economies in African countries are informal. As such, households in these countries tend to face higher levels of informality coupled with a lack of social protection, and have no replacement income or savings in the event of unexpected external shocks, such as COVID-19. Thus, the COVID-19 shock and its negative economic effects triggered a cascade of income losses and bankruptcies that pushed a significant share of households in African countries into poverty. This research analyzes the effect of poverty on the spread of COVID-19 using a sample of 52 African countries. METHO

Pneumococcal serotypes causing non-invasive pneumonia in adults from a South Indian tertiary care hospital and the impact of the newer conjugate vaccines.

BACKGROUND: Streptococcus pneumoniae is the leading cause of community-acquired pneumonia (CAP) in adults. Ageing, chronic conditions and comorbidities are important risk factors for pneumococcal pneumonia. PURPOSE: There is lack of data on the pneumococcal serotypes causing non-invasive pneumonia in India. This study aims to determine the prevalent pneumococcal serotypes causing non-invasive pneumonia, the associated comorbidities, and the coverage of both the available pneumococcal vaccines in India and conjugate vaccines that are currently undergoing clinical trials. METHODS: A total of 2

No Such Thing as a Free-Rider? Understanding Drivers of Childhood and Adult Vaccination through a Multicountry Discrete Choice Experiment.

Increased vaccine hesitancy and refusal negatively affects vaccine uptake, leading to the reemergence of vaccine preventable diseases. We aim to quantify the relative importance of factors people consider when making vaccine decisions for themselves, or for their child, with specific attention for underlying motives arising from context, such as required effort (accessibility) and opportunism (free riding on herd immunity). We documented attitudes towards vaccination and performed a discrete choice experiment in 4802 respondents in The United Kingdom, France and Belgium, eliciting preferences

Motivating Seasonal Influenza Vaccination and Cross-Promoting COVID-19 Vaccination: An Audience Segmentation Study among University Students.

In the response to the coronavirus pandemic, much attention has been invested in promoting COVID-19 vaccination. However, the impact of seasonal influenza should not be neglected, particularly during the winter influenza surge. Currently, most influenza vaccination campaigns target at healthcare workers or high-risk population groups, while COVID-19 vaccination programmes are targeting the whole population as a single homogeneous group. There is limited research on the promotion of influenza vaccination for university students who study, live, and socialise in close contact with a large variet

COVID-19 in Children: Where do we Stand?

From the beginning of the coronavirus disease 2019 (COVID-19) pandemic it became evident that children infected with the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) remain mostly asymptomatic or mildly symptomatic. We reviewed the epidemiologic and clinical features of children with SARS-CoV-2 infection. The true prevalence of asymptomatic SARS-CoV-2 infection is most likely underestimated, as asymptomatic children are less frequently tested. Serologic surveys indicate that half of children tested positive for SARS-CoV-2 report no symptoms. Anosmia/ageusia is not frequent in c

COVID-19 mRNA vaccination leading to CNS inflammation: a case series.

The availability of vaccines against the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), provides hope towards mitigation of the coronavirus disease 2019 (COVID-19) pandemic. Vaccine safety and efficacy has not been established in individuals with chronic autoimmune diseases such as multiple sclerosis (MS). Anecdotal reports suggest that the vaccines may be associated with brain, spinal cord, peripheral nervous system, and cardiac inflammation. Based on the high morbidity and unpredictable course of COVID-19, and the need to achieve herd immunity, vaccination has been recommended

Therapeutically administered ribonucleoside analogue MK-4482/EIDD-2801 blocks SARS-CoV-2 transmission in ferrets.

The coronavirus disease 2019 (COVID-19) pandemic is having a catastrophic impact on human health1. Widespread community transmission has triggered stringent distancing measures with severe socio-economic consequences. Gaining control of the pandemic will depend on the interruption of transmission chains until vaccine-induced or naturally acquired protective herd immunity arises. However, approved antiviral treatments such as remdesivir and reconvalescent serum cannot be delivered orally2,3, making them poorly suitable for transmission control. We previously reported the development of an orall
