EZB-type diffuse large B-cell lymphoma cell lines have superior migration capabilities compared to MCD-type.

Diffuse large B-cell lymphoma (DLBCL) represents the most prevalent aggressive B-cell lymphoma. The group is heterogeneous and the outcome is variable. A variety of approaches have been employed with the objective of improving the stratification of DLBCL patients according to their prognosis, based on the cell of origin. Recently, distinct genetic subtypes of DLBCL have been identified. Given the importance of cell migration in immune cells, the objective of this study was to ascertain whether different genetic subtypes of DLBCL exhibit disparate migration abilities. MCD- and EZB-type DLBCL ce

The salvage role of allogeneic hematopoietic stem-cell transplantation in relapsed/refractory diffuse large B cell lymphoma.

To clarify the role of allogeneic hematopoietic stem-cell transplantation (allo-HSCT) in the chimeric antigen receptor T-cell therapy era, we analyzed the clinical characteristics and outcomes of 52 patients treated with allo-HSCT with relapsed/refractory diffuse large B cell lymphoma. Most enrolled patients had previously undergone intensive treatments, the median number of chemotherapy lines was 4, and the median time from diagnosis to allo-HSCT was 27.1 months. Patients were divided into remission-achieved (n = 30) and active-disease (n = 22) groups before allo-HSCT. Over a median follow-up

Immune scoring model based on immune cell infiltration to predict prognosis in diffuse large B-cell lymphoma.

BACKGROUND: Diffuse large B-cell lymphoma (DLBCL) is genetically heterogeneous in both pathogenesis and clinical symptoms. Most studies on tumor prognosis have not fully considered the role of tumor-infiltrating immune cells. This study focused on the role of tumor-infiltrating immune cells in the prognosis of DLBCL. METHODS: The GSE10846 data set from the National Center for Biotechnology Information's Gene Expression Omnibus was used as the training set, and the GSE53786 data set was used as the validation set. The proportion of immune cells in each sample was calculated with the CIBERSORT

Autologous bone marrow transplant in a patient with sickle cell disease and diffuse large B-cell lymphoma.

As the life expectancy of patients with homozygous sickle cell anemia (SCA) improves, SCA care providers are confronted with diseases of the adult SCA population rarely seen before. We report here a 40-year-old woman with SCA who developed diffuse large B-cell non-Hodgkin's lymphoma (NHL) that was treated with eight cycles of chemotherapy consisting of cyclophosphamide, doxorubicin, vincristine, prednisone, and etoposide (CHOPE), without complete remission. She subsequently underwent high-dose cyclophosphamide and total-body irradiation followed by autologous bone marrow transplantation (BMT).

Gastric low-grade MALT lymphoma, high-grade MALT lymphoma and diffuse large B cell lymphoma show different frequencies of trisomy.

Gastric MALT lymphoma is a distinct entity related to Helicobacter pylori gastritis. Some studies suggest a role for trisomy 3 in the genesis of these lymphomas, but they mainly focused on low-grade MALT lymphoma. Gastric MALT lymphoma, however, comprises a spectrum from low- to high-grade cases. Furthermore, its exact relation to primary diffuse large B cell lymphoma (DLBCL) of the stomach is not clear. We applied in situ hybridisation (ISH) with centromeric probes on 43 samples of 39 patients with primary gastric lymphoma (13 samples with low-grade MALT lymphoma, 25 with high-grade MALT lymp

Autologous transplant for patients with chemotherapy-sensitive late relapse of diffuse large B-cell lymphoma.

In the era of chimeric antigen receptor (CAR) T-cell therapy, autologous stem cell transplant (ASCT) for diffuse large B-cell lymphoma (DLBCL) is preferentially offered to patients with late relapse (>12 months) who achieve complete or partial response (CR or PR, respectively) to salvage therapy. We conducted a retrospective cohort study using publicly available data from the Center for International Blood and Marrow Transplant Research in this population. We included adult patients with DLBCL, not otherwise specified, and high-grade large B-cell lymphomas who experienced disease relapsed afte

TP53 mutation predict poor prognosis in diffuse large B-cell lymphoma: a single-center study.

