Centrally applied histamine increases gastric acid secretion in rats.

Administration of 10-100 micrograms of histamine into the lateral cerebral ventricle of anaesthetized rats stimulated gastric acid secretion in a dose-dependent manner, while subcutaneous (s.c.) injections of the same doses produced clearly less pronounced increases in the acid output. In vagotomized rats only a marginal response to histamine given into the lateral ventricle was observed. When injected into the third cerebral ventricle the doses of histamine needed for the stimulation of gastric acid secretion were 1-10 micrograms, the effect being totally abolished by vagotomy. The results in

Centrally mediated opioid induced depression of hepatic glutathione: effects of intracerebroventricular administration of mu, kappa, sigma and delta agonists.

It has recently been demonstrated that morphine produces a loss of hepatocellular glutathione in mice by virtue of its action within the central nervous system. The ability of opioid receptor antagonists to abolish morphine's effect on hepatic glutathione suggests that this action is opioid-receptor mediated. The involvement of opioid receptors in this phenomenon is confirmed in the present study in mice by the ability of naltrexone, 100 micrograms administered intracerebroventricularly (i.c.v.), to completely block the decrease in hepatic glutathione induced by an i.c.v. injection of 100 micr

Increased central cholinergic activity in rat models of hypertension.

The role of brain acetylcholine (ACh) in the development and maintenance of experimental hypertension was evaluated. Single, acute injections of hemicholinium-3 (HC-3), 20 micrograms, were made into the lateral cerebral ventricle of unanesthetized rats during development of hypertension in the spontaneously hypertensive rat (SHR) or following induction of deoxycorticosterone-salt (DOCA), aortic coarctation (AoCo), and Grollman hypertension. HC-3 caused nonsignificant reductions in blood pressure (BP) when injected in SHR and Wistar-Kyoto (WKY) controls at 5 and 8 weeks of age. At 12 weeks in S

Intraventricular interferon in a case of subacute sclerosing panencephalitis.

A patient with subacute sclerosing panencephalitis (SSPE) was treated by direct administration of Interferon B into the cerebral ventricles. Immediately after the first dose intrathecal synthesis of IgG and the CNS antimeasles antibody titer fell significantly, only to rise again to the pretreatment levels 2 months after discontinuation of treatment. There was no improvement in the patient's clinical status.

Pressor responses to central hypertonic NaCl stimulation in conscious turkeys.

The purpose of this investigation was to examine the possible involvement of the central renin-angiotensin system in the pressor response to the intracerebroventricular (icv) injection of hypertonic NaCl in conscious turkeys. The icv injection was accomplished via a stereotaxically implanted stainless steel guide cannula in the lateral cerebral ventricle. The arterial blood pressure (AP) of the turkey was measured by means of a PE catheter in the left brachial artery. The icv administration of hypertonic NaCl caused a dose-dependent increase of AP. The mean AP increases due to 10-microliter ic

Behavioral characteristics of centrally administered adenosine analogs.

Mice were implanted with chronic indwelling cannulae in the lateral cerebral ventricle. A series of adenosine analogs and related compounds were injected into the lateral ventricle (ICVT) and their effects on spontaneous locomotor activity recorded. All analogs produced dose-related decreases in locomotor activity. 5'-N6-ethyl-carboxamidoadenosine (NECA) was the most potent compound tested, with a number of N6-substituted analogs also being effective depressants of activity. Caffeine, administered either intracerebroventricularly or intraperitoneally, antagonized the depressant effects of the

The role of the capsaicin-sensitive innervation of the rat urinary bladder in the activation of micturition reflex.

Capsaicin applied on the serosal surface of the urinary bladder in urethane-anaesthetized rats produces two distinct types of motor effects: a tetrodotoxin-, hexamethonium- and lidocaine-insensitive 'tonic' contraction and a series of tetrodotoxin-, hexamethonium- and lidocaine-sensitive rhythmic contractions. Both 'tonic' and rhythmic contractions are abolished by bladder denervation indicating their neurogenic origin. The rhythmic but not the 'tonic' component of the contractile effect of capsaicin is abolished by spinal cord transection indicating activation of a supraspinal micturition ref

Gastric erosions induced by intracisternal thyrotropin-releasing hormone (TRH) in rats.

