Antibody-drug conjugates (ADC) offer a targeted cancer treatment approach by delivering cytotoxic payloads directly to tumor cells. However, resistance mechanisms, poor tumor penetration, and off-target toxicity often limit clinical efficacy. Vartumab targets oncofetal chondroitin sulfate (ofCS), a pan-cancer target present on tumor cells and in the malignant stroma, with low expression in normal tissues. As part of transitioning Vartumab to clinical evaluation, two linker-payloads known to mediate bystander effects, valine-citrulline (vc)-monomethyl auristatin E (MMAE) and glycine-glycine-phe

Compared with chimeric antigen receptor (CAR) T cell therapy, antibody-drug conjugates (ADCs) offer distinct advantages. Here, we report on two FGFR4-targeted ADCs, using the high-affinity monoclonal antibody 3A11, the same binder used in a CAR T cell format that is being evaluated at the NCI (NCT06865664) for patients with relapsed/refractory rhabdomyosarcoma (RMS). These ADCs, conjugated to monomethyl auristatin E (MMAE) or an exatecan derivative, are rapidly internalized, causing potent fibroblast growth factor receptor 4 (FGFR4)-dependent cytotoxicity in vitro. In subcutaneous RMS xenograf

Effective treatment for metastatic cancer has remained elusive because of the persistence of drug-resistant metastasis stem cells (MetSC) that drive relapse. MetSCs are tumor cell subpopulations enriched for their ability to reinitiate and sustain metastatic growth, displaying phenotypic plasticity and resistance to chemotherapy. These cells express the L1 cell adhesion molecule (L1CAM), a transmembrane protein detected in numerous human solid tumor types and at multiple disseminated organ sites. As a selective surface marker of MetSCs, L1CAM is a promising candidate for molecularly targeted d

In this work, we present a near-infrared (NIR) phospha-rhodamine dye POR-1, featuring a thiomorpholine 1,1-dioxide auxochrome and a carboxyl handle for constructing dye-antibody conjugates for tumor imaging. The unique auxochrome endows the dye not only with markedly improved quantum yields due to the strong inhibition of nonradiative decay, but also with high hydrophilicity without introduction of the multiple charged groups. Moreover, the dye exhibits high photostability and chemical stability alongside strong resistance to self-aggregation in aqueous solution. All of these properties are hi

PURPOSE: Antibody-drug conjugates (ADC) have transformed the treatment of breast cancer. FDA-approved ADC such as trastuzumab deruxtecan (T-DXd), sacituzumab govitecan (SG), and datopotamab deruxtecan (Dato-DXd) have demonstrated clinical benefit; however, drug resistance will almost inevitably emerge in the metastatic setting, and there is no established strategy for selecting subsequent ADC after disease progression following the first exposure to the ADC. This study aimed to define mechanisms of acquired resistance and evaluate rational sequencing approaches.
EXPERIMENTAL DESIGN: We genera

In this issue of Cancer Discovery, Chen and colleagues demonstrate that, in preclinical models, HER2 expression level directly affects trastuzumab deruxtecan internalization and cytotoxicity, with clinical data revealing divergent target dynamics depending on whether HER2 functions as an oncogenic driver or a dispensable antigen. Together with prior preclinical and clinical evidence, these findings support a context-dependent model in which target downregulation predominates in HER2-low disease, whereas payload resistance or rare binding-site mutations may dominate resistance in HER2-addicted

Antibody-drug conjugates (ADCs) have revolutionized breast cancer therapy. HER2 is the only validated biomarker guiding ADC therapy in breast cancer. However, their clinical benefit remains confined to HER2- and TROP2-targeted therapies. Tumor heterogeneity and acquired resistance often limit durable responses, underscoring the need for novel, tumor-specific ADC targets. CD44 variant isoform 9 (CD44v9), a splice variant of the CD44 family, is largely absent in normal tissues but enriched in aggressive breast cancers, where it contributes to stemness, redox regulation, and therapy resistance. Recurrent or metastatic head and neck squamous cell carcinoma (R/M HNSCC) is a highly aggressive disease entity with poor prognosis. Current systemic treatment regimens, including chemotherapy, targeted therapy, and immune checkpoint inhibitors, provide only limited and often short-lived clinical benefits. As an innovative therapeutic approach, antibody-drug conjugates (ADCs) combine the tumor-targeting specificity of monoclonal antibodies with the potent cytotoxicity of the conjugated payload, enabling selective delivery of highly active drugs to cancer cells. Given the urgent need to improve