Whether TP53 mutation can serve as a simplified prognostic surrogate for complex genomic classifiers in diffuse large B-cell lymphoma (DLBCL) remains unclear. we comprehensively analyzed a retrospective cohort of 444 newly diagnosed DLBCL patients. TP53 mutation was an independent prognostic factor for inferior overall survival (OS) and progression-free survival (PFS) (both P < 0.001), correlating with inferior response to R-CHOP and enrichment in refractory/relapsed disease. Furthermore, multivariate analysis revealed that the adverse prognostic effect of TP53 mutation was predominantly drive

LySeqST: a targeted sequencing assay for robust genomic classification of diffuse large B-cell lymphoma.

Diffuse large B-cell lymphoma (DLBCL) is a biologically heterogeneous disease. Two genomic classification systems, LymphGen and DLBclass, are capable of reproducibly classifying single tumors into subtypes with prognostic relevance in the context of standard-of-care R-CHOP (rituximab, cyclophosphamide, doxorubicin, vincristine, and prednisone) chemoimmunotherapy. Preliminary data from subgroup analyses of clinical trials suggest that the distinct biological features are targetable with drugs that exploit subtype-specific vulnerabilities, and large clinical trials are being designed to test thi

Cardiovascular disease risk after radiotherapy and anthracycline-based chemotherapy for diffuse large B-cell lymphoma.

BACKGROUND: Few studies examined treatment-specific long-term risks of cardiovascular diseases (CVD) in diffuse large B-cell lymphoma survivors treated with potentially cardiotoxic radiotherapy and/or chemotherapy with or without rituximab after the 1990s. METHODS: Long-term CVD risk was examined in a multicenter cohort comprising 2356 diffuse large B-cell lymphoma survivors who survived at least 5 years and who were treated at ages 15-61 years in 1989-2012. CVD data were acquired from medical records, general practitioners, and disease registries. Observed CVD numbers were compared with expe

Peritoneal Lymphomatosis Mimicking Advanced Ovarian Malignancy: A Diagnostic Challenge in Diffuse Large B-Cell Lymphoma.

Diffuse large B-cell lymphoma (DLBCL) may rarely present as peritoneal lymphomatosis, closely mimicking peritoneal carcinomatosis on imaging and creating significant diagnostic uncertainty. We report the case of an 82-year-old woman presenting with progressive dyspnoea, fatigue, and systemic symptoms following treatment for presumed community-acquired pneumonia. Contrast-enhanced computed tomography demonstrated ascites, diffuse peritoneal thickening, omental involvement, and widespread lymphadenopathy, initially raising suspicion for advanced ovarian malignancy, supported by an elevated CA125

[Future directions in clinical development of first-line therapy for diffuse large B-cell lymphoma].

For an extended period, R-CHOP therapy was the standard of care for the first-line treatment of diffuse large B-cell lymphoma (DLBCL); nevertheless, approximately 30% of patients experience treatment resistance or relapse, highlighting the limitations of a "one-size-fits-all" treatment strategy. In recent years, however, the development of risk-adapted therapy based on molecular subtype identification and response-adapted therapy using circulating tumor DNA (ctDNA) to detect minimal residual disease has advanced considerably, building on Pola-R-CHP therapy, which became the new standard of car

Renal Diffuse Large B-cell Lymphoma Mimicking Acute Pyelonephritis on 18 F-FDG PET/CT.

A 58-year-old man with a history of lithotripsy for urolithiasis presented with persistent fever and pollakisuria. As empirical anti-infective therapy was ineffective, 18 F-FDG PET/CT was performed to detect potential malignancy, revealing bilateral renal enlargement with multiple nodular cortex uptakes and an enlarged left axillary lymph node, suggestive of pyelonephritis. However, biopsies of the axillary lymph node and right kidney confirmed high-grade diffuse large B-cell lymphoma (DLBCL). Following R-DA-EPOCH chemotherapy, the patient achieved sustained remission. This case highlights tha

Primary splenic diffuse large B-cell lymphoma masquerading as systemic lupus erythematosus: a case report.