Intracisternal injection of TRH (1 microgram) under light ether anesthesia induced within 4 hr gastric lesions in 24-hr fasted rats maintained unrestrained at room temperature. Saline, ovine corticotropin-releasing factor (oCRF, 10 micrograms), or human pancreatic growth hormone-releasing factor [hpGRF(1-40), 10 micrograms] tested under the same conditions did not modify the integrity of the gastric mucosa. TRH injected intravenously (100 micrograms/kg) proved to be ineffective. The production of gastric erosions elicited by intracisternal TRH (0.1-1 microgram) or by a stabilized TRH analog, R

[Effect of monoamines on luliberin levels in various parts of the hypothalamus of the male rat].

The effect of the monoamines--noradrenaline, dopamine and serotonin, injected into the third cerebral ventricle, on luliberin (LH-RH) content in the individual hypothalamic regions and the blood luteinizing hormone (LH) level, was studied in male rats. A reduced LH-RH level in the median eminence area and enhanced blood LH concentration were seen after noradrenaline injection. The number of cases where LH-RH level could be detectable in the blood, augmented following dopamine injection. The results obtained evidence that noradrenaline and dopamine participate in regulation of LH-RH synthesis a

Structure-activity studies on neurobehavioral effects of bombesin (BB) and gastrin releasing peptide (GRP).

In the present study, we evaluated and compared effects of BB, GRP and related C-terminal fragments on body temperature and grooming. Peptides were administered into the left cerebral ventricle of rats. Body temperature and grooming behavior were assessed over a 2 hr period following injections. All peptides significantly decreased body temperature and increased grooming behavior of animals. However, important differences in potency, efficacy and duration of action were found between BB, GRP and their fragments.

Action of anorexigenic peptide injected into the brain: dissociation of effect on body weight and feeding in the rat.

Previous reports on the effect of anorexigenic peptide (AXP) administered systemically in the rodent are inconsistent in terms of the effect of the tri-peptide on food intake and body weight. The purpose of this study was to examine the effect of AXP infused into the brain on these measures. In post-pubescent female rats of the Sprague-Dawley strain, guide cannulae were permanently implanted in the lateral cerebral ventricle for repeated intracerebroventricular (ICV) infusion. Postoperatively, measures of food and water intake and body weight were obtained every day at the same time. After a 7

Is alcohol induced poikilothermia mediated by 5-HT and catecholamine receptors or by ionic set-point mechanism in the brain?

In adult male Sprague-Dawley rats, stainless steel guide cannulae were implanted stereotaxically in either the lateral or third cerebral ventricle. Postoperatively, each animal was maintained at an ambient temperature of 22 degrees C. Just prior to the intragastric gavage of 4.0 g/kg ethyl alcohol (20% solution) individual animals were fitted with a colonic thermistor probe. Then, control CSF, a monoaminergic receptor antagonist, or a Ca++ ion chelating agent, EGTA, was infused into either the lateral or third ventricle (ICV) in a volume of 10.0 microliter. Augmentation of vagal reflex bradycardia be central alpha2-adrenoceptors in the cat.

Vagal reflex bradycardia was induced in anaesthetized cats with high level spinal axotomy by electrical stimulation of either the carotid sinus nerves or a depressor nerve. In both preparations reflex bradycardia increased with the rate of stimulation. Injection of 1 microgram/kg clonidine into a lateral cerebral ventricle augmented reflex bradycardia in response to carotid sinus nerve stimulation while the same dose of clonidine was ineffective when given intravenously. The antagonistic effect of intracerebroventricular yohimbine (50 micrograms/kg) indicated that the effect of clonidine was d

Interaction between nicotine and endogenous opioid mechanisms in the unanesthetized dog.

Nicotine produced a distinct reproducible syndrome in the conscious dog when injected intravenously or intracerebroventricularly. Intravenously administered nicotine (40 micrograms/kg/min for 20 minutes) increased cardiac and respiratory rates and produced analgesia, miosis, hypothermia, behavioral restlessness and emesis. When microinjected into the third cerebral ventricle, nicotine (100-200 micrograms) similarly increased cardiac and respiratory rates and pupillary diameter; and produced behavioral restlessness, emesis, erratic analgesia and maintained wakefulness and a desynchronized EEG.