BACKGROUND: The immunologically "cold" microenvironment of triple-negative breast cancer presents a major obstacle to treatment, conferring resistance to current immunotherapies. Targeting the stimulator of interferon genes (STING) pathway has emerged as a promising strategy to enhance immunotherapy efficacy.
METHODS: In this preclinical study, we leveraged a novel integrated approach by employing a B7-H3-targeted dual-payload antibody-drug conjugate (ADC) to induce innate immune responses and reprogram the tumor microenvironment.
RESULTS: Our preliminary results indicated that the dual-payl

BACKGROUND: Orbital metastases are rare in patients with breast cancer. However, medical management of orbital metastases is limited by the inability of treatment options to penetrate the blood-brain barrier. In patients with triple negative breast cancer (TNBC), these treatment options are further limited. Sacituzumab govitecan, an antibody-drug conjugate, has emerged as a promising agent for metastatic TNBC. However, to date, patients with central nervous system (CNS) disease have been excluded from corresponding clinical trials, making the efficacy of sacituzumab govitecan in patients with

Mirvetuximab soravtansine (MIRV) is an antibody-drug conjugate (ADC) composed of the DM4 payload conjugated to a folate receptor α (FRα)-targeting antibody via the cleavable sulfo-SPDB linker. MIRV targets and binds to FRα with high affinity and specificity, releasing the DM4 payload intracellularly following MIRV-FRα complex internalization and degradation. DM4 and its metabolite S-methyl-DM4 suppress microtubule dynamic instability, which triggers cell cycle arrest and apoptosis. Selective FRα-overexpression in ≥ 90% of epithelial ovarian tumor cells and its ability to internalize large mole

Antibody-based immunotherapy targeting amyloid-β (Aβ) is a promising approach for Alzheimer's disease (AD). However, its efficacy is limited by rapid hepatic sequestration, complement activation, and liver dysfunction. In this study, we synthesized low-immunogenic 450-nm functionalized mesoporous silica nanoparticles (PEG-MSN-1F12) by conjugating the anti-Aβ42 monoclonal antibody 1F12 to polyethylene glycol-modified mesoporous silica nanoparticles to address these challenges. In APP/PS1 mice, intravenous PEG-MSN-1F12 administration markedly enhanced peripheral Aβ clearance, promoted intestinal

BACKGROUND: The Trop-2-directed antibody-drug conjugate sacituzumab govitecan (SG) has shown significant benefit in metastatic triple-negative breast cancer (mTNBC) in clinical trials. However, evidence about its effectiveness and safety in real-world heterogeneous patient populations is limited. This multicenter study, including the largest European and second-largest worldwide real-world cohort of SG-treated patients with mTNBC, offers valuable insights for its use in routine clinical practice.
MATERIALS AND METHODS: This analysis included 256 women with mTNBC treated with SG across 17 Ital

Nectin-4-targeted imaging has emerged as a promising strategy for patient stratification in urothelial carcinoma, yet currently available radiotracers remain limited by rapid renal clearance, urinary background activity, or potential pharmacologic confounding associated with antibody-drug conjugate (ADC)-based constructs. To overcome some of these limitations, we developed a payload-free immunoPET radiotracer based on the native enfortumab antibody conjugated with the bifunctional chelator DFO*-sq and radiolabeled with zirconium-89 (89Zr). The resulting [89Zr]Zr-DFO*-sq-Enfortumab preserved Ne

Aberrant expression of claudin18.2 (CLDN18.2) has frequently been observed in gastric and gastroesophageal junction (G/GEJ) adenocarcinoma, making it a promising therapeutic target for this aggressive cancer. While a monoclonal antibody targeting CLDN18.2 has been approved for G/GEJ adenocarcinoma, antibody-drug conjugates (ADCs) have also emerged as therapeutic modalities. IBI343 is an ADC consisting of a fully humanized anti-CLDN18.2 monoclonal antibody conjugated to exatecan via site-specific glycol conjugation and a cleavable linker with a drug-to-antibody ratio of 4. Targeting molecules, such as antibodies and peptides, play a key role in the precise delivery of cytotoxic payloads to tumor sites by binding to specific tumor-associated antigens or other proteins within the tumor microenvironment. This investigation evaluates the potential therapeutic application of a bispecific antibody (BsAb), which simultaneously targets EphA2, a tumor-associated antigen, and CD11b, a protein expressed by tumor-associated macrophages and myeloid-derived suppressor cells (TAMCs). Recombinantly produced anti-EphA2-CD11b-BsAb was conjugated to a bifunctional chelator, NOTA-S