BACKGROUND: Diffuse large B-cell lymphoma (DLBCL) is the most common subtype of non-Hodgkin lymphoma, whereas primary splenic DLBCL (PS-DLBCL) is exceedingly rare. Lymphomas that mimic systemic lupus erythematosus (SLE) are clinically heterogeneous and share overlapping manifestations with SLE, rendering diagnosis extremely challenging. CASE PRESENTATION: We report a case of an elderly patient presenting with recurrent fever, a hyperinflammatory state, and cytopenias, along with positivity for multiple autoimmune antibodies and lupus anticoagulant, which strongly suggested a clinical diagnosi

Cutaneous diffuse large B-cell lymphoma induces a macrophage immunosuppressive phenotype through IL-10 secretion.

Primary cutaneous diffuse large B-cell lymphoma (PCDLBCL), leg type (PCDLBCL-LT) is a rare subset of DLBCL that exhibits genetic features shared with LBCL of immune-privileged sites (notably the MYD88L265P mutation) and is enriched in tumor-associated macrophages expressing M2 markers. Using a unique PCDLBCL-LT cell line (ARSI cell line) developed in our institute, we sought to mechanistically decipher the interplay between lymphoma cells and macrophages. We demonstrate that ARSI cells induce phenotypic, transcriptional, and functional changes in macrophages obtained from primary monocytes and

Impact of diagnosis-to-treatment interval in relapsed or refractory diffuse large B-cell lymphoma.

The diagnosis-to-treatment interval (DTI) has been identified as a prognostic factor in newly diagnosed diffuse large B-cell lymphoma (DLBCL). However, its significance in relapsed or refractory DLBCL (R/R DLBCL) remains unclear. This study aimed to investigate the impact of DTI on survival outcomes in patients with R/R DLBCL. We reviewed the medical records of patients with R/R DLBCL who received second-line therapy. DTI at relapse or refractory disease (r/r DTI) was defined as the time from radiological diagnosis-to-treatment initiation, with a cut-off of 28 days. The primary endpoint was ov

Case Report: A case of type II cryoglobulinemia secondary to diffuse large B-cell lymphoma.

BACKGROUND: Diffuse Large B-Cell Lymphoma (DLBCL) represents the most prevalent subtype of Non-Hodgkin Lymphoma (NHL). Cryoglobulinemia results from reversible precipitation of serum immunoglobulin complexes at low temperatures, with Type II (mixed) cryoglobulinemia commonly associated with hepatitis C virus (HCV) infection, autoimmune diseases, and B-cell lymphoproliferative disorders. However, its occurrence secondary to DLBCL is exceedingly rare. This report presents a rare case of DLBCL complicated by Type II cryoglobulinemia, acute liver injury, acute kidney injury, and severe infection.

Unraveling novel therapeutic targets for diffuse large B-cell lymphoma: a plasma multi-omics study.

BACKGROUND: Diffuse large B-cell lymphoma (DLBCL), the most common non-Hodgkin lymphoma, exhibits genetic heterogeneity and variable treatment responses. Discovering new therapeutic targets is critical to improve outcomes. Given the lack of plasma protein studies for DLBCL, this study aims to first identify novel therapeutic targets based on plasma multi-omics and genome-wide association studies with further verifications. METHODS: By two-step training (N = 456,948 participants) and validation (N = 288,927 participants), druggable genes and proteins were identified through the mendelian rando

Primary Bone Diffuse Large B-cell Lymphoma (DLBCL) in a Young Adult: A Case Report.

Primary bone diffuse large B-cell lymphoma (DLBCL) is a rare subtype of extranodal non-Hodgkin lymphoma that predominantly involves long bones, particularly the metaphyseal regions. We report the case of a 23-year-old man presenting with painful swelling of the right knee. Imaging revealed an osteolytic lesion of the distal right femur with cortical destruction and soft tissue extension. Staging contrast-enhanced computed tomography of the chest, abdomen, and pelvis showed no nodal or visceral involvement, consistent with localized (limited-stage) disease. Histopathological examination of the