Diagnosis of intraventricular and cisternal cysticercosis by computerized tomography with positive intraventricular contrast medium.

Computerized tomography (CT) has replaced pneumoencephalography and ventriculography in the diagnosis of intraventricular cysticercosis. The authors present a refinement in the use of CT by introducing a positive contrast medium into the ventricles to increase the definition of plain and contrast-enhanced scans in the diagnosis of intraventricular cysticercosis. Eleven cases of Cysticercus cyst are presented, 10 or which were precisely delineated by CT-iodoventriculography. In the remaining case, lack of definition was due to obstruction of the cerebral aqueduct. Surgical confirmation was obta

Transient vasopressin release and thirst in response to prolonged intracerebroventricular infusions of hypertonic mannitol in saline.

In the conscious goat infusions of 0.4 M mannitol in 0.15 M NaCl into the lateral cerebral ventricle (40 or 100 min, 0.02 ml/min) caused slight, transient vasopressin release and temporary thirst, whereas infusions or pure, hypertonic (0.7 M) mannitol did not elicit thirst and inhibited the basic vasopressin release in the nonhydrated animal. In contrast, infusions of equiosmolal (0.35 M) NaCl induced persistent thirst and pronounced elevation of the plasma vasopressin concentration throughout the infusion period. The cerebrospinal fluid (CSF) osmolality was raised by the same order of magnitu

Inhibition of drinking in water-deprived rats by combined central angiotensin II and cholinergic receptor blockade.

The effect of blockade of central angiotensin II (AII) receptors and cholinergic receptors on thirst induced by water deprivation was studied in Sprague-Dawley rats and rats with hereditary hypothalamic diabetes insipidus (DI). Neither central AII nor cholinergic blockade alone affected drinking. Antagonism of both receptors simultaneously, however, significantly inhibited water intake of both Sprague-Dawley and DI rats. This inhibitory effect was not observed in water-deprived, nephrectomized rats. The combined antagonism on water intake was specific, since milk intake in hungry rats was not

Some observations on the anorectic activity of prostaglandin F2alpha.

1 Intracerebroventricular injection of prostanglandin F2alpha (10-40 microgram) decreases food intake in a dose-dependent manner in rats trained to consume their daily total food intake in a 2 h period. 2 This anorexia is also observed in satiated rats, which had ad libitum access to food. 3 The anorectic activity of prostaglandin F2alpha is not modified by changes in the internal environment of the body after food intake, such as increased blood glucose and insulin levels and decreased fatty acid levels, or by the presence or absence of food in the stomach, as is evident from the anorectic ac

Transarterial and Transvenous Embolization for Posterior Condylar Canal Dural Arteriovenous Fistula.

OBJECTIVE: Posterior condylar canal dural arteriovenous fistula (PCC DAVF) is extremely rare, with only four previously reported cases in the English literature. Cases may present tinnitus and radiculopathy. In cases where the drainer is around the brainstem, subarachnoid and intraventricular hemorrhages (IVHs) may occur. We describe the clinical presentation, angiographic imaging, and endovascular treatment strategy of a PCC DAVF. CASE PRESENTATION: A 30-year-old woman presented to our hospital with tinnitus and stiffness of the shoulder. Neuroimaging studies showed DAVF with fistulous point

The effect of lowering the 5-hydroxytryptamine content of the rat spinal cord on analgesia produced by morphine.

1. Injection of 5,6-dihydroxytryptamine (5,6-DHT, 50 mug) into a lateral cerebral ventricle of male rats lowered the 5-hydroxytryptamine (5-HT) content of the lumbar cord to 12.8% and reduced the analgesic effect of low doses of morphine (0.64-1.63 mg/kg), tested by exerting pressure on the foot; after doses of morphine of 1.33-1.63 mg/kg, the analgesic response was reduced or abolished in 33% of the rats, and after 0.64 mg/kg, 58% of the animals failed to respond normally.2.
